    Notes: Abstract Antibodies raised against purified rapeseed 19 kDa oleosin protein were used to screen an embryo-derived λgt11 expression library from Brassica napus. A near full-length cDNA clone, BnV, was isolated. The 781 bp cDNA contained an open reading frame of 549 bp followed by an untranslated region of 222 pb and a poly(A) region of 10 bp. Comparisons between this cDNA and a different oleosin cDNA previously isolated from the same library showed high degrees of sequence similarity in the central domain region and in the 3′ untranslated region. Sequence similarities between the derived protein sequence of this cDNA and all other known oleosin protein sequences are discussed.

    Notes: Abstract The sequence of an oleosin gene from Brassica napus has been determined. This gene contains a single intron of 437 bp and encodes a polypeptide of 195 amino acids. The oleosin gene product has an estimated molecular mass of 21.5 kDa and consists of a highly hydrophobic central domain flanked by relatively polar N- and C-terminal domains. The central domain is highly conserved between all oleosins sequenced to date and contains a run of periodically spaced leucine residues similar to that of a leucine-zipper motif. The gene has been shown to be expressed specifically in the embryo, maximally between 9 and 11 weeks after flowering, i.e. during the seed desiccation stage. Two transcriptional start sites have been mapped to -70 and -21 of the ATG and a putative ABA-responsive element and three repeated motifs have been identified in the promoter. These short promoter sequences could correspond to regulatory elements responsible for embryo-specific gene expression. Up to six genes exist in the oleosin gene family.

    Notes: Abstract In Brassica napus, oleosins are expressed at high levels in the seed during the latter stages of embryo development. The cis-acting regulatory properties of an 872 bp promoter fragment of a B. napus oleosin gene were examined by analysis of β-glucuronidase (GUS) expression in transgenic tobacco plants containing an oleosin promoter-GUS transcriptional fusion. The reporter gene was expressed at high levels only in seeds, specifically in embryo and endosperm tissue and regulated throughout seed development. These data demonstrate that oleosin gene transcription is regulated in a tissue-specific and temporally regulated manner and clearly indicate that oleosin protein expression is co-ordinated primarily at the transcriptional level. Oleosin mRNA was shown to be abscisic acid (ABA) inducible and an ABA-response element in the oleosin promoter was shown to be bound by a protein factor in a sequence-specific manner. Sequence analysis of the oleosin promoter has identified several other putative cis-acting sequences which may direct oleosin gene expression. The presence of a large open reading frame in the bottom strand of the oleosin promoter (ORF2) which encodes a polypeptide similar to the ethylene-induced E4 gene of tomato is reported. A PCR-generated DNA probe containing the ORF2 sequence hybridised with a 1.4 kb transcript in total RNA extracts of a variety of tissues, including leaves and germinated seed cotyledons. This finding suggests that the oleosin gene promoter directs transcription in both directions. It is the first report of a bi-directional nuclear gene promoter in plants.

    Notes: Abstract This report describes a novel nonaqueous polymeric formulation that exhibits low-viscosity fluid behavior for ease of spraying with conventional nebulizer, which when sprayed into the nasal cavity, transforms to a high-viscosity gel for efficient retention and drug absorption. The transformation occurs because of the rheological changes induced by a change in the solvent composition of the polymeric formulation in the moist nasal cavity. Such a rheological change would then facilitate enhanced residence time of the drug at the site of administration in order to avoid drainage losses. This study reports the results of the effects of a variety of factors such as solvent composition and polymer concentration on the rheological properties of a polyacrylic acid polymer. An attempt to correlate viscosity enhancement effects with enhanced and sustained-release behavior of propranolol, a drug that undergoes extensive first-pass effects, from such formulations via nasal administration in beagle dogs is also described.

    Notes: Abstract The objective of the present study is to apply response surface methodology to the design and analysis of composite experiments containing independent covariate(s). The approach is illustrated here by the study of viscosity characteristics of a polymeric mucoadhesive formulation in multicomponent solvent vehicles. The nonaqueous formulation will produce a gel network with significant rheological change when in contact with body fluids. The process of water inclusion will induce not only solvent compositional change of the mixture but also concomitant dilution of the polymer concentration. To study the viscosity change over the solvent compositions and polymeric concentrations of interest, an experimental design is utilized consisting of a 10-point simplex-centroid lattice augmented with three interior points at each polymeric concentration. The contour patterns are compared with the experimental, data using the variance and lack of fit, starting with the Scheffe linear model and building up to the full cubic model including the covariate terms. The fitted model provides information needed to predict optimum formulations, i.e., initial viscosity of less than 100 cP, but yielding rheological profiles commensurate with high degrees of substantivity when diluted with water. For illustrative purposes, the Carbopol resins neutralized with a 1:1 molar equivalent ratio of triethanolamine in three primary solvents, propylene glycol, glycerol formal, and water, were chosen for this study.

