    Notes: Abstract We have studied the effects of various meteorologicalparameters and plant distribution on the airborneconcentrations of three different pollen types:Chenopodiaceae-Amaranthaceae, Quercus and Olea, in the city of Cartagena, situated in SouthwestEurope. The aerobiological study has been carried outfrom March 1993 to March 1997, using a Hirst-typeLanzoni 2000 trap, situated on the roof of the citytrain station. Meteorological data were supplied bythe Environmental Service of Cartagena's Town Hall.Daily pollen concentrations were standardised.Correlation analysis showed the importance ofmeteorological data on airborne pollen concentrations.On the other hand, ANOVA analyses depicted theinfluence of vegetation distribution in relation tosampler location and wind directions. Finally,multiple regression analyses allowed us to developcorrelation models for the three different pollentypes considered, accounting for 32%(Chenopodiaceae-Amaranthaceae), 19% (Quercus),and 26% (Olea) of the variation in the airbornepollen concentration.

    Notes: Abstract  The levels and burdens of Cd, Zn, Cu and Hg were measured in the dorsal mantle, digestive gland and gonads of the squid Illex argentinus, from the Southwest Atlantic Ocean. Mature and immature individuals of both sexes were analysed. Correlations of heavy metal concentrations and burdens with sex and food habits were studied. The highest metal levels were found, particularly for Cd, in the digestive gland, which attained a concentration of up to 270 μg g−1 (wet weight). Dorsal mantle and gonads exhibited the same order of metal enrichment: Zn〉Cu〉Cd. Zinc and Cu levels were higher in ovaries than in testes and varied with the stage of maturation. The dorsal mantle exhibited the lowest heavy metal concentrations. Mercury levels were below the detection limit of the method in all the tissues analysed.

    Notes: Abstract. In planta Agrobacterium-mediated transformation combined with a soil-based herbicide selection for transgenic plants was used to recover large numbers of transgenic Arabidopsis plants for functional genomic studies. A tissue-culture-free system for generating transgenic plants was achieved by infiltrating Arabidopsis plants with Agrobacterium tumefaciens harboring a binary T-DNA vector containing the phosphinothricin acetyltransferase gene from Streptomyces hygroscopicus, and by selecting transgenic Arabidopsis growing in soil by foliar application of the herbicide Finale (phosphinothricin). Analysis of herbicide-resistant plants indicated that all were transgenic and that the T-DNA transformation process occurred late during flower development, resulting in a preponderance of independently derived T-DNA insertions. T-DNA insertions were usually integrated in a concatenated, rearranged form, and using linkage analysis, we estimated that T1 plants carried between one and five T-DNA loci. Using pooling strategies, both DNA and seed pools were generated from about 38,000 Arabidopsis plants representing over 115,000 independent T-DNA insertions. We show the utility of these transgenic lines for identifying insertion mutations using gene sequence and PCR-based screening.

    Notes: Abstract: Analysis was conducted during 1991 into the fecundity and oocyte size distribution of pumpkinseed, roach and rudd in Lake Banyoles. We found that individual fecundities of these species in Lake Banyoles were lower in comparison with other localities. In roach, there was a moderately significant negative relationship between latitude and egg number. The temporal pattern of the pumpkinseed's oocyte size distribution suggests that it is a multispawner fish that spawns several batches of eggs from mid-May to mid-August. In contrast, roach and rudd seem to be monospawners fish that release only one batch of eggs before mid-May. The monospawning behaviour of rudd in Lake Banyoles is in contrast to the evidence from other waters. Variations in fecundity and spawning mode are discussed on the basis that they are due mainly to differences in local conditions.

The New World primate Aotus nancymaae has been recommended by the World Health Organization (WHO) as a model for evaluation of malaria vaccine candidates, given its susceptibility to experimental infection with the human malaria parasites Plasmodium falciparum and Plasmodium vivax. We present here the nucleotide sequences of the complete cDNA of MHC-DQA1 and of the polymorphic exon 2 segments of MHC-DQB1/DQB2. In a group of three nonrelated animals captured in the wild, five alleles of MHC-DQA1 could be identified. They all belong to one lineage, namely Aona-DQA1*27. This lineage has not been described in any other New World monkey species studied. In a group of 19 unrelated animals, 14 Aona-DQB1 alleles could be identified which are grouped into the two lineages Aona-DQB1*22 and Aona-DQB1*23. These lineages have been described previously in the common marmoset and cotton-top tamarin. In addition, two Aona-DQB2 sequences could be identified which are highly similar to HLA-DQB2 sequences. Essential amino acid residues contributing to MHC DQ peptide binding pockets number 1 and 4 are conserved or semi-conserved between HLA-DQ and Aona-DQ molecules, indicating a capacity to bind similar peptide repertoires. These results fully support the use of Aotus monkeys as an animal model for evaluation of future subunit vaccine candidates.

