    Description: Abstract One known method to reduce vortex shedding from the tip of a blade is the use of end plates or winglets. Although the aerodynamic impact of such end plates has been investigated in the past, no studies exist on the effect of such end plates on the far-field noise. The aeroacoustic noise reduction of three different end-plate geometries is experimentally investigated. The end plates are applied to the free end of a wall-mounted symmetric NACA 0012 airfoil and a cambered NACA 4412 airfoil with an aspect ratio of 2 and natural boundary layer transition. Microphone array measurements are taken in the aeroacoustic open-jet wind tunnel at BTU Cottbus-Senftenberg for chord-based Reynolds numbers between 75,000 and 225,000 and angles of attack from 0$$^circ$$ ∘ to 30$$^circ$$ ∘ . The obtained acoustic spectra show a broad frequency hump for the airfoil base configurations at higher angles of attack that is attributed to tip noise. Hot-wire measurements taken for one configuration show that the application of an end plate diffuses the vorticity at the tip. The aeroacoustic noise contribution of the tip can be reduced when the endplates are applied. This reduction is most effective for higher angles of attack, when the tip vortex is the dominant sound source. Graphic abstract

    Description: Extracellular signals drive the nucleation of the NLRP3 inflammasome which leads to the release of cytokines and causes inflammatory events. Hence, the inflammasome has gained enormous momentum in biomedical basic research. The detailed mechanisms of inflammasome generation and regulation remain to be elucidated. Our study was directed toward the design, convergent synthesis, and initial biochemical evaluation of activity-based probes addressing NLRP3. For this purpose, probes were assembled from a CRID3/MCC950-related NLRP3-binding unit, a linker portion and a coumarin 343 fluorophore or biotin. The affinity of our probes to NLRP3 was demonstrated through SPR measurements and their cellular activity was confirmed by reduction of the interleukin 1β release from stimulated bone marrow-derived macrophages. The initial characterizations of NLRP3-targeting probes highlighted the coumarin probe 2 as a suitable tool compound for the cellular and biochemical analysis of the NLRP3 inflammasome.

    Description: Silvicultural interventions such as strip cuttings can change the resource availability of the edge trees. This may alter tree allometry, as light regime, water, and nutrient availability can change at the forest edge. Increased root growth may optimize resource uptake and/or enhance tree anchorage to withstand the altered wind regime. However, little is known about the patterns of the root–shoot allometric responses to strip cuttings. In three alpine stands differing in climate, site productivity, and stand characteristics, we selected 71 Norway spruce trees and took increment cores from stems, root collars, and main roots. This enabled us to study changes in the long-term root-stem allometry for 46 years and short-term allometric responses to intervention. The effects of cutting were compared between edge trees and trees from the stand interior in 10 years before and after the intervention. The long-term allocation to roots increased with stem diameter, with the strongest effects on the regularly managed stand with the tallest and largest trees. These results support the allometric biomass partitioning theory, which postulates resource allocation patterns between different plant organs to depend on plant size. Strip cutting on north-facing slopes boosted edge-tree growth in all plant compartments and enhanced allocation to roots. This change in allometry started 2 years after cutting but disappeared 7–8 years later. In the post-cutting period, the highest root–shoot increase was observed in the small trees independent of the site. This indicates the change in growing conditions to have the strongest effects in formerly suppressed trees. Thus, the effect of such acclimation on the wind firmness of subdominant spruce trees is a question with high importance for optimizing cutting layouts in lowering post-cutting vulnerability to disturbance. The results from this case study contribute to a better understanding of the structural acclimation of spruce trees from high-elevation forests to new forest edges. However, for a more mechanistic understanding of environmental drivers, further analyses of tree-ring stable isotopes are recommended.

    Description: In order to detect the misuse of testosterone (T), urinary steroid concentrations and concentration ratios are quantified and monitored in a longitudinal manner to enable the identification of samples exhibiting atypical test results. These suspicious samples are then forwarded to isotope ratio mass spectrometry (IRMS)–based methods for confirmation. Especially concentration ratios like T over epitestosterone (E) or 5α-androstanediol over E proved to be valuable markers. Unfortunately, depending on the UGT2B17 genotype and/or the gender of the athlete, these markers may fail to provide evidence for T administrations when focusing exclusively on urine samples. In recent years, the potential of plasma steroids has been investigated and were found to be suitable to detect T administrations especially in female volunteers. A current drawback of this approach is the missing possibility to confirm that elevated steroid concentrations are solely derived from an administration of T and cannot be attributed to confounding factors. Therefore, an IRMS method for plasma steroids was developed and validated taking into account the comparably limited sample volume. As endogenous reference compounds, unconjugated cholesterol and dehydroepiandrosterone sulfate were found suitable, while androsterone and epiandrosterone (both sulfo-conjugated) were chosen as target analytes. The developed method is based on multi-dimensional gas chromatography coupled to IRMS in order to optimize the overall assay sensitivity. The approach was validated, and a reference population encompassing n = 65 males and females was investigated to calculate population-based thresholds. As proof-of-concept, samples from volunteers receiving T replacement therapies and excretion study samples were investigated. Graphical abstract
