    Notes: PC12 cells show a classical volume regulatory process when submitted to hypo-osmotic conditions. The present study examined the effects of such osmotic shock on the structural organization of different cytoskeletal elements. Results were obtained by use of different light and electron microscopy techniques combined with immunostaining methods. It appeared that the osmotically induced changes in cell volume were concomitant with important modifications in the organization of the microfilament network. Microfilaments concentrated in the perinuclear area, leaving only radial extensions of poorly organized structures in the cytoplasm. The latter were the only actin structures immunologically stained in the cytoplasm and seemed to anchor to the plasma membrane. Measurements of the fluorescence intensity of PC12 cells treated with FITC-labeled phalloidin indicated a progressive depolymerization, followed by a repolymerization of F-actin. This occurs in parallel with microfilament reorganization and volume regulatory processes. The appearance of microfilament reorganization was a function of both the incubation period and the amplitude of the osmolarity changes. During the first minutes of osmotic shock, a decrease was observed in the density and length of microvilli, which normally cover the PC12 cell surfaces, suggesting an early reorganization of the underlying microfilament network. Microtubules and intermediate filament networks were not affected by the hypo-osmotic conditions. © 1994 Wiley-Liss, Inc.

    Notes: The present study is concerned with the improvement of the previously proposed ‘shifted integration technique’ for the plastic collapse analysis of framed structures using the linear Timoshenko beam element or the cubic beam element based on the Bernoulli-Euler hypothesis. In the newly proposed ‘adaptively shifted integration technique’, the numerical integration points for the evaluation of the stiffness matrices are automatically shifted immediately after. the occurrence of plastic hinges according to the previously established relations between the locations of numerical integration points and those of plastic hinges. By using the adaptively shifted integration technique, sufficiently accurate solutions can be obtained in the non-linear frame analysis by two-linear-element or only one-cubic-element idealization for each structural member. The present technique can easily be implemented in the existing finite element codes utilizing the linear or the cubic beam element.

    Notes: Discrimination among the four γ-hydroxyornithine diastereomers was studied by fast atom bombardment mass spectrometry (FABMS). It is impossible to distinguish among the four diastereomers of this amino acid by positive- and negative-ion FAB and collisionally activated dissociation MS, but benzyloxycarbonyl group protection of the α- and δ-amino groups in γ-hydroxyornithine allows differentiation among the diastereomers in negative-ion FABMS. The negative-ion mass spectra of benzyloxycarbonyl-protected γ-hydroxyornithine diastereomers showed differences among the abundances of the molecule ion [M - H]-, the dehydrated ion [M  -  H  -  H2O]- due to the loss of the γ-hydroxyl group and the fragment ions formed from both [M  -  H]- and [M  -  H  -  H2O]- ions. On the other hand, no difference was found between the fragmentations of the benzyloxycarbonyl-protected enantiomers of ornithine in negative-ion FABMS. These results indicate that the orientation of the γ-hydroxyl group and the existence of two benzene rings in the benzyloxycarbonyl group are important factors which are responsible for the fragmentations of the four benzyloxycarbonyl-protected γ-hydroxyornithine diastereomers in negative-ion FABMS. These studies also showed that the negative-ion FABMS for benzyloxycarbonyl-protected γ-hydroxyornithine diastereomers is a useful method for determining the configuration of each diastereomer of γ-hydroxyornithine.

    Notes: The effect on the fragmentations of N-benzyloxycarbonyl-protected tripeptide ethyl esters due to the existence of L-proline in the gas phase was examined by the collisional-activated decomposition of the deprotonated molecule and the fragment ions produced by the cleavage of the tripeptide derivatives containing the neutral amino acids (L-alanine, L-leucine and L-phenylalanine) and L-proline, in which changes in both the numbers and positions of the prolyl residues were observed, in negative-ion fast atom bombardment mass spectrometry. The cleavage patterns of these ions in the collisional-activated decomposition mass spectra were observed to depend on the numbers and positions of prolyl residues in the peptide derivatives. These results indicate that the conformational differences in the peptide derivatives due to the existence of L-proline affect the decomposition of the ions containing the neutral amino acids in the gas phase.

    Notes: The indirect stimulation of macrophages to produce nitrite was examined by using the macrophage cell line J774. J774 spontaneously produced nitrite, when cultured at high concentration. J774 cultured in low concentration ( 〈 104 cells in 100 μl) barely produced nitrite. J774 cultured in low concentration produced a large amount of nitrite by the co-culture of nonadherent spleen cells or nonadherent peritoneal exudate cells, which were stimulated with con A, anti-CD3, or staphylococcal enterotoxin A. J774 (BALB/c derived: H-2d) cultured with either syngeneic (BALB/c) or allogeneic (B6; H-2b B10BR; H-2k) nonadherent lymphocytes, which were stimulated with conA or anti-CD3, produced nitric oxide. However, J774 produced nitric oxide by stimulation with SEA only when co-cultured with SEA-reactive T lymphocytes. Peritoneal exudate cells from mice, which did not proliferate by the stimulation of conA or anti-CD3, proliferated well by the addition of L-arginine homologue, NG-monomethyl-L-arginine. The proliferation of nonadherent peritoneal exudate cells stimulated with conA or anti-CD3 was suppressed by the addition of peritoneal macrophages. This suppression was abolished by the addition of NG-monomethyl-L-arginine.

    Notes: Insulating to semiconducting transformation was observed for surface layers of the transition metal oxides exposed to 1.5-15 keV He+, Ar+ and Xe+ ions. The surface electrical conductivity increased with the fluence of the ion beams and reached ∼10Ω-1 cm-1 at a fluence of 1 × 1017 Ar+ cm-2. Core-line splittings in the metal 2p region and preferential losses of the surface oxygen observed in the x-ray photoelectron spectra give evidence of the formation of the reduced chemical species Ti(II), Ti(III) and V(III). The ion bombardment also brought about the appearance of a new peak near the Fermi level in the UV photoelectron spectra. These observations suggest that electrons in the O 2p orbital are transferred to the 3d-derived non-bonding level by the losses of atomic oxygen and thus behave as charge carriers, which causes an increase in electrical conductivity.

    Notes: Methods of evaluation of the chemical composition of tantalum oxide films and of the interface between tantalum oxide films and Si substrates were studied by means of sputter-assisted Auger electron spectroscopy (AES). Tantalum oxide films 10-50 nm thick were grown on Si single-crystal substrates by low-pressure chemical vapour deposition (LPCVD). It has been found that reduction of the oxides is induced by ion sputtering during Auger depth profiling. Therefore we optimized the ion-sputtering conditions to minimize the reduction rate. Then, using stoichiometric β-Ta2O5 as reference, it was confirmed that as-deposited tantalum oxide films were oxygen-deficient and that the oxygen content of these films was increased by subsequent annealing in an O2 ambient. By means of Auger depth profiling, we evaluated the thickness of silicon native oxides formed at the Ta2O5/Si interface as 1.5-2 nm. Moreover, we revealed the oxidation mechanism of a silicon nitride intermediate layer (e.g. 10 nm thick) deposited between tantalum oxide film and an Si substrate during heat treatment at 900-1000°C in O2.

A significant difference was found among the intensities of the fragment ions formed by cleavage of the benzyloxycarbonyl group, depending on the numbers and positions of prolyl residues in the derivatives. These results suggest that the fragmentations of the peptide derivatives in negative-ion fast atom bombardment mass spectrometry depend on the conformational differences in the peptide derivatives due to the existence of proline, and permit the prediction of the positions of proline in the peptides.
