Notizen: This study examines the legitimacy of using the reaction kinetics of low molecular weight model compounds in solution to predict the chemical kinetics of polymer-bound species in a homogeneous melt. The reaction under study takes place between an aliphatic secondary amine, diisooctadecylamine (DiOA), and a 5-membered anhydride ring, saturated maleic anhydride (MA), forming an amic acid product. The MA species was present as a pendant graft on either a model compound, dodecane-g-(maleic anhydride) (dodecane-g-MA), or a polymer chain, linear low-density polyethylene-g-(maleic anhydride) (LLDPE-g-MA). Pseudo-second-order kinetics of the anhydride consumption are followed by infrared spectroscopy, either in situ in dodecane solution or by scanning frozen film samples taken from a linear low-density polyethylene melt. It was found that the LLDPE-g-MA/DiOA system reacted at a slightly slower rate than the dodecane-g-MA/DiOA system in the low-viscosity solution at 140°C. In the melt, the dodecane-g-MA/DiOA system experienced a small decrease in the overall reaction rate compared to the same reaction carried out in dodecane. However, the LLDPE-g-MA/DiOA system underwent a 65% decrease in the observed second-order rate constant on going from a solution to the melt. To explain these phenomena, the effects of diffusion, miscibility, and chain entanglements in the melt are examined here. © 1998 John Wiley & Sons, Inc. J Polym Sci A: Polym Chem 36: 1573-1582, 1998

Notizen: Cross-linkable poly[1-(trimethylsilyl)-1-propyne] (PTMSP) films were cast from toluene solutions containing PTMSP and either 4,4′-diazidobenzophenone or 4,4′-(hexafluoroisopropylidene)diphenyl azide. The composite films were clear and homogeneous and were cross-linked by UV irradiation at room temperature or thermal annealing at 180°C. Low levels of the bis(aryl azide) (1-5 wt %) were effective in rendering the films insoluble in toluene and THF, both good solvents for PTMSP. The process is simple and effective, and thus PTMSP can be readily converted to mechanically stable membranes with permeabilities and separation factors comparable or higher than those of poly(dimethylsiloxane). The films were characterized by measuring their density, their permeability toward O2 and N2, and their spectroscopic properties. Compared to PTMSP, films containing bis(aryl azide) cross-linkers had lower permeabilities and higher separation factors, consistent with a reduction in free volume. When the films were cross-linked photochemically, the permeabilities declined further and the separation factor increased. Films cross-linked thermally had permeabilities comparable to their PTMSP/azide precursors, and density and swelling measurements suggest that higher free volumes are obtained in thermally cross-linked films. All films stored in air suffered from a slow decline in permeability which may reflect slow surface oxidation of the films. When stored in vacuum, cross-linked films were stable and showed no loss in permeability, but the permeability of uncross-linked PTMSP films stored under the same conditions fell to 70% of their original value in 1 month. We attribute the permeability decline to densification accelerated by impurities and solvents. © 1998 John Wiley & Sons, Inc. J Polym Sci B: Polym Phys 36: 959-968, 1998

Notizen: Cardiovascular device development often relies upon large-animal models to assess blood biocompatibility prior to initiating clinical trials. Unfortunately, the amount of information gleaned from such trials is limited by simple assays that do not take full advantage of immuno-technological advances that increasingly are applied in clinical studies. Thus we have developed and tested new flow cytometric techniques for measuring circulating activated bovine platelets and platelet microaggregates. Monoclonal antibodies (MAbs) raised against both activated and quiescent bovine platelets were incubated with control and PMA- or ADP-stimulated whole blood. Selected MAbs detected activated bovine platelets and platelet microaggregates in vitro with flow cytometry. Five calves implanted with one of two designs of nonpulsatile ventricular-assist devices (VADs) were followed with these assays prior to and during VAD implantation. Circulating activated bovine platelets and microaggregates increased after implantation in all animals and, alternatively, remained elevated or returned toward preimplant levels. Platelet activation percentages as detected temporally by three MAbs were correlated with one another, and platelet activation was correlated with microaggregate formation. In summary, these new methods for the sensitive measurement of circulating activated bovine platelets and microaggregates may provide valuable information for the development and assessment of future cardiovascular device designs. © 1998 John Wiley & Sons, Inc. Notizen: Four calcium phosphate cement formulations were implanted in the rabbit distal femoral metaphysis and middiaphysis. Chemical, crystallographic, and histological analyses were made at 2, 4, and 8 weeks after implantation. When implanted into the metaphysis, part of the brushite cement was converted into carbonated apatite by 2 weeks. Some of the brushite cement was removed by mononuclear macrophages prior to its conversion into apatite. Osteoclastlike cell mediated remodeling was predominant at 8 weeks after brushite had converted to apatite. The same histological results were seen for brushite plus calcite aggregate cement, except with calcite aggregates still present at 8 weeks. However, when implanted in the diaphysis, brushite and brushite plus calcite aggregate did not convert to another calcium phosphate phase by 4 weeks. Carbonated apatite cement implanted in the metaphysis did not transform to another calcium phosphate phase. There was no evidence of adverse foreign body reaction. Osteoclastlike cell mediated remodeling was predominant at 8 weeks. The apatite plus calcite aggregate cement implanted in the metaphysis that was not remodeled remained as poorly crystalline apatite. Calcite aggregates were still present at 8 weeks. There was no evidence of foreign body reaction. Osteoclastlike cell remodeling was predominant at 8 weeks. Response to brushite cements prior to conversion to apatite was macrophage dominated, and response to apatite cements was osteoclast dominated. Mineralogy, chemical composition, and osseous implantation site of these calcium phosphates significantly affected their in vivo host response. © 1998 John Wiley & Sons, Inc. Notizen: A set of yeast strains based on Saccharomyces cerevisiae S288C in which commonly used selectable marker genes are deleted by design based on the yeast genome sequence has been constructed and analysed. These strains minimize or eliminate the homology to the corresponding marker genes in commonly used vectors without significantly affecting adjacent gene expression. Because the homology between commonly used auxotrophic marker gene segments and genomic sequences has been largely or completely abolished, these strains will also reduce plasmid integration events which can interfere with a wide variety of molecular genetic applications. We also report the construction of new members of the pRS400 series of vectors, containing the kanMX, ADE2 and MET15 genes. © 1998 John Wiley & Sons, Ltd.

