    Notizen: Abstract Fructose 2,6-bisphosphate (Fru-2,6-P2) represents the most powerful activator of 6-phosphofructo-l-kinase, rate-limiting enzyme of glycolysis. Fru-2,6-P2 content is tightly regulated and appears to be under the control of different hormones and growth factors, acting either through covalent modification of isoenzymatic forms of 6-phosphofructo-2-kinase/fructose 2,6-bisphosphatase (PFK-2/FBPase-2), the bifunctional enzyme responsible for the synthesis and the degradation of the compound, or through changes in transcription rate and/or in the expression of different isoforms of the enzyme. In the present study the metabolism of Fru-2,6-P2 was investigated during the differentiation toward megakaryocytes induced by phorbol 12-myristate 13-acetate (PMA) treatment of human leukemia K562 and MEG-O1 cell lines. Fru-2,6-P2 content as well as PFK-2 activity were increased in a dose-dependent manner after 4 days of incubation with PMA. MEG-01 cells resulted more sensitive to the effect of the inducer, anyway in both cell types cytostatic concentrations of the phorbol ester were able to affect Fru-2,6-P2 metabolism. The effect of PMA was maximal at 4 days of incubation in both examined cell lines. Interestingly, the effect induced by the phorbol ester at 4 days was still appreciable subculturing K562 and MEG-01 cells for 3 days in the absence of the inducer and was associated with relevant changes in the molecular properties of PFK-2: namely increased Vmax and Km. This latter finding suggests that the rise in Fru-2,6-P2 content during the differentiation process toward megakaryocytes might result from the expression of a novel PFK-2 isoform.

    Beschreibung: Bacterial abundance, bacterial secondary production (BSP) and potential ectoproteolytic activity (PEA) were measured at 6 stations along the Strait of Magellan, South America, toward the end of summer 1995. Because of hydrological and climatic factors, 3 main areas could be identified in which the bacterial component displayed specific characteristics. In the Pacific Ocean side, subjected to freshwater inputs from rainfalls and melting of glaciers, the bacterial activities showed the highest values (BSP: 228.2 ng C/l h; PEA: 12.2 nmol/l h). The bacterial biomass was greater than the phytoplanktonic biomass, probably due to organic inputs from land stimulating the bacterial growth. The central part of the Strait demonstrated the lowest values (BSP: 32.6 ng C/l h, PEA: 4.6 nmol/l h), although the ratio of bacterial biomass to phytoplanktonic biomass was greater than 1. In the third area, the Atlantic Ocean opening, subjected to strong tidal currents, BSP and PEA displayed high values, 80 to 88.7 ng C/l h and 11.7 nmol/l h respectively. Nevertheless, the ratio of bacterial to phytoplanktonic biomass was less than 1, like in eutrophic areas. On the other hand, no impact of the tide was noted on bacterial parameters. Considering all samples measured in the 0 to 50 m layer, although BSP and PEA were positively correlated with bacterial abundance, the PEA to BSP ratio was negatively correlated with the bacterial biomass (r = -0.72, p 〈 0.001, n = 22). This ratio could be an indicator of trophic conditions in the 3 subsystems of the Strait.

    Beschreibung: The Global Navigation Satellite System (GNSS) represents a primary data source in Solid Earth Sciences. In order to investigate the Earth’s crustal deformation, time series of the estimated daily positions of the stations are routinely analyzed at the Istituto Nazionale di Geofisica e Vulcanologia (INGV) to investigate the deformation of the Earth’s surface caused by tectonic and non-tectonic processes. The GNSS observations of the stations are processed using the three main scientific software: GAMIT/GLOBK, BERNESE, and GIPSY OASIS II. The accuracy and the strength of geodetic solutions often depend on the geometry and spatial density of the network, and the availability and quality of GNSS data. In many circumstances, GNSS networks are deployed for topographic purposes by private or public institutions, and a significant number of GNSS stations in large regions acquire continuous observations. It may happen that such networks do not collect and distribute data according to IGS standards, so it could be difficult to analyze this data using automated data-processing tools. For that reason, this data is often ignored or partially used by the scientific community, despite their potential usefulness in geodynamic studies. We have attempted troubleshooting this problem by establishing a centralized storage facility in order to collect all available GNSS data and standardize both formats and metadata information. Here we describe the processes and functions that manage this unified repository, called MGA (Mediterranean GNSS Archive), which regularly collects GNSS RINEX files from alarge number of CORS (Continuously Operating Reference Station) located across a wide region of mainly the European and African plates. RINEX observation data and metadata information are provided to the analysts through an FTP server and dedicated web-services. The complete data set is stored in a PostgreSQL database in order to easily retrieve pieces of information and efficiently manage the archive content. The system implements many high-level services that include scripts to download files from remote archives and to detect new available data, web applications such as API (Application Program Interface) to interact with the system, and background services that interact with the database. During the development of this product, particular attention was paid to what has already been achieved by EPOS TCS WP10, whose objective was: "[...] to develop an open source platform with programmatic and web interfaces to store and disseminate raw data and metadata from GNSS stations operating in Europe''. Many ideas and tools presented here were inspired by that project.
