Description: Plant-parasitic nematodes are major agricultural pests worldwide and novel approaches to control them are sorely needed. We report the draft genome sequence of the root-knot nematode Meloidogyne incognita, a biotrophic parasite of many crops, including tomato, cotton and coffee. Most of the assembled sequence of this asexually reproducing nematode, totaling 86 Mb, exists in pairs of homologous but divergent segments. This suggests that ancient allelic regions in M. incognita are evolving toward effective haploidy, permitting new mechanisms of adaptation. The number and diversity of plant cell wall–degrading enzymes in M. incognita is unprecedented in any animal for which a genome sequence is available, and may derive from multiple horizontal gene transfers from bacterial sources. Our results provide insights into the adaptations required by metazoans to successfully parasitize immunocompetent plants, and open the way for discovering new antiparasitic strategies.

Notes: Abstract In the Twin Lakes area, central Sierra Nevada, California, most contact metamorphosed marbles contain calcite + dolomite + forsterite ± diopside ± phlogopite ± tremolite, and most calc-silicate hornfelses contain calcite + diopside + wollastonite + quartz ± anorthite ± K-feldspar ± grossular ± titanite. Mineral-fluid equilibria involving calcite + dolomite + tremolite + diopside + forsterite in two marble samples and wollastonite + anorthite + quartz + grossular in three hornfels samples record P± 3 kbar and T± 630° C. Various isobaric univariant assemblages record CO2-H2O fluid compositions of χCO2= 0.61–0.74 in the marbles and χCO2= 0.11 in the hornfelses. Assuming a siliceous dolomitic limestone protolith consisting of dolomite + quartz ° Calcite ± K-feldspar ± muscovite ± rutile, all plausible prograde reaction pathways were deduced for marble and hornfels on isobaric T-XCO2 diagrams in the model system K2O-CaO-MgO-Al2O3-SiO2-H2O-CO2. Progress of the prograde reactions was estimated from measured modes and mass-balance calculations. Time-integrated fluxes of reactive fluid which infiltrated samples were computed for a temperature gradient of 150 °C/km along the fluid flow path, calculated fluid compositions, and estimated reaction progress using the mass-continuity equation. Marbles and hornfelses record values in the range 0.1–3.6 × 104 cm3/cm2 and 4.8–12.9 × 104 cm3/cm2, respectively. For an estimated duration of metamorphism of 105 years, average in situ metamorphic rock permeabilities, calculated from Darcy's Law, are 0.1–8 × 10−6 D in the marbles and 10–27 × 10−6 D in the hornfelses. Reactive metamorphic fluids flowed up-temperature, and were preferentially channellized in hornfelses relative to the marbles. These results appear to give a general characterization of hydrothermal activity during contact metamorphism of small pendants and screens (dimensions ± 1 km or less) associated with emplacement of the Sierra Nevada batholith.

Notes: Extensive examination of large numbers of spatially orientated thin sections of orientated samples from orogens of all ages around the world has demonstrated that porphyroblasts do not rotate relative to geographical coordinates during highly non-coaxial ductile deformation of the matrix subsequent to their growth. This has been demonstrated for all tectonic environments so far investigated. The work also has provided new insights and data on metamorphic, structural and tectonic processes including: (1) the intimate control of deformation partitioning on metamorphic reactions; (2) solutions to the lack of correlation between lineations that indicate the direction of movement within thrusts and shear zones, and relative plate motion; and (3) a possible technique for determining the direction of relative plate motion that caused orogenesis in ancient orogens.

Notes: Advertising is not a process by which gullible consumers are persuaded to buy things they don't want, according to the authors of this paper. Instead, it furnishes consumers with some useful information. It is not so much the claims made by advertisers that are helpful, but the fact that they are willing to spend extravagant amounts of money on a product that is informative. The authors find tentative evidence for this view of advertising and discuss its implications.

Notes: Nonparametric prediction limits can be useful statistical tools for ground water monitoring at facilities regulated under RCRA Subtitle C. Subtitle D. and similar regulations. New, exact tables arc presented for both “1 of m” plans (m chances to gel one observation inbounds at each of r monitoring wells to avoid a statistically significant increase) and “California” plans (first or all of the next m-1 observations inbounds at each well). The tables provide per-constituent significance levels (false positive rates) as a function of the background sample size n. m. r, the prediction limit (the largest or the next to largest, background observation), and the confirmatory resampling plan selected. When used in a monitoring program, future observations from several wells are compared with a prediction limit obtained from a common background sample. The table significance levels therefore depend critically on having IID (independent and identically distributed) observations. In particular, the false positive rate computations are not valid, and the procedures should not be used, with constituents whose measurements exhibit inherent spatial or systematic temporal variability.Recent U.S. EPA guidance explicitly encourages controlling facility-wide false positive rates over both constituents and wells. Nonparametric prediction limits, particularly with California resampling plans, will have greater difficulty in meeting the new. lower per-constituent false positive rate goals than previous ones, especially if many constituents are involved. Nonetheless, nonparametric prediction limits remain superior to other commonly used procedures for dealing with data with high proportions of nondctects.

