    Description: The relationship between larval fish assemblages and coastal oceanography is the basis for much of our understanding of connectivity and productivity of fish populations. Larval fish assemblages were sampled from the upper mixed layer (〈50 m depth) at three prominent circulation features [separation of the East Australian Current (EAC), anticyclonic eddy, and cyclonic eddy] off the southeast Australian coast across three bathymetric zones (shelf, slope and ocean) for each feature. The separation of the EAC from the coast at ~32°S was characterized by warmer, less saline water compared with the cyclonic and anticyclonic eddies further to the south (~34 and ~35°S, respectively), which were both characterized by cooler Tasman Sea water and greater fluorescence. The anticyclonic eddy had separated from the EAC three months prior to sampling, which facilitated the movement of a cyclonic eddy from the Tasman Sea westwards to the shelf at ~34°S. The larval assemblage in the EAC had high numbers of fish of the families Labridae and Stomiidae. The cyclonic eddy was characterized by larval clupeids, carangids, scombrids and bothids, indicating recent entrainment of shelf waters and proximity to major spawning regions. In contrast, the anticyclonic eddy had fewer larval fish, with little evidence for entrainment of shelf assemblages into the near-surface waters. Myctophids were found in high abundance across all oceanographic features and bathymetric zones. The evidence of selective entrainment of coastal larval fish into the near-surface waters of a cyclonic eddy compared with a similar anticyclonic eddy indicates a potential offshore nursery ground.

    Description: We quantify the rate of sea level rise around the Australian continent from an analysis of tide gauge and Global Positioning System (GPS) data sets. To estimate the underlying linear rates of sea level change in the presence of significant interannual and decadal variability (treated here as noise), we adopt and extend a novel network adjustment approach. We simultaneously estimate time-correlated noise as well as linear model parameters and realistic uncertainties from sea level time series at individual gauges, as well as from time-series differences computed between pairs of gauges. The noise content at individual gauges is consistent with a combination of white and time-correlated noise. We find that the noise in time series from the western coast of Australia is best described by a first-order Gauss–Markov model, whereas east coast stations generally exhibit lower levels of time-correlated noise that is better described by a power-law process. These findings suggest several decades of monthly tide gauge data are needed to reduce rate uncertainties to 〈0.5 mm yr –1 for undifferenced single site time series with typical noise characteristics. Our subsequent adjustment strategy exploits the more precise differential rates estimated from differenced time series from pairs of tide gauges to estimate rates among the network of 43 tide gauges that passed a stability analysis. We estimate relative sea level rates over three temporal windows (1900–2011, 1966–2011 and 1993–2011), accounting for covariance between time series. The resultant adjustment reduces the rate uncertainty across individual gauges, and partially mitigates the need for century-scale time series at all sites in the network. Our adjustment reveals a spatially coherent pattern of sea level rise around the coastline, with the highest rates in northern Australia. Over the time periods beginning in 1900, 1966 and 1993, we find weighted average rates of sea level rise of 1.4 ± 0.6, 1.7 ± 0.6 and 4.6 ± 0.8 mm yr –1 , respectively. While the temporal pattern of the rate estimates is consistent with acceleration in sea level rise, it may not be significant, as the uncertainties for the shorter analysis periods may not capture the full range of temporal variation. Analysis of the available continuous GPS records that have been collected within 80 km of Australian tide gauges suggests that rates of vertical crustal motion are generally low, with the majority of sites showing motion statistically insignificant from zero. A notable exception is the significant component of vertical land motion that contributes to the rapid rate of relative sea level change (〉4 mm yr –1 ) at the Hillarys site in the Perth area. This corresponds to crustal subsidence that we estimate in our GPS analysis at a rate of –3.1 ± 0.7 mm yr –1 , and appears linked to groundwater withdrawal. Uncertainties on the rates of vertical displacement at GPS sites collected over a decade are similar to what we measure in several decades of tide gauge data. Our results motivate continued observations of relative sea level using tide gauges, maintained with high-accuracy terrestrial and continuous co-located satellite-based surveying.

    Description: The lack of adipose triglyceride lipase (ATGL), a patatin-like phospholipase domain-containing enzyme that hydrolyzes fatty acids from triacylglycerol (TAG) stored in multiple tissues, causes the autosomal recessive disorder neutral lipid storage disease with myopathy (NLSD-M). In two families of Lebanese and Italian origin presenting with NLSD-M, we identified two new missense mutations in highly conserved regions of ATGL (p.Arg221Pro and p.Asn172Lys) and a novel nonsense mutation (p.Trp8X). The Lebanese patients harbor homozygous p.Arg221Pro, whereas the Italian patients are heterozygotes for p.Asn172Lys and the p.Trp8X mutation. The p.Trp8X mutation results in a complete absence of ATGL protein, while the p.Arg221Pro and p.Asn172Lys mutations result in proteins with minimal lipolytic activity. Although these mutations did not affect putative catalytic residues or the lipid droplet (LD)-binding domain of ATGL, cytosolic LDs accumulated in cultured skin fibroblasts from the patients. The missense mutations might destabilize a random coil (p.Asn172Lys) or a helix (p.Arg221Pro) structure within or proximal to the patatin domain of the lipase, thereby interfering with the enzyme activity, while leaving intact the residues required to localize the protein to LDs. Overexpressing wild-type ATGL in one patient's fibroblasts corrected the metabolic defect and effectively reduced the number and area of cellular LDs. Despite the poor lipase activity in vitro , the Lebanese siblings have a mild myopathy and not clinically evident myocardial dysfunction. The patients of Italian origin show a late-onset and slowly progressive skeletal myopathy. These findings suggest that a small amount of correctly localized lipase activity preserves cardiac function in NLSD-M.