    Notes: Abstract The objective of this study is to investigate the viscoelastic properties of Carbopol 934P polymeric systems in a variety of mixtures of pharmaceutical solvents. Carbopol 934P neutralized with a 1:1 equivalent ratio of triethanolamine was dissolved in various binary or ternary solvent mixtures consisting of propylene glycol, glycerol formal, and water. Dynamic moduli G′ and G″, complex viscosities, η′ and η″, and loss tangent, tanδ, were examined over a frequency range of 10-3 to 10 Hz using an oscillatory viscoelastic rheometer at 30°C. The results indicated that for 0.5-1.5 wt% neutralized Carbopol in ternary mixtures, G′ and G″ increased by 3-4 orders of magnitude and the phase angle decreased from 80 to 25° when the water content in the solvent mixture increased from 10 to 80 wt%. These studies also indicated that the addition of water to nonaqueous Carbopol 934P polymer systems transforms them from low-viscosity solutions to gels with significant elastic behavior involving physical interaction and entanglement of polymer segments with solvents.

    Description: A supplementary investigation has been conducted in the langley 20-foot free-spinning tunnel on a l/24-scale model of the Grumman F11F-1 airplane to determine the spin and recovery characteristics with alternate nose configurations, the production version and the elongated APS-67 version, with and without empty and full wing tanks. When spins were obtained with either alternate nose configuration, they were oscillatory and recovery characteristics were considered unsatisfactory on the basis of the fact that very slow recoveries were indicated to be possible. The simultaneous extension of canards near the nose of the model with rudder reversal was effective in rapidly terminating the spin. The addition of empty wing tanks had little effect on the developed spin and recovery characteristics. The model did not spin erect with full wing tanks. For optimum recovery from inverted spins, the rudder should be reversed to 22O against the spin and simultaneously the flaperons should be moved with the developed spin; the stick should be held at or moved to full forward longitudinally. The minimum size parachute required to insure satisfactory recoveries in an emergency was found to be 12 feet in diameter (laid out flat) with a drag coefficient of 0.64 (based on the laid-out-flat diameter) and a towline length of 32 feet.

    Description: An investigation has been conducted in the Langley 20-foot free-spinning tunnel on a l/19-scale model of the North American T-28C airplane to determine the spin and recovery characteristics. The T-28C airplane is similar to the T-28B airplane except for slight modifications for the arresting hook. The lower rear section of the fuselage was cut out and, consequently, the lower part of the rudder was removed to make a smooth fairing with the fuselage. The T-28B airplane had good recovery characteristics; but these modifications, along with the addition of gun packages on the wings, led to poor and unsatisfactory spin-recovery characteristics during demonstration spins of the T-28C airplane. Model test results indicated that without the gun packages installed, satisfactory recoveries could be obtained if the elevators were held full back while the rudder was fully reversed and the ailerons were held neutral. However, with the addition of gun packages to the wings and the corresponding change in loading, recoveries were considered unsatisfactory. Recoveries attempted by using a larger chord or larger span rudder were improved very slightly, but were still considered marginal or unsatisfactory. Strakes placed on the nose of the model were effective in slowing the spin rotation slightly and, in most instances, decreased the turns for recovery slightly. Recovery characteristics were slightly marginal for the full fuel loading when strakes and the extended-chord rudder were installed; but with the wing fuel partly used, recovery characteristics were again considered unsatisfactory or, at least, definitely on the marginal side. The optimum control technique for recovery is movement of the rudder to full against the spin with the stick held full back (elevators full up) and the ailerons held neutral, followed by forward movement of the stick only after the spin rotation ceases. Inverted-spin test results indicate that the airplane will spin steep and fast and that recovery by full rudder reversal will be satisfactory if the ailerons are held neutral.

    Description: An investigation has been conducted in the Langley 20-foot free-spinning tunnel on a l/18-scale model of the Ryan X-13 airplane to determine the size parachute required for emergency-spin recovery during demonstration spins. Test results for erect spins indicate that a stable 12.73-foot-diameter parachute with a drag coefficient of 1.11 (based on the 12.73-foot projected diameter) is the minimum size required to insure satisfactory recovery in an emergency. The use of a stable parachute is recommended. Test results for inverted spins indicate that the l2.75-foot-diameter parachute is also satisfactory to terminate inverted spins.

    Description: An investigation has been completed in the Langley 20-foot free-spinning tunnel on a l/18-scale model of the Ryan X-13 airplane to determine its spin, recovery, and tumbling characteristics, and to determine the minimum altitude from which a belly landing could be made in case of power failure in hovering flight. Model spin tests were conducted with and without simulated engine rotation. Tests without simulated engine rotation indicated two types of spins: one, a slightly oscillatory flat spin; and the other, a violently oscillatory spin. Tests with simulated engine rotation indicated that spins to the left were fast rotating and steep and those to the right were slow rotating and flat. The optimum technique for recovery is reversal of the rudder to against the spin and simultaneous movement of the ailerons to full with the spin followed by movement of the elevators to neutral after the spin rotation ceases. Tumbling tests made on the model indicated that although the Ryan X-13 airplane will not tumble in the ordinary sense (end-over-end pitching motion), it may instead tend to enter a wild gyrating'motion. Tests made to simulate power failure in hovering flight by dropping the model indicated that the model entered what appeared to be a right spin. An attempt should be made to stop this motion immediately by moving the rudder to oppose the rotation (left pedal), moving the ailerons to with the spin (stick right), and moving the stick forward after the spin rotation ceases to obtain flying speed for pullout. The minimum altitude required for a belly landing in case of power failure in hovering flight was indicated to be about 4,200 feet.
