    Description: Given the phenomenal popularity of mukbang (a live-streamed eating show) among young people, first in Korea and now globally, it is no longer a secret that they like to watch other people eating and cooking in the digital age. This chapter provides an overview of the evolution of mukbang culture in Korea while exploring the sociocultural meanings of this new cultural phenomenon. The chapter suggests that mukbang as a social phenomenon is deeply rooted in the precarious contexts of Korean youth, also known as the ingyeo generation. Young people’s increasing engagement with mukbang illustrates how a shifting sociocultural structure engages with an emerging affective structure through digital mediation. Young Koreans’ negotiation of their precarious present and future through vicarious experiences of binge eating implies how the basic needs of eating are mediated, spectacularized and resignified as a subcultural practice.

    Description: This open access book examines the lived experiences of diasporic Korean youth in light of the transnational flows of South Korean popular culture, known as the Korean Wave, or Hallyu. Drawing on an ethnographic study of Korean Canadian youth and their engagement with the Korean Wave, the book proposes a critical understanding of the interactions between diasporic youth audiences and popular culture. By examining the Korean Wave as diasporic cultural practices rather than the diffusion of national cultural products, the book reveals the diversified ways in which cultural flows are negotiated by audiences who take up relatively ambivalent reception positions between two or more national and cultural contexts. This book expands the scope of transnational audience studies and youth cultural studies by focusing attention on the diasporic media practices of young people.

    AKAP12 Supports Blood-Brain Barrier Integrity against Ischemic Stroke (2020)

    Description: A-kinase anchor protein 12 (AKAP12) is a scaffolding protein that associates with intracellular molecules to regulate multiple signal transductions. Although the roles of AKAP12 in the central nervous system are still relatively understudied, it was previously shown that AKAP12 regulates blood-retinal barrier formation. In this study, we asked whether AKAP12 also supports the function and integrity of the blood-brain barrier (BBB). In a mouse model of focal ischemia, the expression level of AKAP12 in cerebral endothelial cells was upregulated during the acute phase of stroke. Also, in cultured cerebral endothelial cells, oxygen-glucose deprivation induced the upregulation of AKAP12. When AKAP12 expression was suppressed by an siRNA approach in cultured endothelial cells, endothelial permeability was increased along with the dysregulation of ZO-1/Claudin 5 expression. In addition, the loss of AKAP12 expression caused an upregulation/activation of the Rho kinase pathway, and treatment of Rho kinase inhibitor Y-27632 mitigated the increase of endothelial permeability in AKAP12-deficient endothelial cell cultures. These in vitro findings were confirmed by our in vivo experiments using Akap12 knockout mice. Compared to wild-type mice, Akap12 knockout mice showed a larger extent of BBB damage after stroke. However, the inhibition of rho kinase by Y-27632 tightened the BBB in Akap12 knockout mice. These data may suggest that endogenous AKAP12 works to alleviate the damage and dysfunction of the BBB caused by ischemic stress. Therefore, the AKAP12-rho-kinase signaling pathway represents a novel therapeutic target for stroke.

    Establishment of Canine Transitional Cell Carcinoma Cell Lines Harboring BRAF V595E Mutation as a Therapeutic Target (2021)

    Description: Transitional cell carcinoma (TCC) is the most common malignant tumor of the canine urinary tract and tends to have a poor prognosis due to its invasive potential. Recent studies have reported that up to 80% of canine urothelial carcinoma has the BRAF V595E mutation, which is homologous to the human V600E mutation. Activating the BRAF mutation is an actionable target for developing effective therapeutic agents inhibiting the BRAF/mitogen-activated protein kinase (MAPK) pathway in canine cancer as well as human cancer. We established novel canine TCC cell lines from two tumor tissues and one metastatic lymph node of canine TCC patients harboring the BRAF V595E mutation. Tumor tissues highly expressed the BRAF mutant and phosphorylated extracellular signal-related kinases (ERK)1/2 proteins. The derived cell lines demonstrated activated MAPK pathways. We also evaluated the cell lines for sensitivity to BRAF inhibitors. Sorafenib, a multiple kinase inhibitor targeting RAF/vascular endothelial growth factor receptor (VEGFR), successfully inhibited the BRAF/MAPK pathway and induced apoptosis. The established canine TCC cell lines responded with greater sensitivity to sorafenib than to vemurafenib, which is known as a specific BRAF inhibitor in human cancer. Our results demonstrated that canine TCC cells showed different responses compared to human cancer with the BRAF V600E mutation. These cell lines would be valuable research materials to develop therapeutic strategies for canine TCC patients.
