    Notes: Abstract Mouse monoclonal antibody (mAb) Pgf-II specific for a 72-kDa major cell-surface protein (72K-CSP) derived from Porphyromonas gingivalis OMZ 409 was prepared. Immunoblotting analysis revealed that mAb Pgf-II reacted with 72K-CSP but not with 41-kDa fimbrial subunit protein (41K-fimbrilin) derived from P. gingivalis 381. Electron microscopic observation revealed that P. gingivalis OMZ 409 possessed peritrichous, thin fimbriae on their surface. Immunogold electron microscopy also demonstrated that mAb Pgf-II bound to the 72K-CSP examined with the gold particles arranged along the fibril array originating from the cell surface of the bacteria. These findings suggested that P. gingivalis 72K-CSP was identifiable as another fimbriae (termed Pg-II fimbriae) different from the fimbriae (termed Pg-I fimbriae) composed of a 41K-fimbrilin. Using multipin peptide synthesis technology, 102 sequential overlapping peptides covering the entire 514 amino-acid stretch of Pg-II fimbriae were synthesised. Seven immunodominant regions within Pg-II fimbrial protein molecule, which definitely reacted with the serum of patients with periodontal diseases, were detected.

    Notes: Abstract We previously established murine hybridomas producing a monoclonal antibody monospecific against three glucosyl-transferases (I, SI and S) of Streptococcus mutans which contribute to dental caries formation. Here, we developed a new immunochemical technique (cross-dot system) with which individual levels of glucosyltransferases expressed by S. mutans can be evaluated. We also examined glucosyltransferase production and in vitro artificial plaque formation by a reference strain and several clinical isolates of S. mutans. The findings indicate that the levels of glucosyltransferases produced greatly vary with the cells and the culture medium, and that the cells producing high levels of both glucosyltransferase-SI and glucosyltransferase-I enzymes may possess high in vitro artificial plaque forming ability. We suggest that the cross-dot system will be useful for estimating the cariogenic potential of S. mutans isolates.

    Notes: The effects of stereoregularity on the low-temperature relaxation processes were studied by dynamic mechanical measurements on isotactic and syndiotactic polyisobutyl methacrylates (iso-PiBMA and syn-PiBMA). The α, β, and γ relaxation processes were observed in both stereoregular forms. Both the α, and β loss peaks were at lower temperatures for iso-PiBMA than for syn-PiBMA. The γ loss peak was observed at about -155°C at 30 Hz for both forms, and the apparent activation energy of this process was same for both samples within experimental error (6.7 ± 0.5 kcal/mole). It was reduced from these results that the α and β processes are both considerably influenced by the isotactic configuration but the γ process is not.

    Notes: The sorption properties of atactic polypropylene (APP) and isotactic polypropylene (IPP) were studied by equilibrium sorption of various organic solvents. The variation of the Flory-Huggins interaction parameter χ for the APP-CCl4 system at 25°C was expressed as a function of the volume fraction v2 of polymer by the relation: x = 0.113 exp {1.879 v2}. The average molecular weight Mc of the polymer chains between successive crystallites for IPP subjected to different thermal treatments was calculated to be 250 to 350 by the equation of Flory and Rehner. From the variation of Mc with solvent concentration, we estimated the number fraction of polymer chains actually contributing to elastic deformation. The clustering function for solvent in the polymer calculated by the method of Zimm and Lundberg decreased linearly from a positive value to -1 with increasing solvent concentration. Clustering of solvent molecules was found to occur more easily in APP than in IPP.

    Notes: The effects of swelling and annealing treatments on viscoelastic behavior were studied in melt-crystallized and solution-crystallized samples of isotactic polypropylene (iso-PP) over the temperature range -150 to 150°C. The log E″ versus T curves exhibited α, β, and γ peaks in order of decreasing temperature. The β peak of the melt-crystallized sample shifted to higher temperatures after annealing, but was not affected by swelling. The α peak of melt-crystallized polymer was affected by swelling treatments. It increased in height and shifted to lower temperatures almost linearly with the volume fraction of absorbed solvent. The magnitude of the shift was independent of the solvent species - toluene, p-xylene, tetralin, carbon tetrachloride - however, it depended significantly on the temperature at which the sample had been heat treated. For solution-crystallized polymer, no peaks in log E″ were observed in the temperature range of the β peak of melt-crystallized material, but the α peak appeared larger and broader, and at higher temperature than the corresponding peak in the melt-crystallized polymer. After swelling or annealing, the low-temperature component of the α peak of the solution-crystallized sample decreased in height and at the same time a new loss peak appeared at -55 and 0°C, respectively, is swollen and annealed samples. In particular, in the case of annealing treatments, the high-temperature component of the α peak shifted to still higher temperatures. From these results on the solution-crystallized sample it can be deduced that the segmental motions in the amorphous phase are very strongly constrained by surrounding crystalline phases as compared with those in the amorphous phase of the melt-crystallized sample, and the constraints imposed on the segmental motions are released to a great extent by both treatments. Finally, swelling effects on the γ peak were examined. The γ peak of the melt-crystallized sample decreased in height after swelling. On the other hand, the γ peak of the solution-crystallized sample separated into two peaks, which might be attributed to the mechanical relaxations in the crystalline and amorphous phases.

