    Notes: A system comprised of an immobilized yeast reactor producing ethanol, with a membrane pervaporation module for continuously removing and concentrating the produced ethanol, was developed. The combined system consisted of two integrated circulation loops: In one the sugar-containing medium is circulated through the membrane pervaporation module. The two loops were interconnected in a way allowing for separate parameter optimization (e.g., flow rate, temperature, pH) for each loop.The fermentation unit was 2.0 L bioreactor with five equal segments, packed with 5-mm beads of immobilized yeasts. The bead matrix was a crosslinked polyacrylamide hydrazide gel coated with calcium alginate. The fast circulation loop of the bioreactor allowed for efficient liberation of CO2 at the top of the immobilized yeast reactor. Continuous operation of the uncoupled reactor for over 50 days with inflowing defined medium or dilute molasses at a residence time of 1.25 h yielded ethanol at a rate of about 10 g/L h.The pervaporation unit was constructed from four 60-cm-long tubular membranes of silicone composite on a polysulfone support. The output from the fermentor was circulated through the inside of the tubes of a unit with a total surface area of 800 cm2, having an average flux of 150 mL/h, and selectivities to ethanol vs. water up to 7. A vacuum of 30 mb was applied to the outside of the tubes, removing 20-30 g of ethanol per hour, which was collected in condensors. The continuous removal of ethanol, avoiding inhibition of the fermentation process, resulted in an improved productivity and allowed the use of high sugar concentrations (40% wt/vol) offering the potential of a compact system with reduced stillage.The combined system of ethanol production and removal enabled an operative steady state at which the liquid volume of the system, and the concentrations of ethanol within the reactor (˜4% wt/vol), as well as within the flux crossing the pervaporation membrane (17%-20% wt/vol) were kept constant. At the steady state, a 40% wt/vol sugar solution could be continuously added to the fermentor when 12%-20% wt/vol clear ethanol solution was continuously removed by the pervaporation unit. Membrane fouling was reversed by short washing steps, and continuous step operation was maintained by working with two different modules that were interchanged. In this manner, long term continuous operation (over 40 days) was achieved with a productivity of 20-30 g/L h, representing over a twofold increase relative to the continuously operated reactor uncoupled from the membrane and a fivefold increase in comparison with the value obtained fro a corresponding batch fermentation.

    Notes: A new simple method for the preparation of chemically crosslinked chitosan beads is presented. It consists of the dropwise addition of 2-3% (w/v) low molecular weight chitosan solution containing 2% (w/v) glyoxal in 1% (w/v) tetrasodiumdiphosphate, pH 8.0. Immobilized viable baker's yeast (Saccharomyces cerevisiae) could be obtained via gel entrapment within the new beads when means preventing their direct contact with soluble chitosan were provided, “disguising” the cells until gelation and crosslinking were completed. Such means included cell suspension in castor oil or mixing with carboxymethyl-cellulose powder. Application of these means was shown to be necessary, as cells exposed to soluble chitosan immediately lost their viability and glycolytic activity. Yeast disguised in castor oil was also protected from bead reinforcement by glutaraldehyde treatment, significantly strengthening bead stability while operating under acidic conditions. This capability was demonstrated by continuous ethanol production by chitosan entrapped yeast. © 1994 John Wiley & Sons, Inc.

    Notes: The separation of DNA by gel electrophoresis provides a rapid method for determining size distributions of DNA in solution. Densitometric scanning of photographs of gels has been the standard method of analysis of agarose gels. However, analysis of photographs is complicated by the non-linear response of photographic film. Charged-coupled device video cameras have become popular for quantitative densitometry and we have used a charge-coupled device camera to image agarose gels to quantitate DNA damage. We compare video and photographic densitometry for quantitation of ultraviolet radiation (UV)-induced DNA damage and find that the two methods give equivalent results.

