    Notes: Summary The “maritime continent” area to the north of Australia is one of three major global centres of tropical convective activity and has a fundamental role in the general circulation of the atmosphere. Tropical convection within this region is dominated by island thunderstorm activity which is strongly diurnally modulated, is deep and is geographically fixed. The storms over Bathurst and Melville Islands (11.5°S, 131°E) are representative of island thunderstorms that occur throughout the maritime continent and are believed to be primarily a response to sea breeze convergence. In this paper, we produce an indicative climatology of the Melville and Bathurst Island sea breeze (obtained during a three week period of the 1988 transition season), and we examine the surface patterns of island-scale convergence and divergence, especially in relation to the diurnal modulation of island thunderstorm activity. These observations are placed in the context of the broadscale environment and the relevant surface energy balance and radiation regimes.

    Notes: Summary Twenty-five patients with von Willebrand's disease (vWD) type III were analysed with regard to blood coagulation variables and possible deletions. Nine of the probands and their families were further investigated with DNA linkage analyses. Different patterns of heredity can be suggested in our families with vWD type III, on the basis of blood coagulation analyses. The findings suggest homozygosity in five families and the possibility of compound heterozygosity or a new mutation in the proband in three families. The linkage analyses confirm the results of the coagulation analyses. The segregation of the von Willebrand factor (vWF) gene can be followed in the families, and carrier diagnosis can be made in several of the probands' relatives. The possibility of large deletions in the vWF gene of the probands and their parents was investigated with probes representing the whole vWF cDNA. No deletions were found.

    Notes: Abstract This study was planned to determine the number of origins of the mutation underlying Huntington's disease (HD) in Sweden. Haplotypes were constructed for 23 different HD families, using six different polymorphisms [(CCG) n , GT70, 674, BS1, E2 and 4.2], including two within the gene. In addition, extensive genealogical investigations were performed, and the geographical origin of the haplotypes was studied. Ten different haplotypes were observed suggesting multiple origins for the HD mutation in Sweden. Analysis of the two polymorphic markers within the HD gene (the CCG repeat and GT70) indicates that there are at least three origins for the HD mutation in Sweden. One of these haplotypes (7/A) accounts for 89% of the families, suggesting that the majority of the Swedish HD families are related through a single HD mutation of ancient origin. Furthermore, three of the families that were previously considered to be unrelated could be traced to a common ancestor in the 15th century, a finding that is consistent with this hypothesis.

    Notes: Abstract Both cellular proliferation and apoptosis (programmed cell death) have been claimed to be modulated, perhaps even triggered by, changes in intracellular pH. In this review, we summarize the evidence that gave rise to these hypotheses. To facilitate a critical appraisal of the existing data, we briefly review the main pathways involved in cytosolic pH homeostasis and their regulation by mitogens and by apoptosis-inducing agents. The information available at present suggests that cytosolic pH plays a permissive role in cellular growth and proliferation, but is neither a trigger nor an essential step in the mitogenic signal transduction cascade. Concerning apoptosis, it is clear that lowering the pH in vitro can activate DNase II. However, the evidence linking cytosolic acidification with DNA degradation in vivois presently not convincing. We conclude that the cytosolic pH, an essential physiological parameter that is tightly controlled by multiple, complementary, or redundant systems, is unlikely to play a role in signalling either cell growth or death.

    Notes: Abstract The occurrence of the alkaloidsN-formyl andN-acetyl loline, peramine, lolitrem B, and ergovaline and the response of aphids to plants containing these compounds were determined in species and cultivars ofFestuca,Lolium, and other grass genera infected with fungal endophytes (Acremonium spp., andEpichloe typhina). Twenty-nine of 34 host-fungus associations produced one or more of the alkaloids, most frequently peramine or ergovaline. Three alkaloids (lolines, peramine, and ergovaline) were found in tall fescue and in perennial ryegrass infected withA. coenophialum, while peramine, lolitrem B, and ergovaline were present in perennial ryegrass and in tall fescue infected withA. lolii and inF. longifolia infected withE. typhina. WhileA. coenophialum andA. lolii produced similar patterns of alkaloids regardless of the species or cultivar of grass they infected, isolates ofE. typhina produced either no alkaloids or only one or two different alkaloids in the grasses tested. Aphid bioassays indicated thatRhopalosiphum padi andSchizaphis graminum did not survive on grasses containing loline alkaloids and thatS. graminum did not survive on peramine-containing grasses. Ergovaline-containing grasses did not affect either aphid.

