    Notes: Abstract Observations of suspected planet-forming disks provide estimates of protoplanetary disk masses, surface temperatures, and the rate at which mass is infalling onto the disks. Analyses of primitive meteorites and comets and their components constrain the solar nebula's temperature at the locations and times where those components were formed. Theoretical models of disks undergoing the accretion of mass from an infalling cloud envelope predict disk temperatures in good agreement with these constraints: a moderately warm (500-1500 K) inner disk, surrounded by a cool (50-150 K) outer disk. These models have important implications for the depletion of volatiles in the inner Solar System, for mechanisms of disk evolution, and for the orbital distances at which terrestrial and gas giant planets form.

    Notes: Abstract We previously described the isolation of a variant subline of HL-60 cells that does not differentiate in response to nitric oxide (NO)-generating agents or to cGMP analogs [7]. The variant cells have normal guanylate cyclase activity and normal NO-induced increases in the intracellular cGMP concentration. We now show that the variant cells have normal cGMP-dependent protein kinase (G-kinase) activity, both by an in vitro and in vivo assay, and using two-dimensional gel electrophoresis we have identified six G-kinase substrates in the parental cells. Of these six proteins, we found considerably less phosphorylation of one of the proteins in the variant cells than in parental cells, both in vitro and in intact cells, and by 35S-methionine/35S-cysteine incorporation we found much less of this protein in the variant cells than in parental cells. The protein is a shared substrate of cAMP-dependent protein kinase (A-kinase); since cAMP analogs still induce differentiation of the variant cells, it appears that the NO/cGMP/G-kinase and cAMP/A-kinase signal transduction pathways share some but not all of the same target proteins in inducing differentiation of HL-60 cells.

    Notes: Abstract A variety of fibre coatings have been modelled as a means to reduce the thermal residual stresses in sapphire/NiAl composites, including the use of molybdenum as a compliant layer. However, no experimental studies have been performed to quantify the effect of these coatings on the residual stresses. Measuring the effect of the coatings in real composites is a difficult task given the limitations of the most common residual stress measurement methods, such as X-ray and neutron diffraction. In this study, a continuous indentation system was used to measure a spatial hardness distribution both within the molybdenum fibre coatings and the NiAl matrix. Because it has been shown that residual stresses effect the apparent hardness of a material, the hardness distribution should be indicative of the local residual stress state in the sample. The measured hardness distributions showed distinct trends, including an increasing hardness in the molybdenum as the NiAl interface is approached, a substantial increase in NiAl hardness at the fibre/coating boundary, and a decrease in NiAl hardness that is proportional to the distance from the fibre/coating boundary. The potential utility of continuous indentation systems for mapping the residual stresses in composites is briefly discussed. © 1998 Kluwer Academic Publishers

    Notes: A new glycopeptide antibiotic, MDL 63,246 (Hepta-tyr), of the teicoplanin family, has been evaluated in capillary electrophoresis for the resolution of chiral compounds of pharmaceutical and environmental interest. Electrophoretic separations were carried out in a polyacrylamide-coated capillary using the partial filling-counter current mode with aqueous-organic buffers in the pH range 4-6. Experimental parameters affecting resolution, such as antibiotic concentration, buffer pH, organic modifier type and capillary temperature, were studied. The Hepta-tyr antibiotic exhibited a high enantiorecognition capability towards the studied compounds at very low concentrations (1-2 mg/mL). The optimum experimental conditions were achieved by using a buffer at pH 5 containing acetonitrile at 25°C.

    Notes: An evaluation of capillary zone electrophoresis-mass spectrometry (CZE-MS) as an analytical methodology for the separation and characterization of complex glycopeptides and nonglycopeptide structures has been performed. The evaluation employed endoproteinase V8 digested recombinant human erythropoietin (rHuEPO) that was further fractionated by reverse phase chromatography. The peptides were subjected to sequence analysis and evaluated by capillary electrophoresis, with or without mass detection, for peptide purity. The peptide mass determined from the sequence was then compared to the mass obtained from CZE-MS. Glycosylation sites and carbohydrate branch patterns were easily determined, site specific microheterogeneity (either O-acetylation of N-acetylneur-aminic acids or lactosamine extensions of the carbohydrate chain length) was assessed directly, glycosylation site occupancy was evaluated qualitatively, and nonglycopeptides were resolved and analyzed on-line with ease. Incomplete peptide digestion products were detected and identified by CZE-MS. Protein sequence coverage by CZE-MS was 98.2 percent complete from a single map. Offline evaluation of peptide purity by CZE greatly aided the interpretation of multiple sequence analysis and, in validating that, the CZE-MS was detecting all peptides present. All off-line CZE and on-line CZE-MS experiments employed a capillary that was dynamically coated with Polybrene in the presence of polyethylene glycol; separations were conducted in 0.67 M formic acid.
