    Notes: Common bunt caused by Tilletia tritici and T. laevis has occurred worldwide and reduces yield and quality in common and durum wheats. The development of DNA markers linked to bunt resistance to race T1 in the cross, ‘Laura’(S) בRL5407’ (R), was carried out in this study based on the single head derived F4:5 and single seed derived F4:6 populations. Bulked segregant analysis was used to identify two random amplified polymorphic DNA (RAPD) markers linked to the gene for resistance to race T1 in the spelt wheat ‘RL5407′. The two markers identified, UBC548590 and UBC274988, flanked the resistance gene with a map distance of 9.1 and 18.2 cM, respectively. The former was linked in repulsion phase to bunt resistance while the later was in coupling phase. The two RAPD markers and the common bunt-resistance gene all segregated in Mendelian fashion. Use of these two RAPD markers together could assist in incorporating the bunt-resistance gene from spelt wheat into common wheat cultivars by means of marker-assisted selection.

    Notes: Paracoccus pantotrophus grown anaerobically under denitrifying conditions expressed similar levels of the periplasmic nitrate reductase (NAP) when cultured in molybdate- or tungstate-containing media. A native PAGE gel stained for nitrate reductase activity revealed that only NapA from molybdate-grown cells displayed readily detectable nitrate reductase activity. Further kinetic analysis showed that the periplasmic fraction from cells grown on molybdate (3 μM) reduced nitrate at a rate of Vmax=3.41±0.16 μmol [NO3−] min−1 mg−1 with an affinity for nitrate of Km=0.24±0.05 mM and was heat-stable up to 50°C. In contrast, the periplasmic fraction obtained from cells cultured in media supplemented with tungstate (100 μM) reduced nitrate at a much slower rate, with much lower affinity (Vmax=0.05±0.002 μmol [NO3−] min−1 mg−1 and Km=3.91±0.45 mM) and was labile during prolonged incubation at 〉20°C. Nitrate-dependent growth of Escherichia coli strains expressing only nitrate reductase A was inhibited by sub-mM concentrations of tungstate in the medium. In contrast, a strain expressing only NAP was only partially inhibited by 10 mM tungstate. However, none of the above experimental approaches revealed evidence that tungsten could replace molybdenum at the active site of E. coli NapA. The combined data show that tungsten can function at the active site of some, but not all, molybdoenzymes from mesophilic bacteria.

    Notes: Assembly of each Salmonella typhimurium flagellum filament requires export and polymerisation of ca. 30 000 flagellin (FliC) subunits. This is facilitated by the cytosolic chaperone FliS, which binds to the 494 residue FliC and inhibits its polymerisation. Yeast two-hybrid assays, co-purification and affinity blotting showed that FliS binds specifically to the C-terminal 40 amino acid component of the disordered D0 domain central to polymerisation. Without FliS binding, the C-terminus is degraded. Our data provide further support for the view that FliS is a domain-specific bodyguard preventing premature monomer interaction.

    Notes: Using an alkaline phosphatase-based genetic screening method, we identified a number of proteins that are potentially located on the outer surface of Group B streptococcus (Streptococcus agalactiae). In an enzyme-linked immunosorbent assay, antisera raised against two of the proteins, the streptococcal yutD homologue and a subunit of an ABC transporter, recognised clinically important serotypes of Group B streptococcus. In a neonatal rat model, purified IgG from the sera conferred significant levels of protection against a lethal challenge infection. The proteins identified show potential as protein subunit candidates for vaccines against Group B streptococcal disease in neonates.

    Notes: Nitric oxide (NO) has a broad spectrum of signalling and regulatory functions and multiple molecular targets. Recently, the intrabacterial toxicity of NO and mechanisms for NO resistance have been intensively investigated. Here we report for the first time that NO elicits release of zinc from a bacterial protein. Using the zinc-responsive expression of zntA (encoding a Zn-exporting P-type ATPase) fused to lacZ, i.e. Φ(zntA-lacZ), to monitor intracellular zinc, and SmtA (the Synechococcus metallothionein) as zinc store, we have shown that the NO donors NOC-5 and NOC-7 elicit zinc ejection. No increase in Φ(zntA-lacZ) activity was observed in a zntR mutant, indicating the specificity of the zntA promoter response to zinc ions.

    Notes: Juvenile plaice Pleuronectes platessa in a laboratory arena used intensive search behaviour, characterized by short movements and frequent turning, in the five movements before and after attacking a prey in an aggregated distribution. They used extensive search behaviour with, on average, longer movements and less turning at all other times. Intensive search was, apparently, triggered by a high local density of prey but not by isolated prey. This response to local prey density resulted in area-restricted search when prey were aggregated and win-shift behaviour when prey were dispersed. There was no evidence that the use of intensive search increased with experience of aggregated prey. It therefore appears that the fish were able to exploit encountered prey distribution patterns using their immediate perceptions rather than prior experience.

    Notes: When offered a size range of shrimp Crangon crangon, fewer naïve reared turbot Scophthalmus maximus fed than did wild fish, and those that fed took smaller prey. Analysis of feeding behaviour indicated differences between wild and naïve reared fish in the motivation to feed on novel prey and in prey recognition and capture efficiency. Feeding efficiency and motivation increased with experience and reared fish achieved the feeding rate of wild fish within the 9 days of the experiment. The ‘creep’ style of approach to shrimp was innate to naïve reared turbot. A comparative feeding experiment confirmed that experienced reared fish ate more prey than naïve reared fish. Naïve reared turbot selected pellets and attacked stones preferentially to shrimp, in contrast to wild and experienced reared fish, which selected shrimp. Stones were attacked due to the memory of pellet-like visual characteristics and this behaviour persisted in some reared fish for at least 6 weeks, illustrating a cost of memory in which changing environmental conditions cause previously learnt information to become misleading. The experimental data also demonstrated that differences may arise in the prey and feeding behaviour of wild and naïve reared fish, but that such differences diminish with experience.

    Description: Space weather effects can strongly influence high-frequency (HF) communications by changing the ionospheric environment through which the radio waves propagate. Since many systems utilize HF communications, the ability to make real-time assessments of propagation conditions is an important part of space weather monitoring systems. In this paper, we present new techniques for measuring high-latitude HF communications link parameters using data from SuperDARN radars. These techniques use ground-scatter returns to define the variation in skip distance with frequency. From these data, the maximum usable frequency (MUF) as a function of range is determined and ionospheric critical frequencies are estimated. These calculations are made in near-real-time and the results are made available on the World Wide Web. F-region critical frequencies calculated using this method show good agreement with ionosonde data.Key words.
