    Description: 〈p〉G protein–coupled receptors (GPCRs) convert extracellular stimuli to intracellular responses that regulate numerous physiological processes. Crystallographic and biophysical advances in GPCR structural analysis have aided investigations of structure-function relationships that clarify our understanding of these dynamic receptors, but the molecular mechanisms associated with activation and signaling for individual GPCRs may be more complex than was previously appreciated. Here, we investigated the proposed water-mediated, hydrogen-bonded activation switch between the conserved NPxxY motif on transmembrane helix 7 (TMH7) and a conserved tyrosine in TMH5, which contributes to α〈sub〉1B〈/sub〉-adrenoceptor (α〈sub〉1B〈/sub〉-AR) and β〈sub〉2〈/sub〉-AR activation. Disrupting this bond by mutagenesis stabilized the α〈sub〉1B〈/sub〉-AR and the β〈sub〉2〈/sub〉-AR in inactive-state conformations, which displayed decreased agonist potency for stimulating downstream IP〈sub〉1〈/sub〉 and cAMP signaling, respectively. Compared to that for wild-type receptors, agonist-mediated β-arrestin recruitment was substantially reduced or abolished for all α〈sub〉1B〈/sub〉-AR and β〈sub〉2〈/sub〉-AR inactive-state mutants. However, the inactive-state β〈sub〉2〈/sub〉-ARs exhibited decreased agonist affinity, whereas the inactive-state α〈sub〉1B〈/sub〉-ARs had enhanced agonist affinity. Conversely, antagonist affinity was unchanged for inactive-state conformations of both α〈sub〉1B〈/sub〉-AR and β〈sub〉2〈/sub〉-AR. Removing the influence of agonist affinity on agonist potency gave a measure of signaling efficacy, which was markedly decreased for the α〈sub〉1B〈/sub〉-AR mutants but little altered for the β〈sub〉2〈/sub〉-AR mutants. These findings highlight the pharmacological heterogeneity of inactive-state GPCR conformations, which may facilitate the rational design of drugs that target distinct conformational states of GPCRs.〈/p〉

    Description: Effect of LiNbO 3 ferroelectric substrate on the carrier mobility in top gated graphene field-effect transistors (G-FETs) is demonstrated. It is shown that, at the same residual concentration of the charge carriers, the mobility in the G-FETs on the LiNbO 3 substrate is higher than that on the SiO 2 /Si substrate. The effect is associated with reduction of Coulomb scattering via screening the charged impurity field by the field induced in the ferroelectric substrate, but significant only for mobilities below 1000 cm 2 /V s. Raman spectra analysis and correlations established between mobility and microwave loss tangent of the Al 2 O 3 gate dielectric indicate that the charged impurities are located predominantly at the gate dielectric and/or at the gate dielectric/graphene interface and are likely associated with oxygen vacancies.

    Description: In the vicinity of magnetic reconnection, magnetic energy is transferred into kinetic energy. A reconnection region hence corresponds to a load and it should manifest itself as large and positive values of the power density, E · J 〉〉 0, where E is the electric field and J the current density. In this article we analyze Cluster plasma sheet data from 2001–2004 to investigate the use of the power density for identifying possible magnetic reconnection events from large sets of observed data. From theoretical arguments we show that an event with  pW/m 3 in the Earth's magnetotail observed by the Cluster instruments ( X 〈− 10 R E and ) is likely to be associated with reconnection. The power density can be used as a primary indicator of potential reconnection regions, but selected events must be reviewed separately to confirm any possible reconnection signatures by looking for other signatures such as Hall electric and magnetic fields and reconnection jets. The power density can be computed from multi-spacecraft data, and we argue that the power density can be used as a tool for identifying possible reconnection events from large sets of data, e.g., from the Cluster and the MMS missions.

    Description: Clay rocks are being considered for radioactive waste disposal, but relatively little is known about the impact of microbes on the long-term safety of geological repositories. Thus, a more complete understanding of microbial community structure and function in these environments would provide further detail for the evaluation of the safety of geological disposal of radioactive waste in clay rocks. It would also provide a unique glimpse into a poorly studied deep subsurface microbial ecosystem. Previous studies concluded that microorganisms were present in pristine Opalinus Clay, but inactive. In this work, we describe the microbial community and assess the metabolic activities taking place within borehole water. Metagenomic sequencing and genome-binning of a porewater sample containing suspended clay particles revealed a remarkably simple heterotrophic microbial community, fueled by sedimentary organic carbon, mainly composed of two organisms: a Pseudomonas sp. fermenting bacterium growing on organic macromolecules and releasing organic acids and H 2 , and a sulfate-reducing Peptococcaceae able to oxidize organic molecules to CO 2 . In Opalinus Clay, this microbial system likely thrives where pore space allows it. In a repository, this may occur where the clay rock has been locally damaged by excavation or in engineered backfills.
