Notes: Abstract Temporal changes of the initial susceptibility of ferromagnetic crystals can be measured advantageously via the resonance frequency of an LC-oscillator. Due to its decreasing susceptibility during the magnetic desaccomodation, the crystal under investigation — which is taken as the core of the oscillatory circuit — induces definite changes of the resonance frequency. The essential advantage of this measuring method consists of the digital acquisition of the measured data, which may form the basis of a complete automation of the desaccomodation experiment. This work describes a device, in which all possibilities of automation — offered by this new measuring method — are realized in a consequent manner.

Notes: Summary The influence of streptomycin on grwoth, pigmentation and morphology of three mutants of the green alga Chlamydomonas reinhardii was investigated. 1. In the dark on solid media with sublethal doses of streptomycin both the streptomycin-sensitive strain and the streptomycin-resistant chromosomal mutant (isolated from the former strain) produce yellow colonies, whereas the uniparental resistant mutant grows green. With chloramphenicol under the same conditions no formation of yellow colonies is observed. 2. The streptomycin-induced loss of chlorophyll is accompanied by an almost complete loss of the thylakoid-structure. However, in the remaining chloroplast membrane starch grains and eye-spot are still present. 3. Yellow cells are able to produce under various conditions within 70–100 h green cultures containing normal chloroplasts. 4. In yellow cells the carotenoid/chlorophyll ratio is greatly enhanced. In the presence of streptomycin a general decrease of the pigment content is observed, while the relative amounts of the individual carotenoids and chlorophylls remain constant. 5. Streptomycin concentrations above the resistance level are found to be inhibitory for growth but not lethal. If, under the influence of light the streptomycin content of the medium decreases, green colonies are produced. The average lifetime of these colonies is shorter than that of uninhibited cultures. 6. The participation in thylakoid membrane formation of the different protein synthesizing systems of the cell is diseussed.

Notes: Summary A slide rule has been devised which is based on the general mathematical models of pharmacokinetics. It permits calculation of exact dosage regimens for individual patients from certain basic parameters. First, from the patient's renal clearance, the proportionality constant characterizing renal excretion of a certain drug (a) and its non-renal rate constant of elimination (k nr), the rate constant of total elimination (k e) can be calculated. Second, fromk e, the apparent volume of distribution (V d) and the desired final mean concentration of a drug (c∞), exact values can readily be obtained for the loading dose (D*) and the dosage schedule, which consists of the maintenance dose (D), the dosing intervals (τ) and the infusion rate for intravenous administration. In addition the slide rule provides information about the rate at which c∞ is reached ifD alone is administered at τ, and the fluctuation in the concentration around c∞ to be anticipated during τ. By use of this calculation, the slide rule facilitates the decision whether a loading dose should be given, and what dosage schedule is best suited to the therapeutic problem. It is possible, therefore, to calculate exact dosage regimens for individual patients, even for those with excretory dysfunction. The slide rule should also help physicians to comprehend the nature and significance of pharmacokinetic mechanisms.

Notes: Summary The muscle cells of the pharyngeal bulb of tardigrades Macrobiotus hufelandi and Milnesium tardigradum are up to 15 μm long and form a single layer between the basal lamina and the cuticle-coated lumen. The boundaries between adjacent cells are sinuous. The sarcolemma invaginates between the myofibrils whereby a marked E-system is formed. The sarcoplasmic reticulum is associated to the latter, constituting diads and triads. The myofibrils run radially. Thin filaments originate from hemidesmosome-like structures at the inner and outer sarcolemma in dense bundles which widen in the middle part of the fibrils. Each fibril contains thick and thin filaments in this region. As many as 11 thin filaments could be counted around a thick filament. The latter are not always arranged hexagonally. As it is confirmed by polarisation microscopy each myofibril has a wide A-Zone in the middle which is flanked by shorter I-zones. An H-zone is marked but indistinctly. Each myofibril is interpreted to have the functional and morphological equivalent to one sarcomere. The relevance of these findings in the evolution of Tardigrada is discussed.

Notes: Summary The somatic muscles of the tardigrades Macrobiotus hufelandi and Milnesium tardigradum consist of elongated cells which are structurally independent of each other and insert in the body wall. The contractile part of the muscle cells contains thick and thin myofilaments. The thin filaments run together in typical dense bodies. In relaxed muscles clear A-, I- and H-zones are formed, and the dense bodies are often accumulated in groups. In ribbon like I-zones they are sometimes arranged side by side in rows and resemble fragmented Z-rods. The sarcoplasmic reticulum is localized primarily in the small region between the contractile apparatus and sarcolemma, and is represented by a system of smooth, flat, reticulated cisternae. The contractile apparatus of the somatic muscles is compared to that of smooth and obliquely striated muscles of several invertebrates. On the basis of certain distinguishing features of the contractile part of somatic muscles of tardigrades, it is suggested that these muscles should be referred to as a type intermediate between smooth and obliquely striated muscles.
