    Description: Highlights • Regional brain iron concentrations are heterogenous. • Regional distribution of iron is most consistent with ferritin mRNA expression. • SEC-ICP-MS reveals the protein masses that cytosolic iron is associated with. • More than 50 % of cytosolic iron is associated with ferritin. Iron is essential for brain development and health where its redox properties are used for a number of neurological processes. However, iron is also a major driver of oxidative stress if not properly controlled. Brain iron distribution is highly compartmentalised and regulated by a number of proteins and small biomolecules. Here, we examine heterogeneity in regional iron levels in 10 anatomical structures from seven post-mortem human brains with no apparent neuropathology. Putamen contained the highest levels, and most case-to-case variability, of iron compared with the other regions examined. Partitioning of iron between cytosolic and membrane-bound iron was generally consistent in each region, with a slightly higher proportion (55 %) in the ‘insoluble’ phase. We expand on this using the Allen Human Brain Atlas to examine patterns between iron levels and transcriptomic expression of iron regulatory proteins and using quantitative size exclusion chromatography-inductively coupled plasma-mass spectrometry to assess regional differences in the molecular masses to which cytosolic iron predominantly binds. Approximately 60 % was associated with ferritin, equating to approximately 25 % of total tissue iron essentially in storage. This study is the first of its kind in human brain tissue, providing a valuable resource and new insight for iron biologists and neuroscientists, alike.

    Description: Understanding the sources of potentially toxic elements (PTEs) in soils is a worldwide challenge that requires effective discrimination between geogenic and anthropogenic contributions, particularly in areas with certain geological complexity. This study aims to examine the chemical contents of 23 topsoil samples collected from the surroundings of a fossil fuel power plant in the village of Puerto Libertad (Sonoran Desert, Mexico). The study did not exclusively focus on the source identification of the priority PTEs to evaluate soil pollution. Furthermore, major oxides and immobile trace element (Zr, Hf, and REE: La→Lu) data were provided for a reliable assessment of the provenance of the soils. The relatively high SiO2 contents (65.26–75.42 wt%, anhydrous basis), the Post-Archean Australian Shale (PAAS)-normalized REE patterns, and the uniformity of the values of the Index of Compositional Variability (ICV = 1.11–2.72) and the Chemical Index of Alteration (CIA = 31.65–51.79) suggest that the soils were derived from intermediate to felsic source rocks, controlled by the local weathering of the parent bedrocks, under a low degree of chemical weathering conditions. The PTE data were treated following a robust workflow, which included the use of the enrichment factor (EF), the Spearman rank correlation (ρ), and multivariate statistical analyses allowed the generation of significant elemental associations and the identification of pools related either to the geological background or to anthropogenic activities. The results suggesting that Mo and Zn concentrations present a moderate anthropogenic influence while the concentrations of Pb, Sn, Cu, Cd, As, Cr, and Co are predominantly of geogenic origin. Vanadium (avg. EFV = 3.4) and Ni (avg. EFNi = 4.6) were the most enriched elements in the soils. Moreover, the highest values of the integrated Nemerow Pollution Index (PIN〉3) were recorded at the sampling stations closer to the village, suggesting point-source pollution by the emissions of the power station. Finally, in this paper is traced the extent of the particulate released into the atmosphere, which can be dispersed in a wide area into the Sonoran Desert.

    Description: The delivery of organic carbon (OC) to the ocean's deepest trenches in the hadal zone is poorly understood, but may be important for the carbon cycle, contain crucial information on sediment provenance and event-related transport processes, and provide age constraints on stratigraphic sequences in this terminal sink. In this study, we systematically characterize bulk organic matter (OM) and OC signatures (TOC/TN, C, 14C), as well as those from application of serial thermal oxidation (ramped pyrolysis/oxidation) of sediment cores recovered along an entire hadal trench encompassing high stratigraphic resolution records spanning nearly 2000 years of deposition. We analyze two cores from the southern and northern Japan Trench, where submarine canyon systems link shelf with trench. We compare results with previously published data from the central Japan Trench, where canyon systems are absent. Our analyses enable refined dating of the stratigraphic record and indicate that event deposits arise from remobilization of relatively surficial sediment coupled with deeper erosion along turbidity current pathways in the southern and central study site and from canyon flushing events in the northern study site. Furthermore, our findings indicate deposition of predominantly marine OC within hemipelagic background sediment as well as associated with event deposits along the entire trench axis. This implies that canyon systems flanking the Japan Trench do not serve as a short-circuit for injection of terrestrial OC to the hadal zone, and that tropical cyclones are not major agents for sediment and carbon transfer into this trench system. These findings further support previous Japan Trench studies interpreting that event deposits originate from the landward trench slope and are earthquake-triggered. The very low terrestrial OC input into the Japan Trench can be explained by the significant distance between trench and hinterland (〉180 km), and the physiography of the canyons that do not connect to coast and river systems. We suggest that detailed analyzes of long sedimentary records are essential to understand OC transfer, deposition and burial in hadal trenches.