    Notes: Abstract  Sequences of Aotus nancymaae immunoglobulin kappa light-chain rearrangements were analyzed after reverse transcription polymerase chain reaction. Among 22 in-frame rearrangements analyzed, 12 IGKV genes belonging to the families 1, 2, or 3 were identified. Aotus counterparts for all five human IGKJ genes were found. The identity of the deduced human and Aotus amino acid sequences was between 83% and 92% for junctional regions and 74% for the constant region. Sequence comparisons between rearrangements indicated that somatic mutations, the addition of nongermline-encoded nucleotides, and exonuclease trimming contribute to the generation of diversity of Aotus immunoglobulin kappa chains. The high identity of Aotus and human IGK genes is comparable to that of T-cell receptor genes and further supports the proposal to use the Aotus Plasmodium falciparum infection model for the evaluation of malaria vaccine candidates.

    Notes: Abstract  Inoculation of the oilseed halophyte Salicornia bigelovii Torr. with eight species of halotolerant bacteria, grown in seawater-irrigated pots under environmental conditions native to the plant's habitat, resulted in significant plant growth promotion by the end of the growing season, 8–11 months later. Statistical analysis demonstrated that inoculation with Azospirillum halopraeferens, a mixture of two Azospirillum brasilense strains, a mixture of Vibrio aestuarianus and Vibrio proteolyticus, or a mixture of Bacillus licheniformis and Phyllobacterium sp. significantly increased plant height and dry weight at the end of the season. Some of the bacterial strains also increased the number of side branches and the size of the spikes. The bacteria did not affect the number of seeds or their weight. Inoculation with the mangrove cyanobacterium Microcoleus chthonoplastes had no effect on plant foliage variables. At the end of the growing season, the N and protein content of the plant foliage was significantly reduced by bacterial inoculation; however, the N and protein content of seeds significantly increased. The P content in foliage increased significantly in plants treated with all the bacteria except M. chthonoplastes, whereas the total lipid content of foliage increased significantly only when plants were inoculated with a mixture of A. brasilense strains or with M. chthonoplastes. In three inoculation treatments palmitic acid in seeds significantly increased and linoleic acid significantly decreased. This study demonstrates the feasibility of using bacteria to promote the growth of halotolerant plants cultivated for forage and seed production in proposed seawater-irrigated agriculture.

    Notes: Abstract  This study was carried out to investigate the effect of very high cadmium concentrations (50 and 500 μg Cd g–1 soil) on some biochemical and microbiological measurements under laboratory conditions involving daily soil samplings. The data for both DTPA- and water-soluble Cd showed two distinctive patterns during soil incubation; from 0 to 4 days, values were about 50–500 and 1–100 μg g–1 dry weight soil, whereas they decreased markedly after 7 days. Both daily respiration and the ATP content but not the microbial biomass C determined by the fumigation–extraction method were lowered by high DTPA- and water-soluble Cd concentrations. Dehydrogenase and phosphatase activities as well as both enzyme activity : microbial biomass ratios were decreased by the high DTPA- and water-soluble Cd concentrations. In the first 2 days of incubation, the metabolic quotient (qCO2) was also decreased by the highest values of available Cd. The early (after 6 h) mineralization of l- but not d-glutamic acid to CO2 was inhibited during the 0–4 day incubation period by the highest Cd concentration. Possibly the l-enantiomer was used by a larger fraction of soil microorganisms than the d-enantiomer or, if they were used by the same fraction of soil microorganisms, the d-enantiomer was mineralized at a lower rate. The l- : d-glutamic acid respiration ratio was decreased by the high available Cd content because under polluted conditions soil microorganisms probably discriminated less between the two stereoisomers of glutamic acid.
