    Notes: Raloxifene, also known as Evista®, has recently been approved for the prevention of osteoporosis. Three glucuronidated compounds: raloxifene-6-glucuronide, raloxifene-27-glucuronide, and raloxifene-6,27-diglucuronide are known metabolites of raloxifene in man. Reference standards of the three glucuronides were needed for clinical trials. Although chemical routes exist to make the two mono-glucuronides, these routes were unable to provide material to meet the needs of clinical trial standards. No chemical route existed to synthesize the di-glucuronide. A bioconversion process using the microorganism Streptomyces sp NRRL 21489 was identified and scaled up. The biotranformation products were prepared in a tank fermentation, purified, and characterized by UV, LC/MS and NMR spectroscopy.

    Notes: Abstract Conventional high resolution studies of varved sediments are able to identify clastic and biogenic laminae, but are often unable to resolve the nature of fine-scale lamination contained therein. This intra-annual signal provides us with the highest potential resolution from the sedimentary record and can be resolved using scanning electron microscopy (SEM). Six case studies from lacustrine and marine settings are presented to illustrate the combination of clastic and biogenic fabric types typically found in laminated sediments. Clastic laminae fabrics include those which originate through grain settling and those which are ‘event deposits’. The correct identification of event deposits is essential if varves are to be used chronologically. SEM-based biogenic laminae fabric studies have identified seasonal faunal successions where individual laminae may be less than 100 μ thick and most recently, deep chlorophyll maxima (DCM) summer diatom floras, providing an insight into seasonal scale processes. High resolution lamina fabric studies can provide a basis for generating records of seasonal and inter-annual variability, thus contributing to our understanding of lacustrine and marine processes and palaeoenvironmental interpretation.

    Notes: Abstract In this review of sediment denitrification in estuaries and coastal ecosystems, we examine current denitrification measurement methodologies and the dominant biogeochemical controls on denitrification rates in coastal sediments. Integrated estimates of denitrification in coastal ecosystems are confounded by methodological difficulties, a lack of systematic understanding of the effects of changing environmental conditions, and inadequate attention to spatial and temporal variability to provide both seasonal and annual rates. Recent improvements in measurement techniques involving 15 N techniques and direct N2 concentration changes appear to provide realistic rates of sediment denitrification. Controlling factors in coastal systems include concentrations of water column NO 3 − , overall rates of sediment carbon metabolism, overlying water oxygen concentrations, the depth of oxygen penetration, and the presence/absence of aquatic vegetation and macrofauna. In systems experiencing environmental change, either degradation or improvement, the importance of denitrification can change. With the eutrophication of the Chesapeake Bay, the overall rates of denitrification relative to N loading terms have decreased, with factors such as loss of benthic habitat via anoxia and loss of submerged aquatic vegetation driving such effects.

    Notes: Abstract With the increasingly competitive commercial production of target proteins by hybridoma and genetically engineered cells, there is an urgent requirement for biosensors to monitor and control on-line and in real time the growth of cultured cells. Since growth is accompanied by an enthalpy change, heat dissipation measured by calorimetry could act as an index for metabolic flow rate. Recombinant CHO cell suspensions producing interferon-γ were pumped to an on-line flow calorimeter. The results showed that an early reflection of metabolic change is size-specific heat flux obtained from dividing heat flow rate by the capacitance change of the cell suspension, using the on-line probe of a dielectric spectroscope. Comparison of heat flux with glucose and glutamine fluxes indicated that the former most accurately reflected decreased metabolic activity. Possibly this was due to accumulation of lactate and ammonia resulting from catabolic substrates being used as biosynthetic precursors. Thus, the heat flux probe is an ideal on-line biosensor for fed-batch culture. A stoichiometric growth reaction was formulated and data for material and heat fluxes incorporated into it. This showed that cell demand for glucose and glutamine was in the stoichiometric ratio of ∼3:1 rather than the ∼5:1 in the medium. It was demonstrated that the set of stoichiometric coefficients in the reaction were related through the extent of reaction (advancement) to overall metabolic activity (flux). The fact that this approach can be used for medium optimisation is the basis for an amino-acid-enriched medium which improved cell growth while decreasing catabolic fluxes.

    Notes: Abstract We describe how to express constraints in a functional (semantic) data model, which has a working implementation in an object database. We trace the development of such constraints from being integrity checks embedded in procedural code to being something declarative and self-contained, combining data access and computation, that can be moved around into other contexts in intelligent distributed systems. We see this as paralleling and extending the original vision of functions as values in functional programming systems. It is greatly helped by using a referentially transparent functional formalisation. We illustrate these ideas by showing how constraints can move around within database systems (Colan & Angelic Daplex), being transformed for various uses, or even moved out into other systems and fused into a specification for a configuration problem. We look forward to future directions involving Agents.

    Notes: Abstract The aims of this study were: (1) to analyze individual variation in frog locomotor performance, (2) to compare the thermal sensitivity of jumping and swimming, and (3) to contrast whole animal versus muscle fiber performance at different temperatures. The jumping and swimming performance of Rana temporaria was analyzed at 5, 10, 15 and 20 °C. Muscle fiber bundles were isolated from lateral gastrocnemius and subjected to the length and activation patterns thought to occur in vivo. As temperature increased, locomotor performance in R. temporaria improved with a Q 10 of 1.2 for both jump take-off velocity and mean swimming velocity. The slope of the relationship between performance and temperature (TE) was similar for both locomotor parameters and was described by the equation z-scores of locomotor performance = 0.127 × TE − 1.585. Although some frogs performed better than others relative performance was affected by locomotor type and temperature. Locomotor performance improved with temperature as the power required during take-off and the mean muscle power output increased with Q 10 values of 1.7 and 1.6 respectively. The mean muscle power output during take-off was only 34% of the calculated requirements for the whole animal, suggesting the involvement of elastic strain energy storage mechanisms.
