    Notizen: Abstract Membrane fractions were isolated from dark grown cotyledons of Phaseolus auneus by differential and sucrose density gradient centrifugation. Endoplasmic reticulum-, Golgi apparatus- and plasma membrane-rich fractions were identified by their respective enzymic activities and tested for their ability to transfer glucose from UDP-glucose to endogenous sterols to form steryl glucosides. The glucosyltransferase activity was shown to be located mainly at the plasma membrane.

    Notizen: Abstract Several arguments are in favor of a function of snRNA in the processing of premessenger RNA. A large fraction of snRNA is localized in hnRNP which are assumed to be the site of processing. The different snRNA species are not bound to hnRNP in a unique manner but are associated with both proteins and hnRNA which suggests the possibility of metabolic exchanges in the course of processing. There is approximately 1–2 molecules of snRNA per individual hnRNP. We reexamined the possibility that U1A RNA might serve for the alignment of the extremities of the intron sequences of premessenger RNA insuring correct condition for cutting and splicing. We found that only a UCCA (3′→5′) sequence at position 8–11 of U1A RNA was complementary to an AG-GU (5′→3′) around a putative splice point for 69 different introns sequenced so far. On the basis of secondary structure of U1A RNA, the UCCA sequence would be available for hybridization. The UCCA sequence is also present in U2 RNA and 4.5 S RNAI. It might associate with AG-GU in a manner similar to that of codon-anticodon, the stability of the complex being insured by the configuration of hnRNP. The possible formation of larger hybrids stable by themselves is unlikely upon examination of the nucleotide sequence of various introns adjacent to the splice point. As there is no direct experimental evidence for the function of snRNA in splicing, these considerations are speculative at the present time. The possibility that adenovirus encoded VA RNA would play a role in splicing was also examined. Various arguments suggest that this possibility is rather remote.

    Notizen: Abstract Two amino-acid auxotrophic cell clones ofHyoscyamus muticus, VA5 (His-) and VIIIB9 (Trp-), isolated in a previous experiment have been characterised quantitatively. Studies of growth in the presence and absence of histidine and tryptophan and an examination of dose-response relationships for the two amino acids confirmed the strict auxotrophy of both cell lines. No revertants to prototrophy were detected in either cell line after more than two years in culture. N-methyl-N′-nitro-nitrosoguanidine did not induce reversion in VA5 cell populations. Wild-type aggregates mixed in various combinations with VA5 and VIIIB9 cells could be recovered after plating in selective conditions. No cross-feeding was detected, either between wild-type and auxotrophic cells or between the auxotrophic lines themselves. Both variants were recloned by protoplast culture. All protoplast-derived clones were auxotrophic. The auxotrophic phenotypes behaved as recessive traits in protoplast-fusion experiments.

    Notizen: Abstract Protoplasts of the nicotinamide-deficient Hyoscyamus muticus cell line nic− IVH2 and of the nitrate reductase-deficient Nicotiana tabacum cell line NR− cnx 68 were induced to fuse. Selection for putative interspecific hybrid clones was via auxotroph complementation. Controls included tests for cross-feeding, cross inhibition, PEG-induced variation, culture-induced variation, reversion, viability, delayed selection. Protrotrophic cell lines were recovered exclusively from PEG-treated mixtures of both protoplast types. The putative hybrid clones grew independent of exogenously supplied auxins and cytokinins, and at a faster rate than either parent. The morphogenic potential of different clones varied from ‘non-morphogenic’ to ‘regeneration of fertile plants’. Indirect evidence for the hybrid nature of the clones is provided from a) tight selection, b) hormone-independent growth, c) hybrid vigour, d) extreme morphological variation, e) isoenzyme bands from both parents, f) morphogenic potential. Definite proof for the hybrid nature was, however, provided from species-specific DNA hybridization. Chimerism could be excluded since only the large subunit of Hyoscyamus muticus ribulose bisphosphate carboxylase was found and since species-specific DNA hybridization identified clones which gave no Nicotiana tabacum signal.

