Pfleiderer Cubic chiral magnets crystallizing in space group P 2 1 3 give rise to a spin structure of nontrivial topology, the so-called skyrmion lattice. The strongly doped semiconductor Fe 1 − x Co x Si is a fascinating member of this material class of compounds as it exhibits helimagnetism over a wide compositional range. The authors study the magnetic properties of this itinerant magnet for different values of x , combine their results with previous reports, and infer the magnetic phase diagrams. The latter are rather isotropic but depend on the field and temperature history. In particular, when cooled in an applied magnetic field, the skyrmion lattice may persist down to lowest temperatures. This history dependence, in combination with the variation of temperature, field, and length scale of the magnetic order by means of compositional tuning, makes Fe 1 − x Co x Si an excellent material for future experiments studying skyrmions.     Description: To achieve guide RNA (gRNA) multiplexing and an efficient delivery of tens of distinct gRNAs into single cells, we developed a molecular assembly strategy termed chimeric array of gRNA oligonucleotides (CARGO). We coupled CARGO with dCas9 (catalytically dead Cas9) imaging to quantitatively measure the movement of enhancers and promoters that undergo differentiation-associated activity changes in live embryonic stem cells. Whereas all examined functional elements exhibited subdiffusive behavior, their relative mobility increased concurrently with transcriptional activation. Furthermore, acute perturbation of RNA polymerase II activity can reverse these activity-linked increases in loci mobility. Through quantitative CARGO-dCas9 imaging, we provide direct measurements of cis-regulatory element dynamics in living cells and distinct cellular and activity states and uncover an intrinsic connection between cis-regulatory element mobility and transcription.