    Description: The International Marine Organization 2020 Global Sulfur Cap requires ships to burn fuels with 〈0.50% S and some countries require 〈0.10% S in certain Sulfur Emission Control Areas but little is known about these new types of fuels. Using both traditional GC–MS and more advanced chromatographic and mass spectrometry techniques, plus stable isotopic, δ13C and δ2H, analyses of pristane, phytane and n-alkanes, the organic components of a suite of three 0.50% S and three 0.10% S compliant fuels were characterized. Two oils were found to be near identical but all of the remaining oils could be forensically distinguished by comparison of their molecular biomarkers and by the profiles of the heterocyclic parent and alkylated homologues. Oils could also be differentiated by their δ13C and δ2H of n-alkanes and isoprenoids. This study provides important forensic data that may prove invaluable in the event of future oil spills.

    Description: Adipose tissue is essential to endotherms for thermoregulation and energy storage as well as functioning as an endocrine organ. Adipose derived hormones, or adipokines, regulate metabolism, energy expenditure, reproduction, and immune function in model systems but are less well studied in wildlife. Female northern elephant seals (NES) achieve high adiposity during foraging and then undergo natural fasts up to five weeks long during haul-outs associated with reproduction and molting, resulting in large changes in adipose reserves. We measured circulating levels of four adipokines: leptin, resistin, adiponectin, and kisspeptin-54, in 196 serum samples from female NES at the beginning and end of their breeding and molting fasts. We examined the relationships between these adipokines and life-history stage, adiposity, mass, cortisol, and an immune cytokine involved in the innate immune response interleukin 6 (IL-6). All four adipokines varied with life-history stage. Leptin concentrations were highest at the beginning of the breeding haul-out. Resistin concentrations were higher throughout the breeding haul-out compared to the molt haul-out. Adiponectin concentrations were highest at the beginning of both haul-outs. Kisspeptin-54 concentrations were highest at the end of the breeding haul-out. Leptin, resistin, and adiponectin were associated with measures of body condition, either adiposity, mass, or both. Resistin, adiponectin, and kisspeptin-54 were associated with circulating cortisol concentrations. Resistin was strongly associated with circulating IL-6, a multifunctional cytokine. Adiponectin was associated with glucose concentrations, suggesting a potential role in tissue-specific insulin sensitivity during life-history stages categorized by high adiposity. Increased cortisol concentrations late in lactation were associated with increased kisspeptin-54, suggesting a link to ovulation initiation in NES. This study suggests dramatic changes in circulating adipokines with life-history and body condition that may exert important regulatory roles in NES. The positive relationship between adiponectin and adiposity as well as the lack of a relationship between leptin and kisspeptin-54 differed from model systems. These differences from biomedical model systems suggest the potential for modifications of expression and function of adipose-derived hormones in species that undergo natural changes in adiposity as part of their life-history.

    Description: Continuous measurements from the OSNAP (Overturning in the Subpolar North Atlantic Program) array yield the first estimates of trans-basin heat and salinity transports in the subpolar latitudes. For the period from August 2014 to May 2018, there is a poleward heat transport of 0.50 ± 0.05 PW and a poleward salinity transport of 12.5 ± 1.0 Sv across the OSNAP section. Based on the mass and salt budget analyses, we estimate that a surface freshwater input of 0.36 ± 0.05 Sv over the broad subpolar-Arctic region is needed to balance the ocean salinity change created by the OSNAP transports. The overturning circulation is largely responsible for setting these heat and salinity transports (and the derived surface freshwater input) derived from the OSNAP array, while the gyre (isopycnal) circulation contributes to a lesser, but still significant, extent. Despite its relatively weak overturning and heat transport, the Labrador Sea is a strong contributor to salinity and freshwater changes in the subpolar region. Combined with trans-basin transport estimates at other locations, we provide new estimates for the time-mean surface heat and freshwater divergences over a wide domain of the Arctic-North Atlantic region to the north and south of the OSNAP line. Furthermore, we estimate the total heat and freshwater exchanges across the surface area of the extratropical North Atlantic between the OSNAP and the RAPID-MOCHA (RAPID Meridional Overturning Circulation and Heat-flux Array) arrays, by combining the cross-sectional transports with vertically-integrated ocean heat and salinity content. Comparisons with the air-sea heat and freshwater fluxes from atmospheric reanalysis products show an overall consistency, yet with notable differences in the magnitudes during the observation time period.

