    Description: Spatial and temporal trends in stream chemistry were investigated in a large (1600 km2) alpine watershed in the southern Rocky Mountains of Colorado to help understand mechanisms of streamflow generation. We observed linear increases of concentrations of chemical constituents in streamflow as accumulated drainage area increased along the main channel of Saguache Creek. We tested two conceptual models of streamflow generation against our stream chemistry observations. One model is essentially two-dimensional and treats streamflow generation at the large watershed scale as the aggregation of runoff responses from individual hillslopes, primarily surface and shallow subsurface flow paths. Alternatively, a fully three-dimensional conceptual model treats streamflow generation as being controlled by a distribution of large-scale groundwater flow paths as well as surface and shallow subsurface flow paths. The structure and magnitude of groundwater contributions in streamflow as a function of increasing scale provided a key distinction between these two conceptual models. End-member mixing analysis and measurements of hydraulic head gradients in streambeds were used to quantify basin-scale groundwater contributions to streamflow with increasing spatial scale in the Saguache Creek watershed. Our data show that groundwater contributions are important in streamflow generation at all scales and, more importantly, that groundwater contributions to streamflow do increase with increasing watershed scale. These results favor the three-dimensional conceptual model in which long groundwater flow paths provide a streamflow generation process at large scales that is not operative at smaller scales. This finding indicates that large watersheds may be more than simply the aggregation of hillslopes and small catchments.

    Description: We investigate the representation of the Sierra Barrier Jet (SBJ) in four numerical models at different resolutions, primarily documenting its representation within a high-resolution (6 km), 11-year WRF reanalysis downscaling (WRF-RD). A comprehensive validation of this dynamical downscaling is undertaken during 11 cool seasons (water years 2001–2011, October to March) using available wind profiler data at Chico, CA (CCO). We identify SBJ cases in the observed CCO wind profiler data, as well as in WRF-RD at the closest grid point. WRF-RD's representation of the SBJ at CCO is compared with that of other reanalysis products with coarser horizontal resolutions (i.e., the North American Regional Reanalysis (NARR), the California Reanalysis downscaling, and the NCEP/NCAR Reanalysis) to assess whether downscaling is necessary to correctly capture this topographically induced low-level jet. Detailed comparisons across California between WRF-RD and NARR suggest downscaling is necessary: Only WRF-RD at 6 km resolution is well-capturing this dynamical feature. A catalog of modeled SBJ events that have significant timing overlap with observations is created and used to further assess WRF-RD's representation of SBJ events. In addition, observation-model comparisons of other meteorologically important variables (e.g., precipitation melting level, wind profiles, temperature, and relative humidity) are performed in order to evaluate WRF-RD's ability to capture the dynamical evolution of the SBJ. The detailed, case-by-case comparisons reveal WRF-RD accurately represents 56 percent of the 256 observed SBJ cases occurring during these 11 cool seasons, albeit with a weak wind bias that increases with jet maximum wind strength.

