    Notes: The Alexandrite laser belongs to the class of vibronical lasers. The broad spectral laser gain of Alexandrite laser gain of Alexandrite suggests the possibility to generate femto-second laser pulses with passive mode locking. Experiments showed, however, that a limit of pulsewidth shortening is reached near 10 ps. This effect is caused by the limited mode-locking capability of the passive absorber. The results of eight different passive absorbers used to generate mode locking are presented. The effect of bandwidth narrowing of the laser emission spectrum of Alexandrite on pulsewidth and pulse-train energy was measured for these absorbers. The dependence of laser output energy with the temperature on the active medium was also investigated.

    Notes: Experimental current–versus–bias-voltage data obtained on Hg-diffused, p-n homojunction, infrared photodiodes are critically examined against a theoretical model. A satisfactory agreement between measured and predicted data requires that the thermal impedance of the device be included into the model. Thermal calculations confirm the magnitude of the thermal impedance suggested by the model. Values of several thousand K/W are possible for front-side-illuminated detectors.

    Notes: Pneumocystis carinii organisms were isolated from viral antibody-negative rats that had been infected by intratracheal intubation of organism preparations tested negative for common bacteria and fungi. Infection scores of lungs from infected animals at the time of parasite isolation was 〉 5 (100-1,000 organisms/oil immersion field). Electron microscopy of heavily infected lungs revealed that the pathogens adhered to Type I pneumocytes and to each other, resulting in obstructions up to several cell layers thick, which extended into the alveolar lumen. Protocols for purifying the organisms were developed to optimize separation from each other and from host cells, and to optimize preparation purity, recovery efficiency, and organism viability. The study tested mucolytic agents, sieving, various centrifugation speeds, lysis of host cells by osmotic shock and filtration through membranes of different pore diameter. Final preparations contained no intact host cells as determined by light microscopy. Only minor amounts (〈 5%) of host debris were detected by electron microscopy. Most organisms and their pellicles were ultrastructurally intact but no longer adhered to one another. The final preparation was characterized biochemically by quantitation of the specific lung surfactant marker surfactant protein A, which indicated 〉 99.5% purity. The total non-P. carinii protein in the final preparation (〈 6%, depending on the level of infection) was estimated by the protein content of pelletable material resulting from processing uninfected lungs in an identical manner. Elimination of free cholesterol and phospholipids from host lung tissue was monitored during the purification process. Exogenous stigmasterol, added as an extracellular marker, decreased during the purification process and was undetectable in the final organism preparation. Yields of 108-109 organisms/rat were routinely obtained. Viability, assessed by the calcein acetoxymethyl ester-propidium iodide assay, was 80–95%.

    Notes: The standard theoretical model of two-level systems in low-temperature glasses is modified so that the temperature dependence of the effective homogeneous optical linewidth is in agreement with experiment. This alters the time dependence of the width due to spectral diffusion. The new results fit recent experiments without the need for gaps in the distribution function of flip rates of the two-level-systems or the addition of extra distribution functions. © 1996 American Institute of Physics.

    Notes: Abstract Although mycoplasmas lack cell walls, they are in many respects similar to the gram-positive bacteria with which they share a common ancestor. The molecular biology of mycoplasmas is intriguing because the chromosome is uniquely small (〈600 kb in some species) and extremely A-T rich (as high as 75 mol% in some species). Perhaps to accommodate DNA with a lower G + C content, most mycoplasmas do not have the "universal" genetic code. In these species, TGA is not a stop codon; instead it encodes tryptophan at a frequency 10 times greater than TGG, the usual codon for this amino acid. Because of the presence of TGA codons, the translation of mycoplasmal proteins terminates prematurely when cloned genes are expressed in other eubacteria, such as Escherichia coli. Many mycoplasmas possess strikingly dynamic chromosomes in which high-frequency changes result from errors in DNA repair or replication and from highly active recombination systems. Often, high-frequency changes in the mycoplasmal chromosome are associated with antigenic and phase variation, which regulate the production of factors critical to disease pathogenesis.

    Notes: We have studied optical dephasing and spectral diffusion of the S1←S0 0–0 transition of bacteriochlorophyll-a (BChl-a) in the glass 2-methyltetrahydrofuran (MTHF) at ambient (Δp=0) and high pressure (Δp=3.6 GPa) between 1.2 and 4.2 K by time-resolved hole-burning. The "effective" homogeneous linewidth Γhom′ follows a power law dependence on temperature, Γhom′=Γ0′+aT1.3±0.1, where Γ0′=Γ0+Γ0ET+Γ0ET→SD(td) is the residual linewidth and a=aPD+aSD(td)+aET→SD(td) is the coupling constant. The separate contributions to Γ0′ and a are the fluorescence decay rate Γ0=(2πτfl)−1, the "downhill" energy-transfer rate Γ0ET, the coupling constants due to "pure" dephasing aPD and "normal" spectral diffusion aSD(td), and two terms related to "extra" spectral diffusion induced by energy transfer, Γ0ET→SD(td) and aET→SD(td). We have quantitatively analyzed these contributions at ambient and high pressure. The results show that "normal" SD, "extra" SD, and ET→SD are strongly influenced by pressure. We have interpreted our findings in terms of a change in the number of two-level-systems, the low-frequency modes characteristic for the glassy state. © 1999 American Institute of Physics.