Beschreibung: The objective of this study is to investigate the effect of nonlinear aerodynamics, especially at high angles-of-attack with leading-edge separation, on the TCA flutter properties at transonic speeds. In order to achieve that objective, flutter simulations with Navier-Stokes CFD must be performed. To this end, time-marching Navier-Stokes solutions are computed for the TCA wing/body configuration at high angles-of-attack in transonic flight regimes. The approach is to perform non-linear flutter calculations on the TCA at two angles-of-attack, the first one being a case with attached flow (a=2.8 degrees) and the second one being a high angle-of-attack case with a wing leading edge vortex (a=12.11 degrees). Comparisons of the resulting histories and frequency damping information for both angles-of-attack will evaluate the impact of high-alpha aerodynamics on flutter.

Beschreibung: We present the analysis of a coordinated set of observations from the POLAR Ultraviolet Imager (UVI), ground magnetometers, incoherent scatter radar, solar wind monitors, DMSP and GOES satellites, focused on a traveling convection vortex (TCV) event on 24th July 1996. Starting at approximately 10:48 UT, around magnetometers in Greenland and northern Canada observe pulsations consistent with the passing overhead of a series of alternating TCV filed-aligned current pairs. Azimuthal scans by the Sondrestrom incoherent scatter radar located near Kangerlussuaq (formerly Sondrestrom), Greenland, at this time show strong modulation in the strength and direction of ionospheric plasma flow. The magnetometer pulsations grow in magnitude over the next hour, peaking in intensity at 11:39 UT, at which time images form the UVI instrument show a localized intensification of auroral emissions over central and western Greenland. Subsequent images show the intensification grow in strength and propagate westward (tailward) until approximately 11:58 UT at which time the intensification fades. These observations are consistent with the westward passage of two pairs of moderately intense TCVs over central Greenland followed by a third very intense TCV pair. The intensification of auroral emissions at 11:39 UT is associated with the trailing vortex of the third TCV pair, thought to be the result of an upward field aligned current. The modulated flow observed by the radar is the result of the strong electric fields associated with the impulsive TCV related field aligned current systems as they pass through the field of view of the radar. Measurements of the solar wind from the V;IND and IMP-8 spacecraft suggest that a pressure change may be responsible for triggering the first two pairs of TCVS, and that a subsequent sudden change in the orientation of the interplanetary magnetic field may have produced the intensification of the third TCV pair and the associated auroral brightening. Magnetometer data from the GOES satellite located over the eastern United States at geostationary orbit is consistent with a series of field-aligned moving tailward past the satellite. DMSP particle data indicated that the TCVs occur on field lines which map to the boundary plasma sheet (BPS).

Beschreibung: A spacecraft includes heat-generating payload equipment, and a heat transport system with a cold plate thermally coupled to the equipment and a capillary-wick evaporator, for evaporating coolant liquid to cool the equipment. The coolant vapor is coupled to a condenser and in a loop back to the evaporator. A heated coolant reservoir is coupled to the loop for pressure control. If the wick is not wetted, heat transfer will not begin or continue. A pair of check valves are coupled in the loop, and the heater is cycled for augmentation pumping of coolant to and from the reservoir. This augmentation pumping, in conjunction with the check valves, wets the wick. The wick liquid storage capacity allows the augmentation pump to provide continuous pulsed liquid flow to assure continuous vapor transport and a continuously operating heat transport system. The check valves are of the ball type to assure maximum reliability. However, any type of check valve can be used, including designs which are preloaded in the closed position. The check valve may use any ball or poppet material which resists corrosion. For optimum performance during testing on Earth, the ball or poppet would have neutral buoyancy or be configured in a closed position when the heat transport system is not operating. The ball may be porous to allow passage of coolant vapor.