    Description: This paper analyses the performance of an integrated Hydrological Ensemble Prediction System (HEPS) adapted to small to mid-sized catchments (100 up to 600 km 2 ) situated in the Cévennes-Vivarais region (Southern France) and characterized by short lag-times (3 to 12 hours). In this framework, flood forecasts need hourly Probabilistic Quantitative Precipitation Forecasts (PQPF) so as to provide early warning with 24 to 72 hours of anticipation. Here, two sources of PQPF at daily and sub-daily (6 hours) meteorological time-steps are considered: EPS from ECMWF and analogy-based PQPF provided in real-time at LTHE (Laboratoire d'étude des Transferts en Hydrologie et Environnement). The two PQPF are firstly disaggregated in order to respect the required hydrological hourly time-step, through either the use of a stochastic rainfall generator or the application of a multi-model approach. Then, disaggregated PQPF are used as input to the hydrological model TOPSIMPL in order to provide hourly ensemble discharge forecasts up to 48 hours ahead. Illustration and evaluation of ensemble discharge forecasts issued in near real-time conditions are given for some recently observed flash flood events. It is shown that hourly discharge forecasts are first-order conditioned by the accuracy of PQPF at daily or sub-daily time-step. 6-hour EPS correctly reproduce the rainfall temporal dynamics, while daily analogy-based PQPF are less underdispersive in terms of rainfall amounts. As a result, the merging of the two sources of PQPF substantially increases the performance of discharge forecasts, the contribution of a more sophisticated hourly rainfall generator becoming marginal.     Description: Promoting osteoblastogenesis remains a major challenge in disorders characterized by defective bone formation. We recently showed that the alpha 5 integrin subunit (ITGA5) is critically involved in human mesenchymal cell osteoblast differentiation. In this study, we determined the potential of pharmacological ITGA5 activation by a synthetic cyclic peptide (GA-CRRETAWAC-GA) on murine osteoblast differentiation and function in vitro and bone formation in vivo . Peptide-mediated activation of ITGA5 in murine C3H10T1/2 mesenchymal cells resulted in the generation of the integrin-mediated cell signals FAK and ERK1/2-MAPKs. In vitro , peptide-based activation of ITGA5 protected from cell apoptosis but did not affect cell adhesion or replication, while it enhanced the expression of the osteoblast marker genes Runx2 and type I collagen and increased extracellular matrix mineralization as also found with BMP2, a standard bone anabolic factor. When injected on adult mouse cranial bone for 3 weeks, the peptide-mediated activation of ITGA5 increased bone thickness by 2-fold, an effect also induced by BMP2. Histomorphometric analysis showed that this anabolic effect resulted from decreased cell apoptosis and increased bone forming surfaces and bone formation rate. We conclude that pharmacological activation of ITGA5 in mesenchymal cells is effective in promoting de novo bone formation as a result of increased osteoprogenitor cell differentiation into osteoblasts and increased cell protection from apoptosis. This peptide-based approach could be used therapeutically to promote the osteogenic capacity of osteoblast progenitor cells and to induce de novo bone formation in conditions where osteoblastogenesis is compromised.     Description: This paper analyzes the proportion of snowfall days relative to precipitation days, in order to assess the impact of changing temperatures on snowfall, while minimizing the impact of variations in precipitation frequency and intensity. We analyzed the ratio of snowfall days to precipitation days for up to 100 years at 76 meteorological stations, spanning elevations from 200 to 2700 m asl in Switzerland. Our results show clear decreasing trends in snowfall days relative to precipitation days. These decreases are connected to increasing temperatures. The decrease in snowfall days was stronger at lower elevations, i.e., at locations with temperatures closer to the melting point. We observed a baseline seasonal temperature threshold of −2.7°C ± 0.8°C in winter and −3.8°C ± 0.6°C in spring, above which the decrease in snowfall days grew rapidly. From these observations, we developed an empirical model that can be used to evaluate the impact of future temperature increases on snowfall, independent of changes in the frequency and intensity of precipitation events.

    Description: Age-related bone loss is characterized by reduced osteoblastogenesis and excessive bone marrow adipogenesis. The mechanisms governing bone marrow mesenchymal stromal cell (BMSC) differentiation into adipocytes or osteoblasts during aging are unknown. We show here that overexpressing N-cadherin ( Cadh2 ) in osteoblasts increased BMSC adipocyte differentiation and reduced osteoblast differentiation in young transgenic (Tg) mice whereas this phenotype was fully reversed with aging. The reversed phenotype with age was associated with enhanced Wnt5a and Wnt10b expression in osteoblasts and a concomitant increase in BMSC osteogenic differentiation. Consistent with this mechanism, conditioned media from young wild type osteoblasts inhibited adipogenesis and promoted osteoblast differentiation in BMSC from old Cadh2 Tg mice, and this response was abolished by Wnt5a and Wnt10b silencing. Transplantation of BMSC from old Cadh2 Tg mice into young Tg recipients increased Wnt5a and Wnt10b expression and rescued BMSC osteogenic differentiation. In senescent osteopenic mice, blocking the CADH2–Wnt interaction using an antagonist peptide increased Wnt5a and Wnt10b expression, bone formation, and bone mass. The data indicate that Cadh2/Wnt interaction in osteoblasts regulates BMSC lineage determination, bone formation, and bone mass and suggest a therapeutic target for promoting bone formation in the aging skeleton.
