    Notes: A new model structure is proposed for the silk I form of the crystalline domains of Bombyx mori silk fibroin and the corresponding crystal form of poly(L-Ala-Gly). It was deduced from conformational energy computations on stacked sheet structures of poly(L-Ala-Gly). The novel sheet structure contains interstrand hydrogen bonds but is composed of antiparallel polypeptide chains whose conformation differs from that of the antiparallel β-sheets that constitute the silk II structure. The strands of the new sheet have a two-residue repeat, in which the Ala residues adopt a right-handed and the Gly residues a left-handed sheet-like conformation. The computed unit cell is orthorhombic, with cell dimensions a = 8.94 Å, b = 6.46 Å, and c = 11.26 Å. The model accounts for most spacings in the observed fiber x-ray diffraction patterns of silk I and of the silk-I-like form of poly(L-Ala-Gly), and it is consistent with nmr and ir spectroscopic data. As a test of the computations, the well-established β-sheet structure of silk II and the corresponding form of poly(L-Ala-Gly) have been reproduced. The computed energies for the two forms of poly(L-Ala-Gly) indicate that the silk-II-like form is more stable, by about 1.0 kcal/mol per residue. The main difference between the two structures is the orientation of the Ala side chains of neighboring strands in each sheet. In the Pauling-Corey β-sheet and in the silk II form, referred to as an “in-register” structure, the Ala side chains of every strand point to the same side of a sheet. In the silk I structure, referred to as “out-of-register,” the side chains of Ala residues in adjacent strands point to opposite sides of the sheet.

    Notes: Dragline silk from the spider, Nephila clavipes, was characterized by thermal analysis (TGA, DSC, DMA), computational modeling, scanning electron microscopy and by quasi-static as well as high rates of strain. Thermal stability to about 230°C was observed by TGA, two transitions by DMA, -75°C, representative of localized motion in the amorphous domain, and a main chain motion associated with partial melt at 210°C. Tensile tests indicated average initial modulus, ultimate tensile strength and ultimate tensile strain of 22 GPa, 1.1 GPa and 9%, respectively. The corresponding properties of the best fibers tested were 60 GPa, 2.9 GPa and 11%, respectively. High strain rates (〉50,000%/sec) indicated similar mechanical properties to the average values indicated above. Microscopy showed compressive and tensile strains to failure of 34%. Computational modeling yielded a crystal modulus of 200 GPa.

    Notes: The systematic characterization and optimization of bovine erythrocyte glutathione peroxidase is described, together with its application in an assay employing amperometry in stirred solutions for the determination of endogenous levels of reduced glutathione (GSH). Initial studies employed UV spectroscopy, cyclic voltammetry, and amperometry in stirred solutions (to construct hydrodynamic voltammograms) for several investigations including the selection of a suitable hydroperoxide second substrate, the investigation of solution conditions on the enzyme reaction rate, and reconstituted enzyme stability. Subsequently, amperometry in stirred solutions was used to monitor the enzyme-catalyzed rate of GSH oxidation and to construct calibration plots over the concentration range 10 to 50 μM (corresponding to normal circulating GSH levels after the envisaged sample preparation step consisting of a simple twenty fold dilution of whole blood). Enzyme selectivity studies were performed using several potential physiological interferents and selected pharmaceuticals that may be encountered in the circulation of patients being treated for rheumatoid arthritis. The coefficient of variation for the enzyme linked amperometric assay was 7.28% (n = 5) for solutions containing 40 μM GSH.

    Notes: Absorbable vascular ligating clips are finding increasing use in intraabdominal surgery. We report the results of a light and scanning electron microscope investigation of the tissue reactivity and clip degradation patterns of two such materials, Absolok (polydioxanone) and Lactomer (copolymer of glycolic and lactic acid), implanted in the fascia and peritoneum of rabbits for intervals of 2 to 70 days. Cellular response to the clips, defined as the number of inflammatory cells/104 μm2, was maximum at day 4 postimplantation, then gradually declined as the duration of implantation increased. This pattern, seen with both types of clips, was similar to that seen with polydioxanone (PDS) suture, but significantly greater than that associated with polypropylene (Prolene) suture. Although cellular response to the clips was greater in peritoneum than in fascia, especially on two occasions associated with adhesion formation, this was not statistically significant. Based on our morphological observations, the signs of clip degradation which were indicated by the appearance of surface crazing and cracks occurred earlier in peritoneum than in fascia.
