    Description: We present the low-resolution spectroscopic and UBVRI broad-band photometric investigations of the Type IIb supernova (SN) 2011fu, discovered in UGC 01626. The photometric follow-up of this event was initiated a few days after the explosion and covers a period of about 175 d. The early-phase light curve shows a rise, followed by steep decay in all bands, and shares properties very similar to that seen for SN 1993J, with a possible detection of the adiabatic cooling phase. Modelling of the quasi-bolometric light curve suggests that the progenitor had an extended (~1  x 10 13 cm), low-mass (~0.1 M ) H-rich envelope on top of a dense, compact (~2  x 10 11 cm), more massive (~1.1 M ) He-rich core. The nickel mass synthesized during the explosion was found to be ~0.21 M , slightly larger than that seen for other Type IIb SNe. The spectral modelling performed with synow suggests that the early-phase line velocities for H and Fe ii features were ~16 000 and ~14 000 km s –1 , respectively. Then, the velocities declined up to day +40 and became nearly constant at later epochs.

    Description: [1]  Field studies of watershed carbon fluxes and budgets are critical for understanding the carbon cycle, but the role of deep regional groundwater is poorly known and field examples are lacking. Here we show that discharge of regional groundwater into a lowland Costa Rican rainforest has a major influence on ecosystem carbon fluxes. This influence is observable through chemical, isotopic, and flux signals in groundwater, surface water, and air. Not addressing the influence of regional groundwater in the field measurement program and data analysis would give a misleading impression of the overall carbon source or sink status of the rainforest. In quantifying a carbon budget with the traditional " small watershed" mass-balance approach, it would be critical at this site and likely many others to consider watershed inputs or losses associated with exchange between the ecosystem and the deeper hydrogeological system on which it sits.

    Description: Phytochromes (phy) are red/far-red–absorbing photoreceptors that regulate the adaption of plant growth and development to changes in ambient light conditions. The nuclear transport of the phytochromes upon light activation is regarded as a key step in phytochrome signaling. Although nuclear import of phyA is regulated by the transport facilitators far red elongated hypocotyl 1 (FHY1) and fhy1-like, an intrinsic nuclear localization signal was proposed to be involved in the nuclear accumulation of phyB. We recently showed that nuclear import of phytochromes can be analyzed in a cell-free system consisting of isolated nuclei of the unicellular green algae Acetabularia acetabulum. We now show that this system is also versatile to elucidate the mechanism of the nuclear transport of phyB. We tested the nuclear transport characteristics of full-length phyB as well as N- and C-terminal phyB fragments in vitro and showed that the nuclear import of phyB can be facilitated by phytochrome-interacting factor 3 (PIF3). In vivo measurements of phyB nuclear accumulation in the absence of PIF1, -3, -4, and -5 indicate that these PIFs are the major transport facilitators during the first hours of deetiolation. Under prolonged irradiations additional factors might be responsible for phyB nuclear transport in the plant.

    Description: Legume plants host nitrogen-fixing endosymbiotic Rhizobium bacteria in root nodules. In Medicago truncatula, the bacteria undergo an irreversible (terminal) differentiation mediated by hitherto unidentified plant factors. We demonstrated that these factors are nodule-specific cysteine-rich (NCR) peptides that are targeted to the bacteria and enter the bacterial membrane and cytosol. Obstruction of NCR transport in the dnf1-1 signal peptidase mutant correlated with the absence of terminal bacterial differentiation. On the contrary, ectopic expression of NCRs in legumes devoid of NCRs or challenge of cultured rhizobia with peptides provoked symptoms of terminal differentiation.     Description: The mechanisms underlying the low efficiency of reprogramming somatic cells into induced pluripotent stem (iPS) cells are poorly understood. There is a clear need to study whether the reprogramming process itself compromises genomic integrity and, through this, the efficiency of iPS cell establishment. Using a high-resolution single nucleotide polymorphism array, we compared copy number variations (CNVs) of different passages of human iPS cells with their fibroblast cell origins and with human embryonic stem (ES) cells. Here we show that significantly more CNVs are present in early-passage human iPS cells than intermediate passage human iPS cells, fibroblasts or human ES cells. Most CNVs are formed de novo and generate genetic mosaicism in early-passage human iPS cells. Most of these novel CNVs rendered the affected cells at a selective disadvantage.
