    Notizen: A crucial step in converting an actively growing Bacillus subtilis cell into a dormant spore is the formation of a cell within a cell. This unusual structure is created by a phagocytosis-like process in which the larger mother cell progressively engulfs the adjacent smaller forespore. Only mutations blocking engulfment at an early stage and affecting genes expressed in the mother cell have been identified. Here we describe a new locus, spoIIQ, which is transcribed in the forespore and which encodes a membrane-bound protein required at a late stage of engulfment. Immunofluorescence microscopy analysis have shown that SpoIIQ is initially targeted to the septum at the boundary between the two cells and then spreads around the entire membrane of the forespore. Septum targeting requires only the first 52 residues of SpoIIQ as well as unidentified forespore-specific components. Electron-microscopy studies of cells engineered to activate the mother-cell program of gene expression independently of the forespore indicate that other as yet uncharacterized genes are involved in engulfment and that this morphological process is driven from both sides of the forespore envelope.

    Notizen: The usefulness of artificial neural networks (ANN) for milk shelf-life prediction by multivariate interpretation of gas chromatographic profiles and flavor-related shelf-life was evaluated and compared to principal components regression (PCR). The training set consisted of dynamic headspace gas chromatographic data collected during storage of pasteurized milk (input information for the neural network used to make a decision) and its corresponding shelflife (prediction or response). ANN had better predictability than PCR. A standard error of the estimate of 2 days in shelf-life resulting from regression analysis of experimental vs predicted values indicated a high predictability of ANN.

    Notizen: Abstract A transformation system has been developed for the pathogen fungus Botrytis cinerea, based on the utilization of the wide host plasmid pUT737 that contains the Sh ble gene, conferring resistance to phleomycin. Transformed protoplasts were regenerated at 10–25 μg ml−1 of phleomycin, at a frequency of 25–40 transformants per μg of DNA, and they were resistant up to 50 μg ml−1. Southern hybridization using undigested and digested total DNA showed the presence of circular autonomously replicating plasmid pUT737 in the transformants. Reisolated plasmid from transformed fungus transformed E. coli and rescued plasmid was identified as pUT737. Transformants were grown for four generations under non-selective conditions and replicative plasmids were still detected. Plasmids present in all transformants at this stage had been modified from native pUT737 and showed the same size and configuration indicating that selection through stabilizing plasmid forms has happened.

    Notizen: Abstract In this paper we present an extension to the co-design approach based on LOTOS presented in Fourth International Workshop on Hardware-Software Co-Design, 1996. In this new version we add a prototyping stage to our design flow, that allows to validate the design at the implementation level. We present the complete approach, stressing the prototyping stage after partitioning. An example of an Ethernet bridge serves us to illustrate our approach and present some results.

    Notizen: Abstract Most siderophores are catabolism resistant because of their non-peptidic or cyclic peptide chemical structures. Siderophore degradation is thus a rare event, and one which has received little attention. Based on literature precedence and the generation of monohydroxamic acid(s) by cell-free extracts of a deferrioxamine B degrading bacterium, a catabolic scheme of the siderophore is proposed. Data are also presented concerning parameters such as the pH profile, inducible nature of the siderophore-degrading metabolism and the ability of the deferrioxamine B degrading bacterium to metabolize some of the hypothesized products of deferrioxamine B dissimilation.

    Notizen: Abstract This paper shows that imprecisely stated discounts in brand promotions offered in the form of a low-probability lottery can lead to higher sales (purchase intentions) and consequently profits than equally costly conventional promotions offering a precise discount on the entire stock. Results from two different experimental studies support our findings. For high-probability lottery-like promotions, imprecise discounts lead to a lower performance for the brand than conventional promotions. We attempt to explain the findings by drawing on the behavioral decision theory literature.

    Notizen: Summary Different assay conditions induce changes in the ferric chelate reductase activities of leaf plasma membrane preparations from Fe-deficient and Fe-sufficient sugar beet. With an apoplasttype assay medium the ferric chelate reductase activities did not change significantly when Fe(III)-EDTA was the substrate. However, with ferric citrate as substrate, the effect depended on the citrateto-Fe ratio. When the citrate-to-Fe ratio was 20 ∶ 1, the effects were practically unappreciable. However, with a lower citrate-to-Fe ratio of 5 ∶ 1 the activities were significantly lower with the apoplast-type medium than with the standard assay medium. Our data also indicate that anaerobiosis during the assay facilitates the reduction of ferric malate and Fe(III)-EDTA by plasma membrane preparations. Anaerobiosis increased by approximately 50% the plasma membrane ferric chelate reductase activities when Fe(III)-EDTA was the substrate. With ferric malate anaerobiosis increased activities by 70–90% over the values obtained in aerobic conditions. However, with ferric citrate the increase in activity by anaerobiosis was not significant. We have also tested the effect of riboflavin, flavin adenine dinucleotide, and flavin mononucleotide on the plasma membrane ferric chelate reductase activities. The presence of flavins generally increased activities in plasma membrane preparations from control and Fe-deficient plants. Increases in activity were generally moderate (lower than twofold). These increases occurred with Fe(III)-EDTA and Fe(III)-citrate as substrates.
