    Description: The HIV-1 BED incidence assay was adopted in China in 2005 for HIV-1 incidence surveillance. A proficiency testing (PT) program was established in 2006 to provide quality assurance services. The BED PT program consisted of two components, an international program provided by the U.S. Centers for Disease Control and Prevention from 2006 and a domestic program started by the National HIV/HCV Reference Laboratory in 2011. Each PT panel consisted of eight coded specimens distributed to participating laboratories semi-annually, and testing results were collected and analyzed. The number of participating laboratories increased progressively from 2006 to 2012. The Chinese HIV-1 incidence laboratory network performed satisfactorily both in international and domestic PT programs. We also demonstrated that the BED assay was highly reproducible among participating laboratories. Our success and lessons learned can be readily replicated in other countries or regions contemplating the establishment of a PT program for assay-based HIV incidence estimation.     Description: As with quantum spin liquids based on two-dimensional triangular and kagome lattices, the two-dimensional honeycomb lattice with either a strong spin-orbital coupling or a frustrating second-nearest-neighbor coupling is expected to be a source of candidate quantum spin liquids. An ammonium salt [(C3H7)3NH]2[Cu2(C2O4)3](H2O)2.2 containing hexagonal layers of Cu2+ was obtained from solution. No structural transition or long-range magnetic ordering was observed from 290 K to 2 K from single crystal X-ray diffraction, specific heat and susceptibility measurements. The anionic layers are separated by sheets of ammonium and H2O with distance of 3.5 Å and no significant interaction between anionic layers. The two-dimensional honeycomb lattice is constructed from Jahn-Teller distorted Cu2+ and oxalate anions, showing a strong antiferromagnetic interaction between S = 1/2 metal atoms with θ = −120 (1) K. Orbital analysis of the Cu2+ interactions through the oxalate-bridges suggests a stripe mode pattern of coupling with weak ferromagnetic interaction along the b axis, and strong antiferromagnetic interaction along the a axis. Analysis of the magnetic susceptibility shows that it is dominated by a quasi-one-dimensional contribution with spin chains that are at least as well isolated as those of well-known quasi-one-dimensional spin liquids.     Description: The mechanical property of extracellular matrix and cell-supporting substrates is known to modulate neuronal growth, differentiation, extension and branching. Here we show that substrate stiffness is an important microenvironmental cue, to which mouse hippocampal neurons respond and integrate into synapse formation and transmission in cultured neuronal network. Hippocampal neurons were cultured on polydimethylsiloxane substrates fabricated to have similar surface properties but a 10-fold difference in Young's modulus. Voltage-gated Ca2+ channel currents determined by patch-clamp recording were greater in neurons on stiff substrates than on soft substrates. Ca2+ oscillations in cultured neuronal network monitored using time-lapse single cell imaging increased in both amplitude and frequency among neurons on stiff substrates. Consistently, synaptic connectivity recorded by paired recording was enhanced between neurons on stiff substrates. Furthermore, spontaneous excitatory postsynaptic activity became greater and more frequent in neurons on stiff substrates. Evoked excitatory transmitter release and excitatory postsynaptic currents also were heightened at synapses between neurons on stiff substrates. Taken together, our results provide compelling evidence to show that substrate stiffness is an important biophysical factor modulating synapse connectivity and transmission in cultured hippocampal neuronal network.     Description: The role of bone marrow-derived mesenchymal stem cells(BMSCs)in the pathogenesis and therapy of osteoporosis has drawn increasing attention in recent years. In the development of osteoporosis, it has been demonstrated that many changes occurred in the behavior of BMSCs. For example, the biological system of FasL pathways mediated differentiation of ERK and GSK-3β-catenin pathway was damaged. Here we found that 0.35 mg/L Licochalcone A (L-A) had a strong effect in increasing the osteogenic differentiation and mineralization of BMSCs both in vivo and in vitro by up-regulating FasL and further playing a role in regulating the ERK and GSK-3β-catenin systems. It has also demonstrated that the administration of L-A could restore the biological function of the damaged BMSCs differentiation by recovering or protecting bone mass in a disease state through activating the endosteal bone formation and partially inhibiting bone resorption in acute estrogen deficiency model. Results of our study suggested that careful titration of MSC was response to L-A and up-regulated FasL pathways mediating differentiation of ERK and GSK-3β-catenin biological systems under disease state in vivo, restore the impaired function, is one of the ways of L-A relieve or treatment osteoporosis.     