    Notes: Abstract Wet-only rainwater composition on a weekly basis was determined at four sites in West Java, Indonesia, from June 1991 to June 1992. Three sites were near the extreme western end of Java, surrounding a coal-fired power station at Suralaya. The fourth site was ∼100 km to the east in the Indonesian capital, Jakarta. Over the 12 months study period wet deposition of sulfate at the three western sites varied between 32–46 meq m−2 while nitrate varied between 10–14 meq m−2. Wet deposition at the Jakarta site was systematically higher, at 56 meq m−2 for sulfate and 20 meq m−2 for nitrate. Since sulfate and nitrate wet deposition fluxes in the nearby and relatively unpopulated regions of tropical Australia are both only ∼5 meq m−2 anthropogenic emissions of S and N apparently cause significant atmospheric acidification in Java. It is possible that total acid deposition fluxes (of S and N) in parts of Java are comparable with those responsible for environmental degradation in acid-sensitive parts of Europe and North America.

    Notes: A 1.3-Methyl Shift along the Unsaturated BCSi SkeletonThe synthesis of the methoxy[tris(trimethylsilyl)methyl]boranes X-(MeO)B-C(SiMe3)3 (1a-d; X = Cl, Br, Ph, PhCH2) is described. The methylenoboranes Me-B=C(SiMe2Hal) (3a, b), the 2,3-dihydro-1H-1-sila-3-boraindene species 6, and the 1,2,3,4-tetrahydro-1-sila-3-boranaphthalene species 7 are formed by the thermal elimination of MeOSiMe3 from 1a/b or 1c or 1d, respectively. The boranes 3a, b undergo cyclodimerization at the B=C bond at 25 and 69°C, respectively, the diboretanes 4a, b being formed. The oxaboretanes 5a, b are isolated from the reaction of 3a, b with Ph2CO. The thermal elimination of MeOSiMe3 from Me′-(MeO)B-C(SiMe3)3 (1g;; Me′ = CD3) and subsequent addition of Ph2CO gives the oxboretanes 5g′/5g″ in the ratio of 1:6, showing that an equilibrium Me′-B=C(SiMe32 ⇄ Me - B=C(SiMe3(SiMe2Me′) with a statistical distribution of Me′ among seven methyl places is established in the hot tube before Ph2CO finally attacks the double bond. All these results are in accord with the primary formation of the methyleneboranes X-B=C(SiMe3)2 from 1a-d, followed by a 1,3-shift of Me along the unsaturated BCSi skeleton and the subsequent transformation into the isolated products.

    Notes: Bafilomycin A1, a specific inhibitor of H+-ATPases of the vacuolar type, was in the present study shown, at similar concentrations, to induce secretion of lysosomal enzyme and to elevate lysosomal pH in mouse macrophages. These results lend support to the previous suggestion of a triggering role for an increase in lysosomal pH and a permissive role for cytosolic pH in the exocytosis of macrophage lysosomal enzyme. Vacuolar H+-ATPases are present in the macrophage plasma membrane as well as in intracellular membranes, for example, those of the lysosomal and phagosomal compartments. Phagosomal acidification was shown to be achieved in part by a mechanism with a similar sensitivity to bafilomycin A1 as lysosomal H+ transport and in part by an early, bafilomycin A1-insensitive mechanism. We found a lesser sensitivity towards bafilomycin A1 of the lysosomal and phagosomal H+-ATPase than that localized in the plasma membrane, indicating differences among H+-ATPases at the subcellular level. Also, by attempts to mobilize lysosomal H+-ATPase to the plasma membrane, support was obtained for the notion that subcellular H+-ATPase populations differ and thus possibly could be differentially regulated. © 1995 Wiley-Liss, Inc.