    Description: This paper is a summary of some research activities in calibrating DC-8 SAR data conducted in 1989/90 by the author and others. Firstly, by examining the process of 'symmetrization' in the production of DC-8 Stokes' matrix data, it is shown that an exact solution to the problem of calibrating this data exists, given certain assumptions. Secondly a new, more general, system model for radar polarimeters is proposed, together with a simple approach for verifying the model's validity. The importance of the orientation angle about the radar line of sight for some types of calibration devices is discussed and some results of an experiment to cross-calibrate the C-Band VV data from the DC-8 and DLR E-SAR sensors are presented.

    Description: Active radar calibrators are used to derive both the amplitude and phase characteristics of a multichannel polarimetric SAR from the complex image data. Results are presented from an experiment carried out using the NASA/JPL DC-8 aircraft SAR over a calibration site at Goldstone, California. As part of the experiment, polarimetric active radar calibrators (PARCs) with adjustable polarization signatures were deployed. Experimental results demonstrate that the PARCs can be used to calibrate polarimetric SAR images successfully. Restrictions on the application of the PARC calibration procedure are discussed.

    Description: Imaging radar polarimeters are usually implemented using a Synthetic Aperture Radar (SAR) approach to give a high resolution image in two dimensions: range and azimuth. For each pixel in the image a polarimetric SAR gives sufficient information to characterize the polarimetric scattering properties of the imaged area (or target) as seen by the radar. Using a polarimetric SAR system as opposed to a single-polarization SAR system provides significantly more information about the target scattering mechanisms and allows better discrimination between different types of surfaces. In these notes a brief overview of SAR polarimetry is offered. The notes are intended as a text to accompany a lecture on SAR polarimetry as part of an AGARD-NATO course. Covered in the notes are the following: the polarization properties of electromagnetic waves; the concepts of radar scattering and measuring radar backscatter with a SAR; polarization synthesis; scattering matrix, Stokes matrix, and covariance matrix representations of polarimetric SAR data; polarization signature plots; design and calibration of polarimetric SAR systems; polarization filtering for target detection; fitting a simple model to polarimetric SAR measurements of naturally occurring features; and supervised classification of polarimetric SAR data.

    Description: SIR-C/X-SAR is currently scheduled for launch in April 1994. SIR-C is an L-Band and C-Band, multi-polarization spaceborne SAR system developed by NASA/JPL. X- SAR is an X-Band SAR system developed by DARA/ASI. One of the problems involved in calibrating the SIR-C instrument is to make sure that the horizontal (H) and vertical (V) polarized beams are aligned in the azimuth direction, i.e.. that they are pointing in the same direction. This is important if the polarimetric performance specifications for the system are to be met. To solve this problem, we have designed and built a prototype of a low-cost ground receiver capable of recording received power from two antennas, one H-polarized, the other V-polarized. The two signals are mixed to audio then recorded on the left and right stereo channels of a standard audio cassette player. The audio cassette recording can then be played back directly into a Macintosh computer, where it is digitized. Analysis of.

    Description: Polarimetric SAR data can provide a great deal of information about the scattering behavior of the surface under observation. Polarimetric SAR systems often measure the scattering matrices of the areas under observation in linear polarizations (H and V). From the scattering matrix commonly used forms such as the covariance matrix and the Stokes matrix can be easily derived. Other measures derived from polarimetric SAR data include the standard deviation of texture, correlation coefficients between scattering matrix terms, and the mode and variance of phase differences between scattering matrix terms. The effects of additive system noise on these measurements is not often considered in the literature on this subject.

    Description: The Alaska SAR Facility has been receiving and processing SAR data from the J-ERS-1 satellite since Spring 1992. Corner reflectors have been set up for J-ERS-1 SAR calibration at a site near Delta Junction, in central Alaska. Image quality and calibration analysis results from the Delta Junction site and others will be presented in this paper. The impact of the 3-bit Analog-to-Digital Converter and the automatic stepping of the gain as a function of range in the J-ERS-1 radar receiver on calibration performance has been assessed. Preliminary observations on J-ERS-1 SAR data are that the average Signal-to-Noise ratio is generally fairly low, in the range 5-6dB. Azimuth ambiguity levels are higher than preflight analysis indicated. Over land, the dynamic range in the backscatter at L-band for approximately 36 degree incidence angle is often fairly high.
