    Description: Non-intrusive optical measurement techniques have been examined in the context of developing robust instruments which can routinely yield data of engineering utility in high speed turbomachinery test rigs. The engineering requirements of such a measurement are presented. Of particular interest were approaches that provide both velocity and state-variable information in order to be able to completely characterize transonic flowfields. Consideration of all of the requirements lead to the selection of particle image velocimetry (PIV) for the approach to velocity measurement while laser induced fluorescence of oxygen (O2 LIF) appeared to offer the most promise for gas temperature measurement. A PIV system was developed and demonstrated on a transonic turbine stage in the MIT blowdown turbine facility. A comprehensive data set has been taken at one flow condition. Extensive calibration established the absolute accuracy of the velocity measurements to be 3-5 %. The O2 LIF proved less successful. Although accurate for low speed flows, vibrational freezing of O2 prevented useful measurements in the transonic, 300-600 K operating range of interest here.

    Description: Extended spaceflight under microgravity conditions leads to significant atrophy of weight-bearing muscles. Atrophy and hypertrophy are the extreme outcomes of the high degree of plasticity exhibited by skeletal muscle. Stimuli which control muscle plasticity include neuronal, hormonal, nutritional, and mechanical inputs. The mechanical stimulus for muscle is directly related to the work or exercise against a load performed. Little or no work is performed by weight-bearing muscles under microgravity conditions. A major hypothesis is that focal adhesion kinase (FAK) which is associated with integrin at the adherens junctions and costa meres of all skeletal muscles is an integral part of the major mechanism for molecular signaling upon mechanical stimulation in all muscle fibers. Additionally, we propose that myotonic protein kinase (DMPK) and dystrophin (DYSTR) also participate in distinct mechanically stimulated molecular signaling pathways that are most critical in type I and type II muscle fibers, respectively. To test these hypotheses, we will use the paradigms of hindlimb unloading and overloading in mice as models for microgravity conditions and a potential exercise countermeasure, respectively, in mice. We expect that FAK loss-of-function will impair hypertrophy and enhance atrophy in all skeletal muscle fibers whereas DYSTR and DMPK loss-of-function will have similar but more selective effects on Type IT and Type I fibers, respectively. Gene expression will be monitored by muscle-specific creatine kinase M promoter-reporter construct activity and specific MRNA and protein accumulation in the soleus (type I primarily) and plantaris (type 11 primarily) muscles. With these paradigms and assays, the following Specific Project Aims will be tested in genetically altered mice: 1) identify the roles of DYSTR and its pathway; 2) evaluate the roles of the DMPK and its pathway; 3) characterize the roles of FAK and its pathway and 4) genetically analyze the mechanisms and interactions between the FAK, DYSTR, and DMPK-associated pathways in single and specific combinations of mutants. The identification of potential signaling mechanisms may permit future development of pharmacological countermeasures for amelioration and prevention of the microgravity-induced atrophy in extended spaceflight, and the analysis of both overloading and unloading paradigms may provide further support for development of exercise-based countermeasures. Understanding the basic mechanisms of molecular signaling in muscle plasticity may aid our understanding and treatment of skeletal muscle atrophy not only in spaceflight but in similar problems of the aging population, in prolonged bed rest, and in cachexia associated with chronic disease.

    Description: The remarkable order of striated muscle is the result of a complex series of protein interactions at different levels of organization. Within muscle, the thick filament and its major protein myosin are classical examples of functioning protein machines. Our understanding of the structure and assembly of thick filaments and their organization into the regular arrays of the A-band has recently been enhanced by the application of biochemical, genetic, and structural approaches. Detailed studies of the thick filament backbone have shown that the myosins are organized into a tubular structure. Additional protein machines and specific myosin rod sequences have been identified that play significant roles in thick filament structure, assembly, and organization. These include intrinsic filament components, cross-linking molecules of the M-band and constituents of the membrane-cytoskeleton system. Muscle organization is directed by the multistep actions of protein machines that take advantage of well-established self-assembly relationships. Copyright 1999 John Wiley & Sons, Inc.

    Description: We report on measurements of the angular stability, commonly called 'jitter', of the STS Orbiter during normal operations in space. Measurements were carried out by measuring optically the Orbiter's roll and pitch orientation relative to the solar vector as the orbiter was held in a -Z(sub 0) solar inertial orientation (orbiter bay oriented toward the Sun). We also report observations of an interesting perturbation to the orbiter's orientation noted by the crew during the STS-60 mission. These data may be useful in analyzing the in-orbit response of the Orbiter to thruster firings and other applied torques, and may aid in the planning of future experiments that require fine-pointed operations by the orbiter.

    Description: The low frequency continuum (LFC) noise between 30 and 200 kHz has been investigated from the ISEE 3 spacecraft in the solar wind by means of a radio astronomy experiment more sensitive than previously available. It is demonstrated that the LFC radiation observed in the solar wind is in the form of longitudinal plasma waves rather than transverse electromagnetic waves. The observed spectral characteristics are found to be a function of antenna length. In addition, both the absence of antenna spin modulation and the fact that these plasma waves do not propagate to large distances imply a local origin for the LFC.

Its optical system provides nearly stigmatic imaging of selected portions of the sun over the spectral range from 22.5 to 44.0 nm.     Description: Observations simultaneously made at visual (B, V, and R) wavelengths, at 1, 1.4, and 3.4 mm and at 1.3, 2, 6, and 20 cm of 9 QSOs and BL Lac objects are reported. The range of the millimeter-visual spectral index alpha sub mv was only 0.65-0.82, typical of optically thin synchrotron emission. This may indicate that the electrons radiating synchrotron emission in this portion of the spectrum are not subjected to large radiation losses, and therefore relativistic bulk motion with Doppler factors of about 10 are required. The visual spectral index is much more broadly distributed and typically larger than alpha sub mv. The spectral energy distributions have not changed much in the last two to five years except for 2251+15 and perhaps 0235+164. Only 1749+09 shows a sharp spectral break shortward of 1 mm. Sixteen other sources were observed at 1 mm, of which seven were detected.

    Description: Hardware for the International Solar Polar Mission (ISPM) Unified Radio and Plasma (URAP) wave experiment is presented. The URAP determines direction and polarization of distant radio sources for remote sensing of the heliosphere, and studies local wave phenomena which determine the transport coefficients of the ambient plasma. Electric and magnetic field antennas and preamplifiers; the electromagnetic compatibility plan and grounding; radio astronomy and plasma frequency receivers; a fast Fourier transformation data processing unit waveform analyzer; dc voltage measurements; a fast envelope sampler for the solar wind, and plasmas near Jupiter; a sounder; and a power converter are described.

    Description: Deuterium-enriched hydrogen is present in organic matter in such meteorites as noncarbonaceous chondrites. The majority of the unequilibrated primitive meteorites contain hydrogen whose D/H ratios are greater than 0.0003, requiring enrichment (relative to cosmic hydrogen) by isotope exchange reactions taking place below 150 K. The D/H values presented are the lower limits for the organic compounds derived from interstellar molecules, since all processes subsequent to their formation, including terrestrial contamination, decrease their D/H ratios. In contrast, the D/H ratios of hydrogen associated with hydrated silicates are relatively uniform for the meteorites analyzed. The C-13/C-12 ratios of organic matter, irrespective of D/H ratio, lie well within those observed for the earth. Present findings suggest that other interstellar material, in addition to organic matter, is preserved and is present in high D/H ratio meteorites.
