    Notes: . Transmission of malaria parasites from vertebrate blood to the mosquito vector depends critically on the differentiation of the gametocytes into gametes. This occurs in response to environmental stimuli encountered by the parasite in the mosquito bloodmeal. Male gametogenesis involves three rounds of DNA replication and endomitosis, and the assembly de novo of 8 motile axonemes. Azadirachtin, a plant limnoid and insecticide with an unkown mode of action, specifically inhibits the release of motile gametes from activated microgametocytes but does not inhibit growth and replication of asexual blood stages. We have combined confocal laser scanning microscopy and transmission electron microscopy to examine the effect of azadirachtin on the complex reorganisation of the microtubule cytoskeleton during gametogenesis in Plasmodium berghei. Neither the replication of the genome nor the ability of tubulin monomers to assemble into microtubules upon gametocyte activation were prevented by azadirachtin. However, the drug interfered with the formation of mitotic spindles and with the assembly of microtubules into typical axonemes. Our observations suggest that azadarachtin specifically disrupts the patterning of microtubules into more complex structures, such as mitotic spindles and axonemes.

    Notes: . Encephalitozoon microsporidia proliferate and differentiate within a parasitophorous vacuole. Using the fluorescent probe, calcein, and the mitochondrial probe, MitoTracker-CMXRos, a vital method was developed that confirmed ultrastructural reports that the host cell mitochondria frequently lie in immediate proximity to the parasitophorous vacuole. Morphometry failed to demonstrate any infection-induced increase in host cell mitochondria as there was no correlation between the mitochondrial volume and the extent of infection as judged by the parasitophorous vacuole volume. The total ATP concentration of infected cells did not differ from that of uninfected cells in spite of the increased metabolic demands of the infection. Treatment with 10-6 M albendazole, more than ten times the antiparasitic IC50 dose, and demecolcine had no subjective effect on the proximity of mitochondria to the parasitophorous vacuole membrane when studied by either transmission electron microscopy or by confocal microscopy even though these drug concentrations affected microtubule structure. Thus, once the association between mitochondria and the parasitophorous vacuole has been established, host cell microtubule integrity is probably not required for its maintenance. It is unlikely that the antimicrosporidial action of albendazole involves physically uncoupling developing parasite stages from host cell organelle metabolic support.

    Notes: . Microsporidia of the genus Encephalitozoon infect mammalian cells and have become a source of morbidity and mortality in immunocompromised humans. Encephalitozoon microsporidia develop and mature within parasitophorous vacuoles, enlarging the vacuole over time until it eventually occupies most of the cytoplasm of the host cell. The ability of the host cell to accommodate such a large burden for several days suggests that the parasite subverts normal host cell processes to ensure optimal environmental conditions for its growth and development. Since this environment would be threatened if cell division of the host cell occurred, we have formulated the hypothesis that infection with Encephalitozoon microsporidia induces an arrest in the cell cycle of the host cell. In support of this hypothesis, we have found that mitotic index and DNA duplication are reduced in infected cells as compared to uninfected cells. The number of host cell nuclei in S phase is increased. The levels of cyclin D1 and the percentage of cells in G1 are reduced; however, the levels of cyclin B1 are elevated even though the percentage of cells in G2/M is decreased. These results suggest that host cells infected with Encephalitozoon microsporidia are blocked at multiple points in the cell cycle.

    Notes: : Increasingly, residential development in urbanizing areas is accomplished by large housing projects, composed of clusters of townhouses or garden partments. It is hypothesized that the runoff from such developments should carry more pollution than that from the same number of housing units on separate plots, because the runoff is conveyed directly to drainage channels rather than being drained across lawns and gardens, which may absorb part of the pollutants. In order to evaluate this effect, storm event data were obtained from a planned unit development near Hightstown, N. J., using samples taken every 10 minutes throughout the storm at two different storm sewers. Results show heavy metals pollution about what had been anticipated, in accordance with the hypothesis given above, and BOD ammonia and phosphates higher than predicted. The results are significant for areawide water quality planning in metropolitan areas, where projections of future pollution loadings depends upon the land use.

