    Notes: Abstract The existing methods to solve the problems of pulsatile flow in the cardiovascular system are based on either linear axisymmetric equations or non-linear one-dimensional equations. The solutions thus obtained give only a mediocre comparison with measurements. In this paper, a non-linear axisymmetric theory is proposed. The starting point of the present theory is a third degree polynomial representation of the velocity profile. Integral methods are then applied to obtain the governing equations. To ascertain the accuracy of the theory proposed above, the calculations for a simple case involving pulsatile flow in a long rigid tube were performed. The results are: (a) the average velocities compare very well with exact solutions and (b) the velocity profiles for a given frequency agree very well with exact solutions for flow in small tubes, but tend to differ as tube size is increased.

    Notes: Abstract Initial observations of low fertility, reduced sexual activity of males, and a high frequency of abnormalities in sperm differentiation of the wild type strain (Sevelen, Zürich) of Drosophila melanogaster, normally used in this laboratory, have lead to a study of this phenomenon and its causes. The abnormalities occur during all spermiogenetic stages and are not unique but are found with much lower frequency in normally fertile flies (Oregon R). Growth of Sevelen flies at high but sublethal temperatures (30°C) results in complete sterility, highly abnormal sperm differentiation, and a failure to recover fertility after return to normal temperatures (25° C) in the time period in which normal flies recover. — The principal factor, or factors, controlling normal sperm differentiation are located on the Y chromosome, but are thus far not precisely localized. Expression of the phenotype is modified by genetic background in which the Y chromosome is found.

    Notes: Summary The free amino acid contents in the anthers of male fertile, cytoplasmic male sterile (cms) and genic male sterile (gms) petunia lines were compared at different developmental stages of the male gametophyte. Quantitative differences in the amounts of free amino acids were found between the fertile and male sterile lines and between the cms and gms lines. The differences between the sterile lines were correlated with the different developmental stages at which the breakdown in microsporogenesis occurred. In the Rosy Morn (RM) cms line, where breakdown of microsporogenesis occurred at the end of prophase 1, there was an associated increase in asparagine and decrease in the other amino acids. In the RM gms line, in which breakdown occurred at the tetrad stage, an accumulation of asparagine in the anthers corresponded with an accumulation of glutamine beginning at prophase 1. Compared with fertile anthers, the sterile anthers accumulated much proline at the early meiotic stages, but no γ-aminobutyric acid. Comparison of the free amino acids of the fertile and the male sterile lines indicates that certain biochemical events leading to breakdown of microsporogenesis precede the observed cytological breakdown. The results from adding asparagine and glutamine to extracts of anthers at different developmental stages suggest that the amino acid balance may contribute to the changes in pH in the fertile and male sterile anthers which we observed previously.

    Notes: Abstract We have analyzed Ca2+ currents in two neuroblastoma-motor neuron hybrid cell lines that expressed normal or glutamine-expanded human androgen receptors (polyGln-expanded AR) either transiently or stably. The cell lines express a unique, low-threshold, transient type of Ca2+ current that is not affected by L-type Ca2+ channel blocker (PN 200-110), N-type Ca2+ channel blocker (ω-conotoxin GVIA) or P-type Ca2+ channel blocker (Agatoxin IVA) but is blocked by either Cd2+ or Ni2+. This pharmacological profile most closely resembles that of T-type Ca2+ channels [1-3]. Exposure to androgen had no effect on control cell lines or cells transfected with normal AR but significantly changed the steady-state activation in cells transfected with expanded AR. The observed negative shift in steady-state activation results in a large increase in the T-type Ca2+ channel window current. We suggest that Ca2+ overload due to abnormal voltage-dependence of transient Ca2+ channel activation may contribute to motor neuron toxicity in spinobulbar muscular atrophy (SBMA). This hypothesis is supported by the additional finding that, at concentrations that selectively block T-type Ca2+ channel currents, Ni2+ significantly reduced cell death in cell lines transfected with polyGln-expanded AR.

    Notes: Summary In the locules of fertile Petunia hybrida anthers the in vivo pH during meiosis is 6.8–7.0 and no callase activity can be detected. Towards the end of the tetrad stage, the pH drops to 5.9–6.2 followed by a burst of callase activity. Subsequently, callose in the tetrad walls is digested and the quartets of microspores are released into the anther locules and develop into pollen grains. In the anther locules of one cytoplasmic male sterile (cms) Petunia type the pH drop and strong callase activity are already evident at early meiotic stages. Consequently, the callose already accumulated in the pollen mother cell (PMC) walls is digested and the PMC's cease to develop and are degraded. In another sterile genotype, the pH of the locule remains high (6.8–7.0), no callase activity is detected at the end of tetrad stage and the callose walls remain intact until a very late stage. It is suggested that the timing of callase activity is critical for the normal development of the male gametophyte and that faulty timing may result in male sterility. Measurements of pH in vivo and assays for callase activity in vitro indicate that the low pH is a precondition for the enzyme activity. Furthermore, it is suggested that the activation of callase in vivo is in some way connected with the changes in the pH of the locule.

    Notes: Summary The distribution of vegetative and fruit characters among a large population of parent and F1 hybrid tomato cultivars is described and heritability estimates are given. Comparison of parent and offspring subpopulations revealed higher overall values in the F1 hybrids for vegetative characters and lower overall values for fruit characters than in the parents. A general Galtonean feature of higher offspring values for cross combinations with low parent means and lower offspring values for cross combinations with high parent mean is evident in all traits studied. The predictive value of the data given is evaluated.

A new mouse mutant, punk rocker (allele symbol Kcne1 pkr ), arose spontaneously on a C57BL/10J inbred strain background and is characterized by a distinctive head-tossing, circling, and ataxic phenotype. It is also profoundly and bilaterally deaf. The mutation resides in the Kcne1 gene on Chromosome (Chr) 16 and has been identified as a single base change within the coding region of the third exon. The C to T nucleotide substitution causes an arginine to be altered to a termination codon at amino acid position 67, and predictably this will result in a significantly truncated protein product. The Kcne1 pkr mutant represents the first spontaneous mouse model for the human disorder, Jervell and Lange-Nielsen syndrome, associated with mutations in the homologous KCNE1 gene on human Chr 21.
