    Notes: Summary A strain of Aspergillus niger adapted to grow on gluconate is cultivated on a medium containing gluconic acid lactone as the sole carbon source. Nutritional requirements of the adapted mold were studied. The organism shows optimum growth at 30° C in a medium containing per litre 20 g of gluconic acid lactone, 800 mg of NH4NO3, 100 mg of MgSO4, 7 H2O and 270 mg of KH2PO4 having pH 2.0. Mycelial growth reaches its maximum on the third day. Comparative growth responses of the adapted and parent strains on gluconate, glucose and sucrose were studied.

    Notes: Zusammenfassung Untersuchungen derr II-Region desT 2-Bakteriophagen. Es wurde eine genetische Analyse derr II-Region desT 2-Bakteriophagen durchgeführt zur Feststellung, ob die Befunde, die sich auf die Funktionsteile innerhalb derr II-Region desT 4-Phagen beziehen, in bezug auf dier II-Region desT 2-Phagen verallgemeinert werden können. Die Mutanten wurden mit Hilfe von 5-Bromouracil und Akridineorange isoliert und dieh + r-Mutanten nach demselben Schema wie fürT 4-Phagen weiter differenziert. Sechzig ausgewählte Mutanten wurden nach funktionaler Identität geprüft, was zur Feststellung führte, dass die gesamte Region als eine Einheit funktioniert.

    Notes: Summary Two characters, “Penicillin resistance” and “fructose utilizing ability”, were transferred in the recipient Rhizobium cowpea (source—Ground nut roots) with the help of the DNA extracted from Rhizobium phaseoli culture (source—French bean roots). The recipient culture enters the logarithmic growth phase at the end of 60 min. The percentage transformation frequency is maximum at the beginning of the exponential phase.

    Notes: Summary Sorbitol dehydrogenase has been purified about 26 fold from a strain of Aspergillus niger, growing on sorbitol as the sole source of carbon. An absolute specificity of this enzyme for sorbitol, fructose, NAD and NADH was observed. The K m for sorbitol and fructose were found to be 9.8x10-5 M and 6.6x10-4 M respectively. The enzyme was inhibited by pCMB, NaF and other metal ions studied. The enzyme was slightly activated by Fe+++.

    Notes: Summary Phosphogluconate dehydrogenase was purified about 20 fold from an adapted strain of Aspergillus niger cultivated on gluconate as the sole carbon source. The enzyme showed pH optimum between 7.0 to 7.6. The enzyme was activated by glycyl glycine buffer and magnesium ions. Mn++, Ba++, Ca++ cysteine and reduced glutathione activated the enzyme, while Co++, Zn++, Cu++, Hg++, PCMB and monoiodoacetate inhibit the enzyme. K m values for the substrates, 6-phosphogluconate and NADP, were found to be 1.66×10−4 m and 8.3×10−5 m respectively.

    Notes: Summary A citric acid accumulating strain of Aspergillus niger adapted to grow on gluconic acid lactone as sole carbon source was studied with regards to the enzymatic changes occurring during its adaptive growth and compared with those of the parent strain cultivated on sucrose. Glucose oxidase, glucose dehydrogenase, gluconate dehydrogenase and enzymes of Entner Doudoroff pathway could not be detected in the strain. Gluconokinase was detected in the strain and its inducible nature was established. An adaptive increase of gluconokinase, phosphogluconate dehydrogenase and ribose phosphate isomerase was observed. While the levels of the above enzymes were significantly higher, that of glucose 6-phosphate dehydrogenase were significantly low indicating the repression of the enzyme. Levels of hexokinase and fructose diphosphate aldolase remained more or less the same as that of parent strain. It was inferred from the results that gluconate metabolism occurs via the pentose phosphate pathway following initial phosphorylation.

    Notes: Summary Enzyme make-up of the adapted strain of Aspergillus niger was studied and compared with that of the parent strain. Sorbitol dehydrogenase and fructokinase were induced while glucokinase was repressed in the adapted strain. Intracellular accumulation of fructose in the parent strain failed to induce fructokinase suggesting a sequential induction of sorbitol dehydrogenase and fructokinase in the adapted strain. Studies on the enzymes of the hexose monophosphate pathway suggested elevated rate of operation of this pathway in the adapted strain. The levels of Fructose-1,6-diphosphate aldolase and Isocitrate dehydrogenase in the adapted and the parent strains and their implications on the citric acid producing capacities have been discussed. Sorbitol kinase could not be detected in the adapted strain.
