    Notes: Summary In a cytophysiological study it was investigated whether in juvenile trout gonadal steroids stimulate the gonadotropic (GTH)-cells directly or indirectly via the brain. Pituitaries of donor animals were transplanted into the caudal musculature of testosterone-treated and non-testosterone-treated host fish. Testosterone treatment caused an increase in GTH-content in the in situ pituitaries and in the grafts. Accordingly, the gonadotrops displayed ultrastructural changes such as the appearance of well-developed Golgi systems and large globules. The stimulation of the morphological development of gonadotrops and of synthesis and storage of GTH in the allografted pituitaries indicates that testosterone affects the GTH-cells directly. In untreated juvenile trout the gonadotropin content of the pituitary and the gonadotropin concentration in the plasma vary with the time of year. This variation and the role of testosterone and gonadotropin-releasing hormone on the release of GTH are discussed.

    Notes: Summary Dispersed pituitary cells from male African catfish, Clarias lazera, were fractionated in a density gradient of Percoll. Five fractions were isolated, consisting of about 6, 19, 39, 95 and 83% gonadotrops, respectively. The gonadotrops were identified by their ultrastructural characteristics, by immunocytochemistry, and by measuring their hormone content. After one day in culture, in each fraction the secretion of gonadotropin could be stimulated by a luteinizing hormone-releasing hormone analogue, indicating that the cells had retained their functional integrity. Since the regulatory mechanisms of different cell types from the pituitary have some similarity, purification of the gonadotrops provides a model to study the regulation of gonadotropin secretion.

    Notes: Summary Pituitaries of the African catfish (Clarias lazera) were studied with immunocytochemical methods, at the light-microscopic and ultrastructural levels, for the characterization and localization of gonadotropic and thyrotropic cells. Two immunostaining procedures with the use of different markers were carried out: (i) with peroxidase-antiperoxidase, (ii) with protein A-gold. In routinely stained sections for light microscopy two types of basophils were identified in the proximal pars distalis: (1) large, round, purple cells, and (2) small, angular, light-blue cells. Both types were immunolabeled with antibodies against Clarias α,β-gonadotropin (GTH) and salmon G100-GTH. Only the large basophils were immunolabeled with anti-carp β-GTH, whereas the small basophils were the only cells immunolabeled with anti-human thyrotropin beta subunit (anti-h TSH-β). It was concluded that the large basophils represent the gonadotrops and the small basophils the thyrotrops. At the ultrastructural level the immunostaining of the GTH-cells was confined to three types of inclusions: (i) secretory vesicles, (ii) globules, and (iii) electron-dense, membrane-bound irregular masses. Especially the protein A- gold method, in combination with the use of a highly diluted homologous antiserum, resulted in a distinct localization of GTH. The presence of two types of nerve fibres, synaptically contacting the gonadotrops, is discussed with regard to the presence of a peptidergic (stimulatory) and an aminergic (inhibitory) control of GTH-secretion.

    Notes: Summary A specific HPLC method has been developed for the trace analysis of lower chlorinated aromatic compounds. The method consists of an on-line preconcentration and a post-column reaction step. On-line preconcentration of mono- and dichlorophenols from aqueous samples has been performed using PRP1, a divinylbenzene-styrene copolymeric sorbent as packing material for both pre-and analytical column. Enrichment factors of over 300 were obtained compared to regular (100 μl) loop injections, even for the highly polar monochlorophenols. After reversed-phase separation, post-column photochemical dechlorination is carried out directly in the eluent stream, using a photochemical reactor. Upon dechlorination, fluorescent products are formed, which can be detected selectively. The resulting fluorescence signal shows a linear response to the quantity of solute present over 2 to 3 orders of magnitude (correlation coefficients: 0.990–0.98). For the mono- and dichlorophenols, the detection limit of the photoconversion method is in the lower nanogram range. The method is especially suitable for the analysis of complex matrices such as effluent water samples or biological fluids containing traces of the polar chloroaromatic compounds together with large amounts of other constituents, which interfere when using more general methods of detection like UV absorption. The potential of this technique for automation has been demonstrated by using a microprocessor-controlled column switching unit.

