    Notes: Usually, an Ar gas adding hydrogen gas is used for the fabrication of hydrogenated amorphous silicon (a-Si:H) films by reactive sputtering. The sputtered films contain Ar atoms of more than 5 at. %. These implanted Ar+ ions degrade the film quality, and H atoms, which terminate dangling bonds, leave the film easily by heating to about 300 °C. A He atom has a lighter atomic mass and smaller atomic radius than an Ar atom. Therefore, the reactive sputtered films using the He gas will be less affected by the implanted He+ ions than they will by the implanted Ar+ ions of those sputtered by Ar gas. Infrared absorption (IR) and electron spin resonance (ESR) measurements of the films sputtered by He gas are made and compared with those sputtered by Ar gas.

    Notes: Double- and multiple-layered Co-Cr thin films have been prepared to clarify the dominant factors in the c-axis orientation at the early stage of the film growth. The optimized process condition in the facing-targets sputtering system is likely to promote the c-axis self-orientation in the films since they can grow on the plasma-bombardment-free substrate. This effect was confirmed by depositing the films on the poorly oriented Co-Cr film as the underlayer. Meanwhile, when the well-oriented Co-Cr thin film was used as the underlayer, a continuous growth of crystallites resulted in the improvement of the c-axis orientation in the growing film even under process conditions unfavorable for depositing the well-oriented films. This Co-Cr underlayer effect continues until the thickness of the depositing film reaches about 2000 A(ring), suppressing the influence of the unfavorable sputtering condition.

    Description: The RNase P-mediated endonucleolytic cleavage plays a crucial role in the 3' end processing and cellular accumulation of MALAT1, a nuclear-retained long noncoding RNA that promotes malignancy. The regulation of this cleavage event is largely undetermined. Here we characterize a broadly expressed natural antisense transcript at the MALAT1 locus, designated as TALAM1, that positively regulates MALAT1 levels by promoting the 3' end cleavage and maturation of MALAT1 RNA. TALAM1 RNA preferentially localizes at the site of transcription, and also interacts with MALAT1 RNA. Depletion of TALAM1 leads to defects in the 3' end cleavage reaction and compromises cellular accumulation of MALAT1. Conversely, overexpression of TALAM1 facilitates the cleavage reaction in trans . Interestingly, TALAM1 is also positively regulated by MALAT1 at the level of both transcription and RNA stability. Together, our data demonstrate a novel feed-forward positive regulatory loop that is established to maintain the high cellular levels of MALAT1, and also unravel the existence of sense-antisense mediated regulatory mechanism for cellular lncRNAs that display RNase P-mediated 3' end processing.

    Description: 〈p〉Cells with high ploidy content are common in mammalian extraembryonic and adult tissues. Cell-to-cell fusion generates polyploid cells during mammalian development and tissue regeneration. However, whether increased ploidy can be occasionally tolerated in embryonic lineages still remains largely unknown. Here, we show that pluripotent, fusion-derived tetraploid cells, when injected in a recipient mouse blastocyst, can generate diploid cells upon ploidy reduction. The generated diploid cells form part of the adult tissues in mouse chimeras. Parental chromosomes in pluripotent tetraploid cells are segregated through tripolar mitosis both randomly and nonrandomly and without aneuploidy. Tetraploid-derived diploid cells show a differentiated phenotype. Overall, we discovered an unexpected process of controlled genome reduction in pluripotent tetraploid cells. This mechanism can ultimately generate diploid cells during mouse embryo development and should also be considered for cell fusion–mediated tissue regeneration approaches.〈/p〉

    Description: Seismic wave resonance in sedimentary basins is a well-recognized seismic hazard; however, concentrated areas of earthquake damage have been observed near basin edges, where wave propagation is particularly complex and difficult to understand with sparse observations. The Tokyo metropolitan area is densely populated, subject to strong shaking from a diversity of earthquake sources, and sits atop the deep Kanto sedimentary basin. It is also instrumented with two seismic arrays: the dense MEtropolitan Seismic Observation network (MeSO-net) within the basin, and the High sensitivity seismograph network (Hi-net) surrounding it. In this study, we explore the 3-D seismic wavefield within and throughout the Kanto basin, including near and across basin boundaries, using cross-correlations of all components of ambient seismic field between the stations of these two arrays. Dense observations allow us to observe clearly the propagation of three modes of both Rayleigh and Love waves. They also show how the wavefield behaves in the vicinity of sharp basin edges with reflected/converted waves and excitation of higher modes.

