A diatom-phosphorus weighted averaging (WA) transfer function, derived from a training set of currently oligotrophic to mesotrophic European Alpine lakes, was applied to a high-resolution sediment core with annual laminae from Mondsee, an Austrian pre-alpine lake, in order to reconstruct the eurrophication history of the lake.2. The water chemistry records of total phosphorus (TP) available for Mondsee were compared with the diatom-inferred TP from the model for the period 1975–93. The trend in TP values as inferred by the model paralleled the monitored trend in TP values closely, with matching peaks in 1979/80, a decrease in values from the early 1980s, a second smaller peak in 1986/7, and a further reduction in concentrations in the last 6 years.3. However, there was a clear mismatch between the actual timing of the major TP peak, with the water chemistry records reporting its occurrence in 1979, and the diatom model indicating a small peak in 1980 and the highest concentrations in 1982. This can be attributed to the uncertainty of the sediment chronology for this section of the core, and possibly to the inconsistency between the core resolution and the resolution of the diatom model.4. In terms of the actual concentrations of TP inferred by the model, they compared reasonably well with the measured data, although the model tends to underestimate for the lower core section owing largely to poor diatom assemblage analogues. In the upper part of the core, the diatom-inferred TP values were in extremely close agreement with the monitored chemical data.5. This validation study indicates that diatom-phosphorus transfer functions are robust and are able reliably to infer past-TP concentrations from fossil diatom assemblages in sediment cores. Despite the natural intra- and interannual variability in diatom assemblages and epilimnetic water chemistry, the technique can provide accurate estimates of TP with an annual resolution. The model can be applied to selected sites with suitable sediment records to reconstruct lake TP histories, thus providing a pragmatic management tool for addressing lake eutrophication problems.

The range of morphological variability of theCyclotella bodanica/radiosa, C. distinguenda, C. ocellata, C. comensis andC. stelligera complexes was documented. Significance tests included in the canonical correspondence analysis (CCA) indicated that total phosphorus, conductivity and surface water temperature were significantly correlated withCyclotella taxa assemblage distribution. Numerical species optima and tolerances were calculated in respect to these significant variables using weighted averaging (WA) regression and calibration. CCA and WA were able to provide information about the environmental preferences of the various morphotypes observed.

    Notes: Abstract The surface sediments of 50 Central European low to high mountain, acid-sensitive lakes were examined for chrysophycean stomatocysts. One hundred and twenty sic siliceous stomatocyst morphotypes were differentiated by using scanning electron microscope (SEM), counted and illustrated (light microscopy and SEM). Canonical Correspondence Analysis (CCA) showed that pH is the most important measured variable in explaining the cyst distributions, with a strong correlation to the first axis (r2=-0.78). Simple Weighted Averaging (WA) regression and calibration was performed to generate a transfer function for lake water pH. Finally, the transfer function was evaluated by reconstructing pH for a short annually laminated and 137Cs dated core from Lake Plešné, a strongly acid lake, in the Bohemian Forest. A comparison of the measured and inferred pH values indicates that the model has good predictive power. The cyst-inferred pH and the diatom-inferred pH both indicate that pH was low during the 1970s and that the lake has experienced recovery in recent years.

    Notes: Abstract: Multidisciplinary palaeolimnological approaches were applied to long sediment cores from Längsee, Carinthia. Climate amelioration after deglaciation allowed the expansion of shrubs and timber-line trees and the onset of plankton and probably lake summer stratification. This phase called Pre-Bölling has an upper boundary of approx. 16 kyrs BP. The change from a steppe to a tundra-like vegetation culminated during the following less temperate, humid phase, with lower in-lake productivity (Oldest Dryas, Ia; 16-13.6 kyrs BP). At about 13.6 kyrs BP when reforestation took place, summer surface water temperature increased towards recent temperatures and hypolimnetic anoxia developed. Different anoxic levels in respect to climate change underline the dynamic type of meromixis. Levels varied, being highest during the Preboreal/Boreal, and lowered during the Atlantic towards recent levels. The contrast between low trophic status and sediments enriched in organic matter characterizes "pseudeutrophic lake stratification" before the onset of human impact. Changes in the pelagic cladoceran communities during this time seem to be related to changes in predation. Aside long-term climatic change during the Atlantic, short-term fluctuations of precipitation within climate oscillations are suggested as possible causes of erosion processes, changes in lake level and water temperature. These are indicated for the Oldest and Younger Dryas, the Boreal/Atlantic and the Subboreal/Subatlantic transitions. They most likely act as the trigger mechanism for temporary mixing and changes in redox conditions. Extensive forest clearance in the catchment during Bronze-Age settlement caused a change in sediment type and temporary nutrient enrichment. The perturbations are comparable with erosion processes during the late-glacial when climate oscillations were the only driving forces. Anthropogenic influences on the lake environment vary according to land-use type.

    Notes: Abstract Chemical and biological sedimentary records of a high alpine lake were used to reconstruct palaeoecological conditions and compared with two centuries of instrumental temperature measurements. Air temperature determined the lake water pH throughout the past 200 yr almost regardless of the level of atmospheric deposition. Our data suggest a strong climate forcing of the acid-base balance in sensitive high-altitude lakes. Their physico-chemical conditions and biota strongly depend on the duration of ice and snow cover which is significantly different between warm and cold periods. Beside changes in weathering rates, in-lake alkalinity generation and water-retention time, delayed freezing in autumn and earlier ice-out dates with a shorter duration of CO2 over-saturation could be crucial for the tight temperature-pH coupling.

    Description: Background: Mesenchymal stem cells (MSCs) are increasingly considered to be used as biological immunosuppressants in hematopoietic stem cell transplantation (HSCT). In the early reconstitution phase following HSCT, natural killer (NK) cells represent the major lymphocyte population in peripheral blood and display graft-vs-leukemia (GvL) effects. The functional interactions between NK cells and MSCs have the potential to influence the leukemia relapse rate after HSCT. Until date, MSC-NK cell interaction studies are largely focussed on bone marrow derived (BM)-MSCs. Umbilical cord derived (UC)-MSCs might be an alternative source of therapeutic MSCs. Thus, we studied the interaction of UC-MSCs with unstimulated allogeneic NK cells. Results: UC-MSCs could potently suppress NK cell cytotoxicity in overnight cultures via soluble factors. The main soluble immunosuppressant was identified as prostaglandin (PG)-E2. Maximal PGE2 release involved IL-1β priming of MSCs after close contact between the NK cells and UC-MSCs. Interestingly, blocking gamma-secretase activation alleviated the immunosuppression by controlling PGE2 production. IL-1 receptor activation and subsequent downstream signalling events were found to require gamma-secretase activity. Conclusion: Although the role of PGE2 in NK cell-MSC has been reported, the requirement of cell-cell contact for PGE2 induced immunosuppression remained unexplained. Our findings shed light on this puzzling observation and identify new players in the NK cell-MSC crosstalk.