    Description: The Slope Sea in the Northwest Atlantic Ocean, located between the Gulf Stream and the continental shelf of the Northeast United States, is a recently-documented possible major spawning ground for Atlantic bluefin tuna (Thunnus thynnus). Larval surveys and a habitat modeling study have shown that suitable spawning habitat occurs in the Slope Sea, but the degree to which this habitat varies interannually is an open question. Here, we perform a decade-long (2009–2018) numerical modeling analysis, with simulated larvae released uniformly throughout the Slope Sea, to investigate the interannual variability in the water temperature and circulation criteria deemed necessary for successful spawning. We also quantify the influence of Gulf Stream meanders and overshoot events on larval retention and their effect on habitat suitability rates throughout the Slope Sea, defined as the percentage of simulated larvae released at a given location that satisfy criteria related to water temperature and retention near nursery habitat. Average environmental oceanographic conditions over the decade are most favorable in the western part of the Slope Sea, specifically in the Slope Gyre and away from the immediate vicinity of the Gulf Stream. Variability in domain- and summertime-averaged yearly spawning habitat suitability rates is up to 25% of the mean decadal-averaged values. Yearly habitat suitability correlates strongly with the Gulf Stream overshoot but does not correlate well with other oceanographic variables or indices, so an overshoot index can be used as a sole oceanographic proxy for predicting yearly bluefin spawning habitat suitability in the Slope Sea. Selective spawning can weaken the correlation between habitat suitability and Gulf Stream overshoot. Effort should be put towards collecting observational data against which we could validate our findings.

    Description: Very Low Sulfur Fuel Oils (VSLFO, 〈0.5% S) are a new class of marine fuel oils, introduced to meet recent International Maritime Organization regulations. The MV Wakashio was reported to have released 1000 t of VLSFO when it grounded on a reef in Mauritius on 25th July 2020. A field sample of oily residue contaminating the Mauritian coast was collected on 16th August 2020 and compared with the Wakashio fuel oil. Both oils were analyzed for organic and elemental content, and stable isotope ratios δ13C and δ2H measured. Comprehensive two-dimensional gas chromatography with high-resolution mass spectrometry was used to identify and compare biomarkers resistant to weathering. The aromatic content in the VLSFO was relatively low suggesting that the potential for ecosystem harm arising from exposure to toxic components may be less than with traditional fuel oil spills. The Wakashio oil spill is, to our knowledge, the first documented spill involving VLSFO.

    Description: Extreme events have increased in frequency globally, with a simultaneous surge in scientific interest about their ecological responses, particularly in sensitive freshwater, coastal, and marine ecosystems. We synthesized observational studies of extreme events in these aquatic ecosystems, finding that many studies do not use consistent definitions of extreme events. Furthermore, many studies do not capture ecological responses across the full spatial scale of the events. In contrast, sampling often extends across longer temporal scales than the event itself, highlighting the usefulness of long-term monitoring. Many ecological studies of extreme events measure biological responses but exclude chemical and physical responses, underscoring the need for integrative and multidisciplinary approaches. To advance extreme event research, we suggest prioritizing pre- and postevent data collection, including leveraging long-term monitoring; making intersite and cross-scale comparisons; adopting novel empirical and statistical approaches; and developing funding streams to support flexible and responsive data collection.

    Description: A substantial body of research now exists demonstrating sensitivities of marine organisms to ocean acidification (OA) in laboratory settings. However, corresponding in situ observations of marine species or ecosystem changes that can be unequivocally attributed to anthropogenic OA are limited. Challenges remain in detecting and attributing OA effects in nature, in part because multiple environmental changes are co-occurring with OA, all of which have the potential to influence marine ecosystem responses. Furthermore, the change in ocean pH since the industrial revolution is small relative to the natural variability within many systems, making it difficult to detect, and in some cases, has yet to cross physiological thresholds. The small number of studies that clearly document OA impacts in nature cannot be interpreted as a lack of larger-scale attributable impacts at the present time or in the future but highlights the need for innovative research approaches and analyses. We summarize the general findings in four relatively well-studied marine groups (seagrasses, pteropods, oysters, and coral reefs) and integrate overarching themes to highlight the challenges involved in detecting and attributing the effects of OA in natural environments. We then discuss four potential strategies to better evaluate and attribute OA impacts on species and ecosystems. First, we highlight the need for work quantifying the anthropogenic input of CO2 in coastal and open-ocean waters to understand how this increase in CO2 interacts with other physical and chemical factors to drive organismal conditions. Second, understanding OA-induced changes in population-level demography, potentially increased sensitivities in certain life stages, and how these effects scale to ecosystem-level processes (e.g. community metabolism) will improve our ability to attribute impacts to OA among co-varying parameters. Third, there is a great need to understand the potential modulation of OA impacts through the interplay of ecology and evolution (eco–evo dynamics). Lastly, further research efforts designed to detect, quantify, and project the effects of OA on marine organisms and ecosystems utilizing a comparative approach with long-term data sets will also provide critical information for informing the management of marine ecosystems.