Notes: Lincolns municipal wellfield consists of 44 wells developed in an alluvial aquifer adjacent to the Platte River near Ashland, Nebraska Induced recharge from the river is the primary source of water for the wellfield. Wafer samples were collected on a periodic basis from the Platte River arid two transects of monitoring wells. These samples were analyzed for the herbicide atrazine, which was used as a tracer of induced recharge in this stream-aquifer system. Atrazine concentrations in the river and aquifer were much less than 1.0 ppb during late fall and winter, but increased to as high as 18.9 ppb during spring and summer, associated with runoff from upgradient agricultural lands. There was approximately a 21-day lag time from the first detection of increasing atrazine concentration in the river to the first detection in monitoring wells immediately adjacent to the river. This lag time was relatively constant throughout the year and from one year to the next, even with major fluctuations of river stage and wellfield production. This consistency of lag time indicated that the travel times from the river to the first set of monitoring wells immediately adjacent to the river were fairly constant.Paths of preferential flow were identified in the aquifer at a depth of 25 to 35 feet below land surface. This aquifer zone appeared to play a significant role in movement of water from beneath the river into the wellfield.Aquifer dispersivity was calculated using a method described by Hoehn and Santschi (1987). Macrodispersivity (AL) was shown to increase linearly over the scale of the wellfield. Calculated values of AL were within limits of other reported values for this type of aquifer material and agreed well with values reported by Hoehn and Santschi (1987); These findings will be extremely beneficial for planning and management of the municipal wellfield.

Notes: The aim of the study was to determine whether C5 deficiency in the mouse is associated with resistance to the development of secondary amyloidosis. Chronic inflammation was induced in the F2 progeny, derived from matings between amyloid-susceptible and amyloid-resistance mice, by daily injections of azocasein for thirty days. Using a restriction fragment length polymorphism generated by digestion of genomic DNA with the restriction enzyme HindIII, C5 sufficient and deficient DNA can be clearly differentiated. Eight mice were found to be C5 sufficient, 32 were heterozygotes and 14 were found to be C5 deficient. Grading of the splenic amyloid load from negative to 4+ was performed after staining tissue squashes with Congo red and viewing them under a polarizing microscope. Seventeen mice were noted to have negative to trace, 18 had moderate (1+-2+) and 19 had heavy (3 H+-4+) amyloid deposition. There was no correlation between splenic amyloid load and C5 deficiency. Based on these results it is clear that C5 deficiency and resistance to secondary amyloidosis are not associated.

Notes: Abstract— The present research was conducted to evaluate if there is an interrelationship between methionhe and choline as well as between betaine and choline in feed formulations for channel catfish. A semi-purified basal diet was formulated to contain 28-g crude protein/100-g diet. DL-methionine was added to the basal diet to produce deplete, sufficient, and replete levels of methionine (0.35%, 0.39%, and 0.43%); choline chloride (200, 400, and 600-mg cholinentg diet) was added to the basal diet to produce low, median, and high levels of choline; and free base betaine (0 and 1,000-mg betainelkg diet) was added to the basal diet to produce sufficient and replete levels of betaine in eight experimental diets. Each diet was randomly fed to triplicate groups of fish (15 fishleach group) over a 13-wk growth trial. Added choline in the diets containing 0.35% (deplete) methionine had a significant linear effect on weight gain and feed consumption. There were no significant differences in weight gain, FCR, survival, HIS, and liver lipid between diet 3 (deplete methionhe and high choline) and diet 6 (sufficient methionine and median choline). Both added choline and betaine in the diets containing 0.39% (sufficient) methionine had significant effects on weight gain, feed consumption, and FCR. There were no significant differences in the measured parameters between diet 7 (low choline and excess betaine) and diet 6 (median choline and no betaine). Methionine had no significant Linear effect on weight gain and feed consumption when choline was low. Therefore, in the absence of sufficient methionine, choline or betaine can spare a portion of methionine requirement of channel catfish. In the absence of sufficient choline, methionine cannot spare choline requirement of channel catfish. Further, in the absence of sufficient choline, betaine can spare at least a portion of choline requirement of channel catfish.

Notes: Abstract— Plasma concentrations of insulin-like growth factor-I (IGF-I) and insulin-like growth factor binding proteins (IGFBPs) were compared between market sized sunshine and palmetto bass. Differences between male and female fish were examined and both hybrids were sampled before and after a low-water confinement stress. There was no difference in body weight between the hybrid types, however, females were significantly heavier than males in both hybrids. Plasma IGF-I concentrations were significantly higher in pre-stressed palmetto bass when compared with sunshine bass, and male palmetto bass had significantly higher IGF-I concentrations than females. Two hours after a 15-min low-water stress, a significant decrease in palmetto bass IGF-I occurred, but there was no change in sunshine bass IGF-I concentrations. Levels of a 33-kDa IGFBP were similar in pre-stressed palmetto bass when compared with sunshine bass; however, no differences due to gender were evident for either hybrid. Two hours after a 15-min low-water exposure, there was no change in the levels of a 33-kDa IGFBP in palmetto or sunshine bass. A more complete understanding of IGF-I, IGFBPs, and IGF-I receptors may be necessary to explain the role of IGF-I in regulating fish growth.
