    Description: Arachidonic acid (AA) is the precursor for prostaglandin E2 (PGE2) synthesis and increases growth of prostate cancer cells. To further elucidate the mechanisms involved in AA-induced prostate cell growth, induction of c-fos expression by AA was investigated in a human prostate cancer cell line, PC-3. c-fos mRNA was induced shortly after addition of AA, along with a remarkable increase in PGE2 production. c-fos expression and PGE2 production induced by AA was blocked by a cyclo-oxygenase inhibitor, flurbiprofen, suggesting that PGE2 mediated c-fos induction. Protein kinase A (PKA) inhibitor H-89 abolished induction of c-fos expression by AA, and partially inhibited PGE2 production. Protein kinase C (PKC) inhibitor GF109203X had no significant effect on c-fos expression or PGE2 production. Expression of prostaglandin (EP) receptors, which mediate signal transduction from PGE2 to the cells, was examined by reverse transcription polymerase chain reaction in several human prostate cell lines. EP4 and EP2, which are coupled to the PKA signalling pathway, were expressed in all cells tested. Expression of EP1, which activates the PKC pathway, was not detected. The current study showed that induction of the immediate early gene c-fos by AA is mediated by PGE2, which activates the PKA pathway via the EP2/4 receptor in the PC-3 cells.

    Description: The mechanism by which the proto-oncogene, c-fos, is up-regulated in response to PGE2 in the mouse osteoblastic (MC3T3-E1) cell line was investigated using RT-PCR. c-fos messenger RNA up-regulation by dmPGE2 is rapid, starting 10 min post stimulation, and transient. The specific protein kinase A (PKA) inhibitor, H89, inhibited c-fos induction. Moreover, down-regulation of protein kinase C (PKC) activity by chronic TPA treatment had no effect on the induction of c-fos by dmPGE2. We conclude that up-regulation of c-fos by dmPGE2 is primarily dependent on PKA in MC3T3-E1 osteoblasts. In S49 lymphoma wild-type but not S49 cyc- cells, which are deficient in cAMP signaling, dmPGE2 up-regulates c-fos and increases cell growth compared with unstimulated cells. Thus in S49 lymphoma cells, c-fos induction by PGE2 is also dependent on cAMP signaling. The minimal c-fos promoter region required for dmPGE2-induced expression was identified by transfecting c-fos promoter deletion constructs coupled to the chloramphenicol acetyltransferase (CAT) reporter gene into Vero cells. Transfection of a plasmid containing 99 bp c-fos proximal promoter was sufficient to direct c-fos/CAT expression following stimulation with dmPGE2. Because induction of c-fos is mediated by cAMP, these data are consistent with activation of c-fos via the CRE/ATF cis element.

    Description: Although our understanding of effects of space flight on human physiology has advanced significantly over the past four decades, the potential contribution of stress at the cellular and gene regulation level is not characterized. The objective of this ground-based study was to evaluate stress gene regulation in cells exposed to altered gravity and environmentally suboptimal conditions. We designed primers to detect message for both the constitutive and inducible forms of the heat shock protein, HSP-70. Applying the reverse transcriptase-polymerase chain reaction (RT-PCR), we probed for HSP-70 message in human acute T-cell leukemia cells, Jurkat, subjected to three types of environmental stressors: (1) altered gravity achieved by centrifugation (hypergravity) and randomization of the gravity vector in rotating bioreactors, (2) serum starvation by culture in medium containing 0.05% serum, and (3) temperature elevation (42 degrees C). Temperature elevation, as the positive control, significantly increased HSP-70 message, while centrifugation and culture in rotating bioreactors did not upregulate heat shock gene expression. We found a fourfold increase in heat shock message in serum-starved cells. Message for the housekeeping genes, actin and cyclophilin, were constant and comparable to unstressed controls for all treatments. We conclude that gravitational perturbations incurred by centrifugal forces, exceeding those characteristic of a Space Shuttle launch (3g), and culture in rotating bioreactors do not upregulate HSP-70 gene expression. In addition, we found RT-PCR useful for evaluating stress in cultured cells. Copyright 2000 Wiley-Liss, Inc.

