    Description: Permafrost-affected landscape soils are rich in organic matter and contain a high fraction of organic nitrogen, but much of this organic matter remains inaccessible due to nitrogen limitation. Microbial nitrification is a key process in the nitrogen cycle, controlling the availability of dissolved inorganic nitrogen (DIN) such as ammonium and nitrate. In this study, we investigate the microbial diversity of canonical nitrifiers and their potential nitrifying activity in the active layer of different Arctic cryosols in the Lena River Delta in North-East Siberia. These cryosols are located on Samoylov Island, which has two geomorphological landscapes with mineral soils in the modern floodplain and organic-rich soils in the low-centered polygonal tundra of the Holocene river terrace. Microcosm incubations show that the highest potential ammonia oxidation rates are found in low organic soils, and the rates depend on organic matter content and quality, vegetation cover, and water content. As shown by 16S rRNA amplicon sequencing, nitrifiers represented 0.6% to 6.2% of the total microbial community. More than 50% of the nitrifiers belonged to the genus Nitrosospira. Based on PCR amoA analysis, ammonia-oxidizing bacteria (AOB) were found in nearly all soil types, whereas ammonia-oxidizing archaea (AOA) were only detected in low-organic soils. In cultivation-based approaches, mainly Nitrosospira-like AOB were enriched and characterized as psychrotolerant, with temperature optima slightly above 20 °C. This study suggests a ubiquitous distribution of ammonia-oxidizing microorganisms (bacteria and archaea) in permafrost-affected landscapes of Siberia with cold-adapted AOB, especially of the genus Nitrosospira, as potentially crucial ammonia oxidizers in the cryosols.

    Description: Permafrost deposits have been a sink for atmospheric carbon for millennia. Thaw-erosional processes, however, can lead to rapid degradation of ice-rich permafrost and the release of substantial amounts of organic carbon (OC). The amount of the OC stored in these deposits and their potential to be microbially decomposed to the greenhouse gases carbon dioxide (CO2) and methane (CH4) depends on climatic and environmental conditions during deposition and the decomposition history before incorporation into the permafrost. Here, we examine potential greenhouse gas production as a result of degrading ice-rich permafrost deposits from three locations in the northeastern Siberian Laptev Sea region. The deposits span a period of about 55 kyr from the last glacial period and Holocene interglacial. Samples from all three locations were incubated under aerobic and anaerobic conditions for 134 days at 4 ∘C. Greenhouse gas production was generally higher in deposits from glacial periods, where 0.2 %–6.1 % of the initially available OC was decomposed to CO2. In contrast, only 0.1 %–4.0 % of initial OC was decomposed in permafrost deposits from the Holocene and the late glacial transition. Within the deposits from the Kargin interstadial period (Marine Isotope Stage 3), local depositional environments, especially soil moisture, also affected the preservation of OC. Sediments deposited under wet conditions contained more labile OC and thus produced more greenhouse gases than sediments deposited under drier conditions. To assess the greenhouse gas production potentials over longer periods, deposits from two locations were incubated for a total of 785 days. However, more than 50 % of total CO2 production over 785 days occurred within the first 134 days under aerobic conditions, while 80 % were produced over the same period under anaerobic conditions, which emphasizes the nonlinearity of the OC decomposition processes. Methanogenesis was generally observed in active layer samples but only sporadically in permafrost samples and was several orders of magnitude smaller than CO2 production.

    Description: The currently observed climate warming will lead to substantial permafrost degradation and mobilization of formerly freeze-locked matter. Based on recent findings, we assume that there are substantial stocks of inorganic nitrogen (N) within the perennially frozen ground of arctic ecosystems. We studied eleven soil profiles down to one meter depth below surface at three different sites in arctic eastern Siberia, covering polygonal tundra and river floodplains, to assess the amount of inorganic N stores in arctic permafrost-affected soils. Furthermore, we modeled the potential thickening of the seasonally unfrozen uppermost soil (active) layer for these sites, using the CryoGrid2 permafrost model and representation concentration pathway (RCP) 4.5 and 8.5 scenarios. The first scenario, RCP4.5, is a stabilization pathway that reaches plateau atmospheric carbon concentrations early in the 21st century; the second, RCP8.5, is a business as usual emission scenario with increasing carbon emissions. The modeled increases in active layer thickness (ALT) were used to estimate potential annual N mobilization from permafrost-affected soils in the course of climate-induced active-layer deepening. We observed significant stores of inorganic ammonium in the perennially frozen ground of all investigated soils, up to 40-fold higher than in the active layer. The modeled ALT increase until 2100 under the RCP8.5 scenario was between 19 ± 3 cm and 35 ± 6 cm, depending on the location. Under the RCP4.5 scenario, the ALT remained stable in all investigated soils. Our estimated mean annual N release under the RCP8.5 scenario is between 8 ± 3 mg m−2 and 81 ± 14 mg m−2 for the different locations, which reaches values up to the order of magnitude of annual fixation of atmospheric N in arctic soils. However, the thawing induced release of N represents only a small flux in comparison with the overall ecosystem N cycling.
