    Notizen: Yeast flocculation involves binding of surface lectins to carbohydrate receptors on neighbouring cell walls. Brewing strains of Saccharomyces cerevisiae normally become flocculent in the stationary phase of growth. This paper presents evidence that lectins are synthesized in exponential phase, inserted into the cell wall, and activated later at the time of flocculation onset.Cycloheximide failed to prevent flocculation unless it was added in early growth; with later additions progressively larger degrees of flocculation occurred. Flocculation onset was delayed by cycloheximide but was otherwise cycloheximide insensitive. Preflocculent cells could be artificially activated to full flocculation by heat. Artificial activation of samples from growing yeast cultures confirmed the progressive synthesis of lectins throughout exponential growth. Pronase E treatment of whole cells prior to heating prevented any activation of flocculation.It was concluded that lectins were synthesized continuously from an early stage of growth and rapidly inserted into the cell wall (accessible by pronase E), where they remained inactive for up to 14 h, before being activated at flocculation onset by an as-yet unknown mechanism. It was found that lectin synthesis and activation occurred in all brewing strains tested.

    Notizen: Saccharomyces cerevisiae contains at least four PRS genes, all of which have been cloned and sequenced. Each of the four derived amino acid sequences have more than 60% similarity to the corresponding polypeptides of man, rat, Escherichia coli and Salmonella typhimurium. The PRS1 gene maps on chromosome XI, PRS2 on chromosome V, PRS3 on chromosome VIII and PRS4 on chromosome II. One member of this gene family, PRS1, contains a region of non-homology (NHR) shown by cDNA cloning and sequencing not to be an intron. The results presented here suggest that the presence of this NHR is not detrimental to the function of the gene. To date the possibility of protein splicing can be neither proven nor disputed. The sequences submitted to the EMBL data library are available under the following accession numbers: PRS1 (X70069), PRS2 (X74414) and PRS3 (X74415).

    Beschreibung: The Indus River is the only major drainage in the western Himalaya and delivers a long geological record of continental erosion to the Arabian Sea, which may be deciphered and used to reconstruct orogenic growth if the modern bedload can be related to the mountains. In this study we collected thermochronologic data from river sediment collected near the modern delta. U-Pb ages of zircons spanning 3 Gyr show that only ~5% of the eroding crust has been generated since India-Asia collision. The Greater Himalaya are the major source of zircons, with additional contributions from the Karakoram and Lesser Himalaya. The 39Ar/40Ar dating of muscovites gives ages that cluster between 10 and 25 Ma, differing from those recorded in the Bengal Fan. Biotite ages are generally younger, ranging 0–15 Ma. Modern average exhumation rates are estimated at ~0.6 km/m.y. or less, and have slowed progressively since the early Miocene (~20 Ma), although fission track (FT) dating of apatites may indicate a recent moderate acceleration in rates since the Pliocene (~1.0 km/m.y.) driven by climate change. The 39Ar/40Ar and FT techniques emphasize the dominance of high topography in controlling the erosional flux to the ocean. Localized regions of tectonically driven, very rapid exhumation (e.g., Nanga Parbat, S. Karakoram metamorphic domes) do not dominate the erosional record.

    Beschreibung: The Early Ordovician Grampian Orogeny in the British Isles represents a classic example of collision between an oceanic island arc and a passive continental margin, starting around 480 Ma. The South Mayo Trough in western Ireland preserves a complete and well-dated sedimentary record of arc collision. We sampled sandstones and conglomerates from the Rosroe, Maumtrasna and Derryveeny Formations in order to assess erosion rates and patterns during and after arc collision. U-Pb dating of zircons reveals a provenance dominated by erosion from the upper levels of the Dalradian Supergroup (Southern Highland and Argyll Groups), with up to 20% influx from the colliding arc into the Rosroe Formation, but only 6% in the Maumtrasna Formation (~465 Ma). The 24 dominant source regions lay to the northeast (e.g. in the vicinity of the Ox Mountains, 50 km distant, along strike). The older portions of the North Mayo Dalradian and its depositional basement (the Annagh Gneiss Complex) do not appear to have been important sources, while the Connemara Dalradian only plays a part after 460 Ma, when it supplies the Derryveeny Formation. By this time all erosion from the arc had effectively ceased and exhumation rates had slowed greatly. The Irish Grampian Orogeny parallels the modern Taiwan collision in showing little role for the colliding arc in the production of sediment. Negligible volumes of arc crust are lost because of erosion during accretion to the continental margin.