    Description: Leonardo-BRDF (Bidirectional Reflectance Distribution Function) is a new NASA mission concept proposed to allow the investigation of radiative transfer and its effect on the Earth's climate and atmospheric phenomenon. Enabled by the recent developments in small-satellite and formation flying technology, the mission is envisioned to be composed of an array of spacecraft in carefully designed orbits. The different perspectives provided by a distributed array of spacecraft offer a unique advantage to study the Earth's albedo. This paper presents the flight dynamics analysis performed in the context of the Leonardo-BRDF science requirements. First, the albedo integral is investigated and the effect of viewing geometry on science return is studied. The method used in this paper, based on Gauss quadrature, provides the optimal formation geometry to ensure that the value of the integral is accurately approximated. An orbit design approach is presented to achieve specific relative orbit geometries while simultaneously satisfying orbit dynamics constraints to reduce formation-keeping fuel expenditure. The relative geometry afforded by the design is discussed in terms of mission requirements. An optimal Lambert initialization scheme is presented with the required Delta-V to distribute all spacecraft from a common parking orbit into their appropriate orbits in the formation. Finally, formation-keeping strategies are developed and the associated Delta-V's are calculated to maintain the formation in the presence of perturbations.

    Description: The NASA John H. Glenn Research Center and the U.S. Department of Energy are currently developing a Stirling convertor for use as an advanced spacecraft power system for future NASA deep-space missions. As part of this development, a Stirling Technology Demonstrator Convertor (TDC) was recently tested to verify its survivability and capability of withstanding its expected launch random vibration environment. The TDC was fully operational (producing power) during the random vibration testing. The output power of the convertor was measured during the testing, and these results are discussed in this paper. Numerous accelerometers and force gauges were also present which provided information on the dynamic characteristics of the TDC and an indication of any possible damage due to vibration. These measurements will also be discussed in this paper. The vibration testing of the Stirling TDC was extremely successful. The TDC survived all its vibration testing with no structural damage or functional performance degradation. As a result of this testing, the Stirling convertor's capability to withstand vibration has been demonstrated, enabling its usage in future spacecraft power systems.

    Description: The characterization and understanding of the acoustic field within a launch vehicle's payload fairing (PLF) is critical to the qualification of a spacecraft and ultimately to the success of its mission. Acoustic measurements taken recently for the Cassini mission have allowed unique opportunities to advance the aerospace industry's knowledge in this field. Prior to its launch, the expected liftoff acoustic environment of the spacecraft was investigated in a full-scale acoustic test of a Titan IV PLF and Cassini simulator in a reverberant test chamber. A major goal of this acoustic ground test was to quantify and verify the noise reduction performance of special barrier blankets that were designed especially to reduce the Cassirii acoustic environment. This paper will describe both the ground test and flight measurements, and compare the Cassini acoustic environment measured during launch with that measured earlier in the ground test. Special emphasis will be given to the noise reduction performance of the barrier blankets and to the acoustic coherence measured within the PLF.

    Description: We determine the distances to the z approximately equal to 0.55 galaxy clusters MS 0451.6-0305 and CL 0016+16 from a maximum likelihood joint fit to interferometric Sunyaev-Zel'dovich effect (SZE) and X-ray observations. We model the intracluster medium (ICM) using a spherical isothermal beta-model. We quantify the statistical and systematic uncertainties inherent to these direct distance measurements, and we determine constraints on the Hubble parameter for three different cosmologies. For an OmegaM = 0.3, OmegaL = 0.7 cosmology, these distances imply a Hubble constant of 63(exp 12)(sub -9)(exp +21)(sub -21) km/s/Mpc, where the uncertainties correspond to statistical followed by systematic at 68% confidence. The best fit H(sub o) is 57 km/sec/Mpc for an open OmegaM = 0.3 universe and 52 km/s/Mpc for a flat Omega = 1 universe.