Description: We report on ordered nanoporous films exhibiting a unique magneto-plasmon based response, fabricated by nanosphere-assisted physical deposition. This work focuses on multi-layer Ag/CoFeB/Ag films as examples of such structures. Their microstructure dependent magnetic properties, localized surface plasmon resonance (LSPR) and magneto-optical Kerr effect were investigated. The observed effects of nanopores and Ag layers on the magnetic properties indicate the synergistic interaction between nanopores and Ag layers leading to an enhancement of the ferromagnetic character of the CoFeB film. LSPR spectra reveal that the introduction of Ag layers enhances the light transmission in the nanoporous CoFeB films (having pore sizes exceeding the wavelength of light) due to an enhanced interaction of light with surface plasmons. Periodic nanoporous Ag/CoFeB/Ag films covered by Ag capped nanospheres show a much larger extinction than uncovered nanoporous Ag/CoFeB/Ag films. The correlation between the magneto-optical Kerr effect and the nanostructures suggests a field-tunable Kerr effect owing to the magneto-electric coupling between the magnetic layer and the Ag layers, which is enhanced by the nanopores.     Description: In this paper, we report the in situ growth of NixCu1-x (x = 0, 0.25, 0.50, 0.75 and 1.0) alloy catalysts to anchor and decorate a redox-reversible Nb1.33Ti0.67O4 ceramic substrate with the aim of tailoring the electrocatalytic activity of the composite materials through direct exsolution of metal particles from the crystal lattice of a ceramic oxide in a reducing atmosphere at high temperatures. Combined analysis using XRD, SEM, EDS, TGA, TEM and XPS confirmed the completely reversible exsolution/dissolution of the NixCu1-x alloy particles during the redox cycling treatments. TEM results revealed that the alloy particles were exsolved to anchor onto the surface of highly electronically conducting Nb1.33Ti0.67O4 in the form of heterojunctions. The electrical properties of the nanosized NixCu1-x/Nb1.33Ti0.67O4 were systematically investigated and correlated to the electrochemical performance of the composite electrodes. A strong dependence of the improved electrode activity on the alloy compositions was observed in reducing atmospheres at high temperatures. Direct electrolysis of CO2 at the NixCu1-x/Nb1.33Ti0.67O4 composite cathodes was investigated in solid-oxide electrolysers. The CO2 splitting rates were observed to be positively correlated with the Ni composition; however, the Ni0.75Cu0.25 combined the advantages of metallic nickel and copper and therefore maximised the current efficiencies.     Description: This study demonstrates both prokaryotic and eukaryotic community structures and dominant taxonomies in different positions of the greatest estuary reservoir for drinking water source in the world in four seasons of one year using 454 pyrosequencing method with total of 312,949 16S rRNA and 374,752 18S rRNA gene fragments, including 1,652 bacteria OTUs and 1,182 fungus OTUs. During winter and spring, the community composition at the phylum level showed that microorganisms had similar structures but their quantities were different. Similarly, obvious changes at the genus level were observed among the samples taken in winter and spring between summer and fall. Microorganisms located the reservoir inlet were founded to be different from those in rear at both phylum and genus level. Air temperature had a stronger effect than sampling location on the microbial community structure. Total nitrogen and dissolved oxygen were algae-monitoring indicators during the whole year. Moreover, Bacillus was an efficient indicator during summer and autumn for bacteria OTUs. Scientific Reports 4 doi: 10.1038/srep06966

    Description: Human Rad 9 (hRad9), part of the Rad9-Hus1-Rad1 complex plays an important role in DNA damage repair as an up-stream regulator of checkpoint signaling, however little is known about its role in response to chemotherapy of breast cancer and whether hRad9 inhibition can potentiate the cytotoxic effects of chemotherapy on breast cancer cells remains to be elucidated. Fifty cases of breast cancer receiving neoadjuvant therapy were collected. All these cases were revised and classified into chemotherapy sensitive (CS) or chemotherapy resistant (CR) group according to the Miller and Payne (MP) grading system. Immunohistochemically, hRad9 positive tumours showed nuclear and/or cytoplasmic staining. hRad9 over-expression was associated with an impaired neoadjuvant chemotherapy response. A significant correlation was found between expression of hRad9 and Cyclin D1. In vitro, hRad9 was knocked down using siRNA in breast cancer cell line MCF-7 and MDA-MB-231. Deregulated expression of Rad9 accompanied by down expression of chk1 enhanced the sensitivity of human breast cancer cells to doxorubicin. Our work suggests that hRad9 might be a potential predictor for the response to chemotherapy in patients with breast cancer and its clinical value as a target for improving chemosensitivity needs further exploration.     Description: Clioquinol is an anti-microbial drug, and it was recently found to induce cancer cell death. In the present study, clioquinol was found to trigger autophagy by inducing LC3 lipidation and autophagosome formation which was abolished by an autophagy inhibitor 3-methyladenine. Further study showed clioquinol displayed no effects on PI3KC3 or Beclin 1 expression but downregulated the expression and the enzymatic activity of mammalian target of Rapamycin (mTOR), a critical modulator of autophagy. Moreover, clioquinol inhibited the catalytic activity of the mTOR complex 1, thus suppressing phosphorylation of P70S6K and 4E-BP1, two major proteins associated with autophagy in the mTORC1 signaling pathway. Clioquinol induced leukemia and myeloma cell apoptosis, however, addition of autophagy inhibitor 3-methyladenine attenuated this kind of cell death. Therefore, this study demonstrated that clioquinol induces autophagy in associated with apoptosis in leukemia and myeloma cells by disrupting mTOR signaling pathway.