    Description: We studied the seismicity rate increase in the Kanto region around Tokyo following the 2011 Tohoku-Oki earthquake ( M w 9.0) to examine whether this increase was correlated with the static increases in the Coulomb failure function (CFF) of the Tohoku-Oki earthquake sequence. Because earthquakes in the Kanto region exhibit various focal mechanisms, the receiver faults for the CFF were assumed to be the focal mechanism solutions for nearly 19 000 earthquakes that previously occurred. Our results showed that the number of earthquakes for which the mechanism solutions had a positive CFF (~12 000) is much larger than those that had a negative CFF (~2000). Comparison of the CFF values for earthquakes before and after the Tohoku-Oki earthquake showed that the latter had more positive values; this supports the hypothesis that the coseismic stress change transferred from the Tohoku-Oki earthquake sequence is the major contributing factor to the increased seismicity rate in the Kanto region.

    Description: Since the initial discovery of hydrothermal vents in 1977, these ‘extreme’ chemosynthetic systems have been a focus of interdisciplinary research. The Okinawa Trough (OT), located in the semi-enclosed East China Sea between the Eurasian continent and the Ryukyu arc, hosts more than 20 known vent sites but all within a relatively narrow depth range (600–1880 m). Depth is a significant factor in determining fluid temperature and chemistry, as well as biological composition. However, due to the narrow depth range of known sites, the actual influence of depth here has been poorly resolved. Here, the Yokosuka site (2190 m), the first OT vent exceeding 2000 m depth is reported. A highly active hydrothermal vent site centred around four active vent chimneys reaching 364°C in temperature, it is the hottest in the OT. Notable Cl depletion (130 mM) and both high H 2 and CH 4 concentrations (approx. 10 mM) probably result from subcritical phase separation and thermal decomposition of sedimentary organic matter. Microbiota and fauna were generally similar to other sites in the OT, although with some different characteristics. In terms of microbiota, the H 2 -rich vent fluids in Neuschwanstein chimney resulted in the dominance of hydrogenotrophic chemolithoautotrophs such as Thioreductor and Desulfobacterium . For fauna, the dominance of the deep-sea mussel Bathymodiolus aduloides is surprising given other nearby vent sites are usually dominated by B. platifrons and/or B. japonicus , and a sponge field in the periphery dominated by Poecilosclerida is unusual for OT vents. Our insights from the Yokosuka site implies that although the distribution of animal species may be linked to depth, the constraint is perhaps not water pressure and resulting chemical properties of the vent fluid but instead physical properties of the surrounding seawater. The potential significance of these preliminary results and prospect for future research on this unique site are discussed.

    Description: To better understand the relationship between P-wave velocities and ice content in saturated, unconsolidated saline permafrost, we constructed an effective-medium model based upon ultrasonic P-wave data that were obtained from earlier laboratory studies. The model uses a two-end-member mixing approach in which an ice-filled, fully frozen end member and a water-filled, fully unfrozen end member are mixed together to form the effective medium of partially frozen sediments. This mixing approach has two key advantages: (1) It does not require parameter tuning of the mixing ratios, and (2) it inherently assumes mixed pore-scale distributions of ice that consist of frame-strengthening (i.e., cementing and/or load-bearing) ice and pore-filling ice. The model-predicted P-wave velocities agree well with our laboratory data, demonstrating the effectiveness of the model for quantitatively inferring ice content from P-wave velocities. The modeling workflow is simple and is largely free of calibration parameters — attributes that ease its application in interpreting field data sets.