    Notes: Summary A new photochemical reaction method for the on-line fluorimetric detection of natural vitamin K homologs was developed. In addition to its high sensitivity, the method features an interesting selectivity for the determination of these compounds in a complex matrix such as human serum. The formation of different photolysis products as a function of reaction conditions, and the optimalisation of a segmented flow reaction system will be discussed. The results indicate that the method is 4 times more sensitive than UV detection, and at least as selective as fluorescence detection after post-column chemical reduction.

    Notes: Summary The retention behaviour of nucleobases and nucleosides on unmodified silica with dichloromethane-methanol-water mixtures has been systematically investigated. The degree and order of retention can be varied over a wide range by changing the pH, the type and concentration of the acidic additives and by the methanol and water content of the mobile phase. The retention process cannot be considered as simple adsorption but rather as a very complex mixed distribution process of adsorption and absorption including the involvement of ion-pair formation. Further data on the effect of the type of silica (source of supply) on retention behaviour, column efficiency and column reproducibility are reported. The practical application of unmodified silica for the separation of nucleobases and nucleosides is demonstrated by the analysis of a hydrolysate of calf thymus DNA.

    Notes: Summary An improved interface for the coupling of a liquid chromatograph (LC) and an electron-capture detector (ECD) allows the use of the LC-ECD combination for the determination of various types of chlorinated aromatic compounds. The addition of up to 6% of dioxan to the hexane used as mobile phase can be tolerated without serious deterioration of detector performance. Derivatization of polar compounds such as anilines, phenylurea herbicides, chlorophenols and hydroxylated polychlorobiphenyls with fluorine-and non-fluorine-containing acid anhydrides is a useful tool to extend the application range of LC-ECD. As an example, the analysis of a spiked soil sample is discussed.

    Notes: Summary Six different cyclodextrins with varying cavity size and rim substitution were used as chiral agents for the enantiomeric separation of eight chromane compounds or analogues using capillary electrophoresis. It is shown that the cyclodextrin type and concentration have a large influence on the enantiomeric separation obtained for these compounds. A chiral resolution of 1.4 or better could be obtained for all the substances with either substituted heptakis(2,3,6-tri-O-methyl)-β-cyclodextrin or unsubstituted γ-cyclodextrin as the chiral selector. The influence of the γ-cyclodextrin concentration, ionic strength and pH on the chiral separations was also investigated with a multivariate screening design. The detection limit and resolution of the present method allow determinations of the investigated compounds down to a chiral impurity of less than 0.1 % (area/area).

    Notes: Summary A method has been developed for the analysis of environmentally important aldehydes in rain water. The method is based on the derivatization of the aldehydes with hydrazino benzene sulfonic acid, separation of the hydrazones formed by capillary electrophoresis and UV detection at 280 nm. Derivatization was shown to be complete in 15 min at 50°C. The aldehyde derivatives could be separated from each other and from the excess of reagent using a pH 9 borate buffer as background electrolyte, with an analysis time of less than 6 min. The repeatability was better than 0.5% for the peak mobilities and in the order of 2–5% for the peak areas. Detection limits of 0.8–3 μmol L−1 (0.02–0.2 ppm) were obtained. The method was applied for the determination of formaldehyde and acetaldehyde in rain water samples.

    Notes: Summary Various methods are currently under investigation to improve concentration detection limits in capillary electrophoresis (CE). Stacking and isotachophoresis coupled in-line with free-solution electrophoresis are the techniques most frequently used. Samples containing a high and/or varying salt concentration, such as urine and serum, are difficult to handle: usually a significant loss in electrophoretic efficiency is observed compared with samples possessing a low electric conductivity. In the present paper an alternative approach is developed, i.e. a liquid chromatographic (LC)-type of sample pretreatment is coupled on-line with CE. To demonstrate the feasibility of this approach, the separation of three model compounds (benzoates) in water containing up to 400 mM of sodium chloride is studied using a 50 mM borate CE buffer of pH 9.5. The direct injection of samples with high salt concentrations in CE results in peak splitting and/or serious band broadening. These problems are not encountered when using the present LC-CE system. In addition, the detection limits are hardly influenced by the salt concentration of the sample; this underlines the robustness of the system.