    Notizen: Summary Plants of three common wheat (Triticum aestivum L. em. Thell) cultivars and one randomly selected doubled-haploid line derived by anther culture from each of the three cultivars were each grown in three environments, a field environment, a greenhouse environment, and a growth chamber environment. Anthers containing largely miduninucleate to late uninucleate microspores were cultured and calli were induced to regenerate plants in order to assess the effects of cultivar, cultivar family (cultivar and corresponding doubled-haploid derivative), anther-donor plant environment, and cultivar X environment interaction on androgenic responses. Large differences in response were observed among cultivars as well as between cultivars and doubled-haploids. Differences between cultivar and doubled-haploid within cultivar family usually resulted from higher frequency of response in the cultivar, contrary to the hypothesis that anther culture per se constitutes a general selective device for superior androgenic responses. Also, in a second experiment, anther callusing frequency was greater in the cultivar ‘Kitt’ than in any of five unique doubled-haploid lines derived from ‘Kitt’. Significant effects were also observed in the first experiment for the interactions of cultivar family X environment as well as doubled-haploid vs. cultivar X environment, although the effect of environment itself was less significant than these interactions.

    Notizen: Summary Frequency of callus formation in wheat (Triticum aestivum L. em. Thell) anthers cultured in vitro and the frequency of subsequent plantlet formation from such calli were examined in a diallel population produced from five inbred spring wheat cultivars. Two of the five cultivars were believed to possess relatively high frequencies of response and the other three relatively low response frequencies, based on previous studies. General and specific combining abilities were estimated and found to be highly significant for both traits. Reciprocal effects were also estimated and were highly significant for both traits. Of the 25 entries, the largest mean callus formation frequency was observed on anthers of ‘Kitt’ x ‘Olaf’, while the largest mean plantlet formation frequency was observed using anthers of the cultivar, ‘Fielder’. No significant correlation was observed between the two traits. Heritability estimates in the range of 0.6–0.7 suggested, however, that both traits were highly heritable, so that rapid gain from selection for these traits should be possible. Current limitations due to genetic variation in responses therefore may not constitute a major obstacle to application of in vitro techniques by wheat breeders.

    Notizen: Summary Mitochondrial DNA (mtDNA) restriction patterns were studied in mutant, cybrid and somatic hybrid plants regenerated from Nicotiana protoplasts. It has been shown that neither components of the culture media used for protoplast culture and plant regeneration, nor the antibiotics streptomycin and lincomycin used for the mutant selection induced alterations in the mtDNA. No rearrangements were detected in the mtDNA of plants derived from homoplasmic fusions where the mtDNA of the parents was identical as judged by mtDNA restriction patterns. There were rearrangements, however, in the mtDNA of each of the cybrid plants derived from heteroplasmic fusions. Restriction patterns generated by BamHI and SalI restriction endonucleases were different from those of both parents, and were composed of parental and non-parental fragments.

    Notizen: Summary Systemic administration of monosodium-1-gluta-mate by single injections of 4 mg/g body weight in infant rats (2–10 days of age) results in acute swelling of cytoplasm and nuclear pyknosis of neurons in the stratum zonale and stratum griseum superficiale of the superior colliculus. Multiple daily doses of 4 mg/g body weight monosodium-1-glutamate result in an almost complete loss of neurons in these two superficial layers. The deeper layers appear not to be affected. No pathological effects were observed in the lateral geniculate body or pretectal complex. Light-and electron-microscopic studies reveal that the optic nerves are remarkably shrunken and many myelinated as well as unmyelinated axons are lost. Injection of 3Hproline into the vitreous body of one eye results in limited transport to the suprachiasmatic nucleus, lateral geniculate body and to lateral portions of the superior colliculus. The small percentage of intact axons in the optic nerve, as well as the limited proline transport from the eye, suggest that administration of monosodium-1-glutamate leaves intact some optic fibers, a portion of which belongs to the retinohypothalamic tract.