    Notes: : For many years, a commonly used strategy for source water protection in Taiwan has been setting up arbitrary, fixed-width buffer zones near sensitive waters, such as water-supply reservoirs, and prohibiting any development in their watersheds. However, such regulations are now often viewed as infringing by the government on landowners' property rights, a situation that has led to citizen protests. This paper describes a proposed strategy that is water-quality based and uses a quantitative zoning approach. A reservoir's watershed is divided into several zones beginning from the normal water line to the divide. Different levels of best management practices (BMPs) are required for controlling runoff pollution in different zones. The layout of the management zones is based on a number of factors such as reservoir classification, water quality conditions, and physical characteristics of the watershed. The goal of promoting such an approach is to try to balance the needs of watershed development and water quality protection. A case study using the Tapu Reservoir Watershed in Northern Taiwan as an example for illustrating the proposed zoning approach is presented.

    Notes: Modern hydro-electric power schemes operate without storage, abstracting a proportion of the flow in the river whenever the level is above a specified minimum. The electricity is passed to the national grid and the water returns to the river without delay or contamination.The ecological aspects of this form of abstraction mean that they are generally acceptable. Fisheries is one of several topics which requires close attention, and another is the consequences and effects brought about by changes to the flow regime. The purpose of this paper is to explore the interaction between those flows which are reduced (only some are affected) and the associated fishery issues.The effects of typical abstraction regimes on down-river hydrographs, and in particular on the magnitude and timing of flows, are explained in the context of the particularly variable conditions which occur in upland rivers, i.e. the preferred location for many so-called run-of-river hydroelectric schemes. The limited evidence which is available for migratory fish movement through such reaches is used to assess how these movements could be affected by typical abstraction regimes.

    Notes: : Effects of glycerol, xylitol, and sorbitol on selected physical properties of whey protein isolate (WPI) films were examined. Increasing glycerol or sorbitol content led to increases in moisture content, water vapor permeability, and % elongation; and decreases in tensile strength, elastic modulus, and glass transition temperatures of the films. Increasing levels of xylitol had no effect on permeability, moisture content, or glass transition temperature of the films, but decreased % elongation, tensile strength, and elastic modulus. Moisture content of the films correlated well with glass transition temperatures. Differences in measured physical properties of films with plasticizer type and concentration may be attributed to differences in the hygroscopic and crystalline properties of the plasticizers.

    Notes: Storage resistance of a powdered synthetic orange-flavored drink (OFD) to nonenzymic browning as measured by formation of three browning products at 38°C was compared with that for natural instant orange juice (IOJ). OFD was freeze dried to afford a homogenous OFD mixture comparable to IOJ and its storage stability was compared with that of IOJ. Although OFD was more resistant than IOJ to nonenzymic browning when OFD had a lower moisture level than IOJ, at the same moisture level, OFD was no more stable. This indicated that the greater storage stability of OFD is due to its lower moisture content, rather than to its discrete separation of components which might retard sugaracid reactions.

    Notes: Witch flounder from an inshore stock were caught by gill-net and their storage life in ice was determined by sensory analysis. Chemical analyses were made of ATP, hypoxanthine, lactic acid, and trimethylamine. ATP breakdown occurred at a slower rate than in most fish, and after 18 days storage, the major components of the nucleotide degradation were still intermediates as production of hypoxanthine was low. Glycogen levels at death were low, and this metabolite degraded in the manner expected from past experience. The relationship between pH and lactate was highly significant (P ≤ 0.01). Trimethylamine values depended on whether KOH or K2CO3 was used during the analysis, and discrepencies between these two methods are discussed. Analysis of sensory and chemical results indicated that trimethylamine was a good indicator of quality (P ≤ 0.01), and that while hypoxanthine correlated well with quality, its quantity was such that it was unlikely to have any effect on sensory evaluation.