    Description: A green synthesis of silver nanoparticles (AgNPs) was conducted using the stem extract of Piper chaba, which is a plant abundantly growing in South and Southeast Asia. The synthesis was carried out at different reaction conditions, i.e., reaction temperature, concentrations of the extract and silver nitrate, reaction time, and pH. The synthesized AgNPs were characterized by visual observation, ultraviolet–visible (UV-vis) spectroscopy, dynamic light scattering (DLS), scanning electron microscopy (SEM), transmission electron microscopy (TEM), x-ray diffraction (XRD), energy dispersive x-ray (EDX), and Fourier transform infrared (FTIR) spectroscopy. The characterization results revealed that AgNPs were uniformly dispersed and exhibited a moderate size distribution. They were mostly spherical crystals with face-centered cubic structures and an average size of 19 nm. The FTIR spectroscopy and DLS analysis indicated that the phytochemicals capping the surface of AgNPs stabilize the dispersion through anionic repulsion. The synthesized AgNPs effectively catalyzed the reduction of 4-nitrophenol (4-NP) and degradation of methylene blue (MB) in the presence of sodium borohydride.

    Description: Steelmaking slags are a promising resource as artificial seaweed beds for the reconstitution of marine environments. To grow seaweed well, the formation of biofilms is an essential process in biofouling. This study focused on the formation of initial biofilms on steelmaking slag samples and analyzed the resulting bacterial communities using the next-generation sequencing technique. Three types of steelmaking slag were submerged in an area of Ise Bay in Mie Prefecture, Japan, for 3 and 7 days in the summer and winter seasons to allow the formation of biofilms. The bacterial communities of these biofilms were richer in sulfur-oxidizing bacteria compared to the biofilms formed on polyurethane sponges. It was found that Helicobacteraceae dominantly grew on the biofilms formed on the slag samples. This shows that steelmaking slags have potential to be used as artificial seaweed beds and marine water purifiers.

    Description: The anomalous nanoscale electromagnetic field arising from light–matter interactions in a nanometric space is called a dressed photon. While the generic technology realized by utilizing dressed photons has demolished the conventional wisdom of optics, for example, the unexpectedly high-power light emission from indirect-transition type semiconductors, dressed photons are still considered to be too elusive to justify because conventional optical theory has never explained the mechanism causing them. The situation seems to be quite similar to that of the dark energy/matter issue in cosmology. Regarding these riddles in different disciplines, we find a common important clue for their resolution in the form of the relevance of space-like momentum support, without which quantum fields cannot interact with each other according to a mathematical result of axiomatic quantum field theory. Here, we show that a dressed photon, as well as dark energy, can be explained in terms of newly identified space-like momenta of the electromagnetic field and dark matter can be explained as the off-shell energy of the Weyl tensor field.

    Description: Skin waste from tuna processing needs to be utilized, such as extraction of its collagen and gelatin. Their functional properties can be improved by enzymatic hydrolysis for conversion to peptides. Thus, the research objectives were to examine the characteristics and antioxidant activity of collagen, gelatin, and the derived peptide from yellowfin tuna skin. Collagen was extracted using 0.75 M acetic acid at 4 °C, while gelatin was prepared using 0.25% citric acid and extracted at 65 °C. Hydrolysis was carried out with 2% Alcalase, followed by fractionation with a molecular weight cut off sieve for both collagen and gelatin. Collagen yield was 22.6% with pH value of 6.63 and whiteness of 96.7%. Gelatin yield was 20.0% with pH value of 4.94 and whiteness of 51.0%. Hydrolysis for three hours resulted in 52.7% and 45.2% degree of hydrolysis for collagen and gelatin, respectively. The molecular weights of collagen peptides ranged from 2.94 to 11.93 kDa, while those of gelatin peptides ranged from 3.54 to 16,620 kDa.     Description: Xanthomonas campestris is one of bacteria carrying a type III secretion system which transports their effector proteins into host plant cells to disturb host defense system for their infection. To establish a genome editing system without introducing any foreign gene, we attempted to introduce genome editing enzymes through the type III secretion system. In a test of protein transfer, X. campestris pv. campestris (Xcc) transported a considerable amount of a reporter protein sGFP-CyaA into tobacco plant cells under the control of the type III secretion system while maintaining cell viability. For proof of concept for genome editing, we used a reporter tobacco plant containing a luciferase (LUC) gene interrupted by a meganuclease I-SceI recognition sequence; this plant exhibits chemiluminescence of LUC only when a frameshift mutation is introduced at the I-SceI recognition site. Luciferase signal was observed in tobacco leaves infected by Xcc carrying an I-SceI gene which secretes I-SceI protein through the type III system, but not leaves infected by Xcc carrying a vector control. Genome-edited tobacco plant could be regenerated from a piece of infected leaf piece by repeated selection of LUC positive calli. Sequence analysis revealed that the regenerated tobacco plant possessed a base deletion in the I-SceI recognition sequence that activated the LUC gene, indicating genome editing by I-SceI protein transferred through the type III secretion system of Xcc.
