    Notes: A new rheometer is described. It consists of a transducer unit supplied with an electric impedance analyzing unit. The transducer unit converts a mechanical impedance into an electrical impedance by the piezoelectric effect. A detailed quantitative analysis of the interaction between the sample and the transducer is given. The real and imaginary parts of the shear modulus of a viscoelastic sample can be found in the frequency range of 1 mHz–50 kHz, modulus range of 0.1 MPa–10 GPa, and the temperature range of 150–300 K. The sample volume is 0.3 cm3 and the strain amplitude is exceedingly small. The small size of the transducer allows for good temperature control and equilibration. It has a simple construction based on inexpensive components. Results on the supercooled liquid 2-metyl-2,4-pentandiol at the glass transition obtained by the method are included. © 1995 American Institute of Physics.

    Notes: The precise determination of total organic carbon (TOC) in aquifer materials is important in the estimation of retardation coefficients for organic compound transport. Commonly used methods for TOC determination after carbonate removal by acidification include: weight loss on ignition of solids at 550° C, acid-dichromate oxidation, and combustion at 800–950° C. Since carbonate carbon frequently is more abundant than organic carbon all TOC methods depend on efficient and complete carbonate removal prior to the TOC quantification step. A method for the determination of TOC in aquifer solids involving carbonate removal by 0.73 M sulfurous acid and subsequent combustion at 800° C was tested on solids from three aquifers and on mineral standards. The carbon quantification by combustion and infrared detection of CO2 was accurate at solid organic carbon contents between 30 and 10,000 μgC/g. The acid treatment added small amounts of carbon to the sample, and siderite removal was incomplete. For samples without siderite or dolomite, the accuracy of TOC determinations was enhanced by grinding the solids. Measurement of very low TOC contents (50–100 /μgC/g) requires supplementary testing of grinding and acid treatment effects.

    Notes: Changes in the properties of soil solution in the rhizosphere of developing radish plants were investigated. Variations in these properties were expected to affect the distribution and speciation of metals in the soil and soil solution. Applications of essential nutrients were linked to plant transpiration rates and prevented excess addition of nutrient ions, so that subtle changes in soil solution composition would not be obscured. Soil solution pH, the concentration of dissolved organic carbon (DOC) and the concentrations of major and trace elements in solution were found to vary over time. Strict control of fertilizer additions led to the maintenance of a relatively low ionic strength in the soil solution, and under such conditions trace metal solubility appeared to be highy influenced by the concentration of DOC. A chemical speciation analysis was performed which showed that, while dissolved Cd and Zn were largely uncomplexed in unplanted soil, Cd and Zn in the rhizosphere existed mainly as complexed forms. It is hypothesized that this is partly a result of Ca-metal-ligand equilibrium in solution, with higher Ca concentrations in unplanted soil leading to more of the Cd and Zn in solution existing in the uncomplexed state. Changes in the concentrations of uncomplexed Cd and Zn with time gave the best correlations with changes in plant uptake of these metals over time, supporting the hypothesis that plants mainly absorb the free metal ion from soil solution.

    Notes: Abstract The chemical and temporal features of the sex-pheromone emitted by Heliothis virescens females are encoded by a diverse array of output pathways from the male-specific macroglomerular complex (MGC) in the antennal lobe. Most output neurons (29 out of 32) were activated by antennal stimulation with the principal component of the sex-pheromone blend of this species, (Z)-11-hexadecenal. Six neurons were excited solely by this component, 8 neurons also responded to the second essential blend component, (Z)-9-tetradecenal, and 14 neurons displayed equivalent responses to the two. Many neurons also effectively encoded the onset and duration of the stimulus. In one additional neuron, a prolonged excitatory response (synergism) was evoked only by the blend of the two components, indicating that some MGC neurons function as ‘blend detectors’. In contrast to the situation in Helicoverpa zea, none of the MGC neurons in H. virescens responded selectively to (Z)-9-tetradecenal, suggesting that these two noctuid species employ different neural strategies to encode information about their respective pheromone blends. Three MGC-output neurons responded selectively to (Z)-11-hexadecenyl acetate, an odorant released by some sympatric species that disrupts normal upwind flight to pheromones. Thus, changes in the attractant and deterrent chemical signals, as well as the physical features of these odor plumes, are encoded in the MGC across a diverse parallel array of output pathways to the protocerebrum.

    Notes: Abstract In situ microcosms were successfully used to study the degradation of a range of organic compounds in two pristine aquifers, one aerobic (Vejen) and one anaerobic (Villa Farm). Degradation and sorption behavior in the laboratory column microcosms packed with Villa Farm sediment was very similar to that in the in situ microcosms. However, when the columns were packed with quartz and equilibrated with aerated Villa Farm groundwater, behavior mirrored that at Vejen, indicating that oxygen rather than sediment or groundwater composition was the critical parameter. The aromatic and polyaromatic compounds (benzene, toluene,o-xylene, naphthalene) degraded under aerobic conditions only. The organochlorine compounds (trichloroethylene, tetrachloroethylene, 1,1,1-trichloroethane, 1,4-dichlorobenzene and 1,2-dichlorobenzene) showed little or no sign of degradation either aerobically or anaerobically. Interpretation of the data was complicated by strong sorption to the Villa Farm sediment but tetrachloromethane, nitrobenzene, ando-nitrophenol appeared to degrade under anaerobic conditions only. Phenol degraded rapidly under both sets of conditions.

    Notes: SUMMARY Calcitonin (CT) and other bone-active peptides have been restrained in clinical use by the need for parenteral administration. Although nasal and other transmucosal routes can be used for CT treatment, bioavailability and bioactivity of the peptide thus delivered are limited. We have evaluated the intrapulmonary route (IP) for the delivery of salmon calcitonin (SCT) in normal subjects. SCT was administered with a dry powder delivery inhaler. For comparison, each subject also received intramuscular (IM) SCT. Inhaled SCT produced significant hypocalcemia in all subjects as did injected SCT, and the peptide could be readily measured in serum by immunoassay. Compared by dose, IP SCT had 66% of the bioactivity and 28% of the bioavailability of IM SCT. This intrapulmonary route of administration should enhance the clinical acceptability of SCT and could also be applicable to other bone-active peptides.

    Notes: Abstract We have cloned a regulatory gene for amylase synthesis in Aspergillus oryzae. This gene, amyR, encodes a 604-amino acid transcriptional activator with a Cys6 zinc cluster, that shows extensive homology to the DNA binding domain of GAL4 from Saccharomyces cerevisiae. The DNA binding domain of amyR binds to two types of sequences found in a number of promoters from Aspergillus genes coding for starch-degrading enzymes. One type of binding site is characterized by two CGG triplets separated by eight nucleotides. The other type has only one CGG triplet, which is followed by the sequence AAATTTAA.

    Notes: The clean single-crystal VC0.75(100) surface, its oxidation, and the stability of the resulting oxide were examined with electron spectroscopy (AES, XPS and UPS), electron diffraction (low-energy and reflection high-energy electron diffraction) and atomic force microscopy (AFM). The clean VC0.75 surface exhibits carbon vacancy-related features in valence band spectroscopy data. This surface was exposed to O2 at 500 °C for up to 10 000 Langmuirs. The oxidation leads to the formation of vanadium oxide, VOx(100), crystallites on the VC0.75(100) substrate. The AFM images indicate that oxide crystallites grow with an elongated shape along 〈001〉 type directions. When heated above 1000 °C, the oxide is unstable and a clean VC0.75(100) surface is produced.
