    Description: Sediment cores collected along a transect in Crooked Pond, southeastern Massachusetts, provide evidence of water-level changes between 15,000 cal yr B.P. and present. The extent of fine-grained, detrital, organic accumulation in the basin, inferred from sediment and pollen stratigraphies, varied over time and indicates low water levels between 11,200 and 8000 cal yr B.P. and from ca. 5300 to 3200 cal yr B.P. This history is consistent with the paleohydrology records from nearby Makepeace Cedar Swamp and other sites from New England and eastern Canada and with temporal patterns of regional changes in effective soil moisture inferred from pollen data. The similarities among these records indicate that (1) regional conditions were drier than today when white pine (Pinus strobus) grew abundantly in southern New England (11,200 to 9500 cal yr B.P.); (2) higher moisture levels existed between 8000 and 5500 cal yr B.P., possibly caused by increased meridonal circulation as the influence of the Laurentide ice sheet waned; and (3) drier conditions possibly contributed to the regional decline in hemlock (Tsuga) abundances at 5300 cal yr B.P. Although sea-level rise may have been an influence, moist climatic conditions during the late Holocene were the primary reason for a dramatic rise in water-table elevations.

    Description: Deschampsia antarctica Desv. is one of two vascular plants from the Maritime Antarctic. It is usually exposed to cold, salt, and desiccating winds. We hypothesize that D. antarctica has genes that encode dehydrin proteins and their expression is regulated by low temperature, salt or osmotic stress. To test this hypothesis a fragment of a dehydrin gene from D. antarctica was identified and used as a probe to study dehydrin expression under low temperature, salt, and osmotic stress, and exogenous ABA (abscisic acid) treatments. An anti-dehydrin antibody was also used to study dehydrin protein accumulation under the same treatments. Southern analysis of genomic DNA treated with different endonucleases showed more than four bands recognized by the probe, suggesting that D. antarctica has several dehydrin genes. Northern analysis showed two putative dehydrin transcripts of 1.0 kb accumulated only under exogenous ABA and 1.6 kb under osmotic and salt treatments, suggesting that D. antarctica would have ABA-dependent and - independent pathways for regulation of dehydrin expression. Western analysis showed seven dehydrin proteins (58, 57, 55, 53, 48, 30 and 27 kDa) under the different stress treatments. Cold-accumulated dehydrin proteins were immunolocalized, showing that they are associated with vascular and epidermal tissue, which are preferential ice nucleation zones.

    Description: Deschampsia antarctica is a freezing-tolerant plant and the only native Poaceae that grows in the Maritime Antarctic. During the long days of the growing season this plant accumulates sucrose (Suc) in the leaves to 36% of the dry weight. The mechanism that leads to this high accumulation is unknown. The effect of day length and low temperature on sucrose phosphate synthase (SPS) (EC: 2.4.1.14) activity and sugar accumulation was studied in D. antarctica and compared with other Poaceae. Three different day lengths: short (SD) (8/16 h), medium (MD) (16/8 h) and long (LD) (21/3 h); and two temperatures: 4°C (cold-acclimated) and 15°C (non-acclimated) were tested. The highest contents in total soluble sugars (TSS) and Suc were reached in crowns and leaves, respectively, in cold-acclimated plants under LD. TSS and Suc contents and SPS activity with cold acclimation were higher in D. antarctica than in other agricultural (wheat, oat and barley) and non-agricultural (D. caespitosa and D. beringensis) Poaceae species. Suc/TSS ratio was higher in all Deschampsia species than in agricultural Poaceae species. SPS activity and sucrose content in leaves were positively correlated only in LD cold acclimated plants. This result shows that SPS activity is responsive to day length in D. antarctica.