    Beschreibung: The Indus Delta is constructed of sediment eroded from the western Himalaya and since 20 ka has been subjected to strong variations in monsoon intensity. Provenance changes rapidly at 12-8 ka, although bulk and heavy mineral content remains relatively unchanged. Bulk sediment analyses shows more negative {varepsilon}Nd and higher 87Sr/86Sr values, peaking around 8-9 ka. Apatite fission track ages and biotite Ar-Ar ages show younger grains ages at 8-9 ka compared to at the Last Glacial Maximum (LGM). At the same time {delta}13C climbs from -23 to -20{per thousand}, suggestive of a shift from terrestrial to more marine organic carbon as Early Holocene sea level rose. U-Pb zircon ages suggest enhanced erosion of the Lesser Himalaya and a relative reduction in erosion from the Transhimalaya and Karakoram since the LGM. The shift in erosion to the south correlates with those regions now affected by the heaviest summer monsoon rains. The focused erosion along the southern edge of Tibet required by current tectonic models for the Greater Himalaya would be impossible to achieve without a strong summer monsoon. Our work supports the idea that although long-term monsoon strengthening is caused by uplift of the Tibetan Plateau, monsoon-driven erosion controls Himalayan tectonic evolution.     Beschreibung: This study provides 39 new thermochronometric analyses from the western part of the Greater Caucasus, a region in which existing data are extremely limited and of questionable quality. The new results are consistent with field studies that identify Triassic to Middle Jurassic (Cimmerian) and Oligo-Miocene (Alpine) orogenic erosional events. An inverse relationship between the fission track and depositional ages of Oligo-Miocene sedimentary samples also implies some degree of Eocene erosion of the Greater Caucasus and intermediate sediment storage. Cooling ages and field relationships within the core of the range, west of Mt Elbrus, require ~5 km of Permo-Triassic exhumation and restrict the overall amount of Cenozoic exhumation to ~2.5 km. Current exhumation rates are typically low, and do not support a Plio-Pleistocene increase in climate-driven denudation. High (~1 km Ma-1) rates of exhumation are restricted to the southern flank of the range in northwest Georgia. Despite a general lack of significant seismicity within the study region, this exhumation peak is close to the largest instrumentally recorded earthquake in the Caucasus (Ms = 7.0). This may suggest that exhumation is associated with the decoupling of the sedimentary succession from its crystalline basement in the southern part of the range and the inversion of the largely Jurassic fill of the Greater Caucasus basin. Rates of exhumation are compatible with this being driven by active shortening. Further sampling and analysis are required to provide a higher-resolution, low-temperature thermochronology of Alpine exhumation, to isolate the drivers for Palaeogene Dziruli Massif cooling and uplift, and to constrain better the extent of the current, localized phase of rapid exhumation.

    Beschreibung: The Harappan Culture, one of the oldest known urban civilizations, thrived on the northwest edge of the Thar Desert (India and Pakistan) between 3200 and 1900 BCE. Its demise has been linked to rapid weakening of the summer monsoon at this time, yet reorganization of rivers may also have played a role. We sampled subsurface channel sand bodies predating ca. 4.0 ka and used U-Pb dating of zircon sand grains to constrain their provenance through comparison with the established character of modern river sands. Samples from close to archaeological sites to the north of the desert show little affinity with the Ghaggar-Hakra, the presumed source of the channels. Instead, we see at least two groups of sediments, showing similarities both to the Beas River in the west and to the Yamuna and Sutlej Rivers in the east. The channels were active until after 4.5 ka and were covered by dunes before 1.4 ka, although loss of the Yamuna from the Indus likely occurred as early as 49 ka and no later than 10 ka. Capture of the Yamuna to the east and the Sutlej to the north rerouted water away from the area of the Harappan centers, but this change significantly predated their final collapse.

    Beschreibung: We have developed a Bayesian method for Dix inversion and illustrated it with examples from the North Sea. The method is a constrained Dix inversion in which the uncertainty of the estimated interval velocities is an integral part of the solution. The method combines available geologic prior knowledge with the information in the picked rms velocities so that the prior model stabilizes and constrains the inversion. The definition of layers or intervals is flexible. One possibility is the classical layering whereby the top and bottom of the intervals are defined directly from the times of the picked rms velocities, but the intervals can be defined arbitrarily and independent of the picked velocities. Even a pseudocontinuous velocity profile with dense time sampling can be predicted with corresponding uncertainty. The Dix inversion is conventionally formulated as a 1D inversion problem in the locations with picked rms velocities. Under the assumption that Dix inversion is adequate, we have defined a method for spatial prediction at any position in a 3D model. This involves spatial smoothing and interpolation, and provides a method for building a velocity cube with the corresponding uncertainty on any grid. Explicit analytic expressions are found for both the optimal solution and the uncertainty. In general, the uncertainty of the estimated interval velocities increases with depth and with decreasing layer thickness. The uncertainty of the spatial prediction decreases with increasing spatial correlation. Because the solution is of an explicit analytic form, the Bayesian Dix inversion is computationally fast and does not require stochastic simulation.
