    Notes: A sudden overaccumulation of methylglyoxal (MG) induces, in Saccharomyces cerevisiae, the expression of MG-protective genes, including GPD1, GLO1 and GRE3. The response is partially dependent on the transcriptional factors Msn2p/Msn4p, but unrelated with the general stress response mechanism. Here, we show that the high-osmolarity glycerol (HOG)-pathway controls the genetic response to MG and determines the yeast growth capacity upon MG exposure. Strains lacking the MAPK Hog1p, the upstream component Ssk1p or the HOG-dependent nuclear factor Msn1p, showed a reduction in the mRNA accumulation of MG-responsive genes after MG addition. Moreover, hyperactivation of Hog1p by deletion of protein phosphatase PTP2 enhanced the response, while blocking the pathway by deletion of the MAPKK PBS2 had a negative effect. In addition, the activity of Hog1p affected the basal level of GPD1 mRNA under non-inducing conditions. These effects had a great influence on MG resistance, as hog1Δ and other HOG-pathway mutants with impaired MG-specific expression displayed MG sensitivity, whereas those with enhanced expression exhibited MG resistance as compared with the wild-type. However, MG does not trigger the overphosphorylation of Hog1p or its nuclear import in the parental strain. Moreover, dual phosphorylation of Hog1p appears to be dispensable in the triggering of the transcriptional response, although a phosphorylable form of Hog1p is fundamental for the transcriptional activity. Overall, our results suggest that the basal activity of the HOG-pathway serves to amplify the expression of MG-responsive genes under non-inducing and inducing conditions, ensuring cell protection against this toxic glycolytic by-product.

    Notes: : A novel method to assess the ripening state of Persea Americana Mill avocados was developed. The method was validated and its results compared with standard ones such as the penetrometer method. Also, comparison were made between the proposed method and the ultrasonic system reported by other authors. The correlation between the proposed method and both the standard and former ultrasonic method is quite good. The measurement system overcomes the problem of alignment between the emitter and receiver found in the resent execution development for the same purposes. Extension of proposed systems could allow the measurement with a single ultrasonic source and possibility the development of an online evaluation system for all the fruits.

    Notes: Wounding lettuce (Lactuca sativa L., var. Longifolia) leaves induced an eight-fold increase in the activity of phenylalanine ammonia-lyase (PAL; EC 4.3.1.5), and the subsequent accumulation of phenolic compounds and tissue browning at 10°C. PAL is a key enzyme in the synthesis of phenolic compounds. A PAL cDNA was previously isolated by reverse-transcription PCR using total RNA from wounded lettuce leaves. RNA gel blots showed that maximum accumulation of both PAL mRNA and PAL enzyme activity occurred 24 h after wounding. A 2-min heat shock at 45°C administered within 5 min of wounding delayed the wound-induced increase in PAL activity, but did not delay the increase in wound-induced PAL mRNA. Changes in the content of PAL protein were also followed by immunoblot using anti-PAL antibody raised against the bacterially expressed protein from the cDNA. Immunoblots showed that the level of PAL protein in wounded lettuce tissue was significantly reduced by the heat shock treatment. These data suggest that heat shock reduces the rise in wound-induced PAL enzyme activity by reducing the translation of wound-induced PAL mRNA, or by increasing the turnover of the induced PAL protein.

    Notes: Abstract  Basalts and tonalites dredged from the Amami Plateau in the northern West Philippine Basin have the geochemical characteristics of intraoceanic island arc rocks: low 87Sr/86Sr (0.70297–0.70310), intermediate 143Nd/144Nd (0.51288–0.51292), moderate light rare earth element (LREE) enrichment (La/Yb = 4.1–6.6) and high La/Nb (1.4–4.3). The incremental heating of hornblende from tonalites yielded well-defined plateaus and 40Ar/39Ar isochron ages of 115.8 ± 0.5 Ma and 117.0 ± 1.1 Ma, while plagioclase yielded disturbed Ar release patterns, with ages ranging from 70 to 112 Ma. Taken together, these results show that the Amami Plateau was formed by subduction-related magmatism in the Early Cretaceous period, earlier than indicated by prior K/Ar results. The results support tectonic models in which the West Philippine Basin was opened within a complex of Jurassic–Paleocene island arc terranes, which are now scattered in the northern West Philippine Basin, the Philippine Islands and Halmahera. The Amami Plateau tonalites and basalts have higher Sr/Y and lower Y and 87Sr/86Sr compared with younger tonalitic rocks from the northern Kyushu–Palau Ridge and the Tanzawa complex, which were formed by the subduction of the Pacific Plate beneath the Philippine Sea Plate. Based on the geochemical characteristics of the basalts, the Early Cretaceous subduction zone that formed the Amami Plateau may have been the site of slab melting, which suggests that a younger and hotter plate was being subducted at that time. However, the Amami tonalites were probably formed from basaltic magma by fractional crystallization or by partial melting of basaltic arc crust, rather than by melting of the subducted slab.

    Notes: An experimental campaign was conducted for monitoring the formation of hydrocarbongaseous compounds during bioleaching of copper sulphide ores with mesophilic microorganisms.Three different mineral samples were used: a pyrite concentrate, a chalcopyrite concentrate and acopper sulphide ore rock from El Teniente mine containing 1 % copper. Mineral samples werebioleached in 250 ml shake flasks containing 100 ml of basal medium inoculated either with a purestrain of Acidithiobacillus ferrooxidans or a natural bacteria consortium obtained from the acidleaching of El Teniente ore in columns. Each sealed system shake flask was fed with a flow ofsynthetic air and the exit stream was passed through a column containing an adsorbent material andnext through a water trap as to avoid back air contamination from the environment. Compoundspresent in the adsorbent material after 90 days of bioleaching were analysed using a gaschromatography mass spectrometry technique with a procedure that detects 162 differenthydrocarbon gaseous compounds (SGH). Results showed that the bacterial communities in coppersulphide bioleaching results in the formation of some hydrocarbon gaseous compounds, mainlyvarious types of methyl benzene and branched alkanes. The formation of these organic compounds islikely to be related to the presence of heterothrofic microorganisms present in the natural populations

    Notes: Cell behaviour such as adhesion, morphology, proliferation and functional activity are highly influenced by surface properties including hydrophobicity, roughness, texture and morphology. These surface properties may be controlled using a mixture of additive coating techniques to produce glass coatings by sol-gel process and soft lithography on dental ceramics. The purpose of this work was to compare cell adhesion and early orientation of Human Bone Marrow (HBM) cells cultured on micro-patterned (micro-PGC) and on flat glass coatings (FGC) produced by sol-gel processing. Spin coating was used to apply SiO2 flat coatings on glass substrates as model surfaces. Photolithography was applied to produce master patterns with microscale dimensions. A moulding technique was used to print micropatterned SiO2 glass coatings produced by a sol-gel process. The coatings were then sintered, sterilized and cultured with HBM cells derived from primary cultures, using a standardized protocol, for 1 and 7 days. Cell morphology and orientation were observed using scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). Flat and MPGC with line shaped features were produced. Cells presented a typical osteoblastic morphology on flat surfaces while slimmer, preferentially oriented and more elongated morphologies could be seen on line micro-patterned surfaces. HBM cells cultured on flat glass coatings showed increased tendency to spread and to assume more randomized proliferation when compared to the cells on the micro-patterned glass coatings. Micro-patterned glass coatings showed higher orientation control and smaller delay in the rate of proliferation, in early stages of in vitro culture as compared to flat coatings. These preliminary studies revealed that Micro-PGC induce significant morphological changes and controlled orientation of HBM cells during early stages of cell proliferation
