    Notes: The temperature dependence of plant growth rate is related to the temperature dependence of respiratory metabolism. To determine how the effects of temperature on respiration rate and efficiency are transmitted to growth, this study measured the dark metabolic heat rate (q) and CO2 production rate (RCO2) in excised shoots of seedlings of 14 maize cultivars (Zea mays L.) at several temperatures. The temperature coefficients of q and RCO2 differ within a given cultivar and also differ among the cultivars. Both q and RCO2 exhibit an isokinetic temperature of 20 ± 3 °C. The measured temperature dependences of q and RCO2 were used to model the temperature dependences of both growth and substrate carbon conversion efficiency. This procedure may be useful in determining the suitability of cultivars for growth in a given climate and in understanding metabolic adaptation to climate.

    Notes: Surface roughness has been shown to affect differentiation and local factor production of MG63 osteoblast-like cells. This study examined whether surface roughness alters cellular response to circulating hormones such as 1α,25-(OH)2D3. Unalloyed titanium (Ti) disks were pretreated with HF/HNO3 (PT) and then were machined and acid-etched (MA). Ti disks also were sandblasted (SB), sandblasted and acid etched (CA), or plasma sprayed with Ti particles (PS). The surfaces, from smoothest to roughest, were: PT, MA, CA, SB, and PS. MG63 cells were cultured to confluence on standard tissue culture polystyrene (plastic) or the Ti surfaces and then treated for 24 h with either 10-8M or 10-7M 1α,25-(OH)2D3 or vehicle (control). Cellular response was measured by assaying cell number, cell layer alkaline phosphatase specific-activity, and the production of osteocalcin, latent (L) TGFβ, and PGE2. Alkaline phosphatase activity was affected by surface roughness; as the surface became rougher, the cells showed a significant increase in alkaline phosphatase activity. Addition of 1α,25-(OH)2D3 to the cultures caused a dose-dependent stimulation of alkaline phosphatase activity that was synergistic with the effect caused by surface roughness alone. 1α,25-(OH)2D3 also caused a synergistic increase in osteocalcin production as well as local factor (LTGFβ and PGE2) production on the rougher CA, SB, and PS surfaces, but it had no effect on the production on smooth surfaces. The inhibitory effect of surface roughness on cell number was not affected by 1α,25-(OH)2D3 except on the SB surface. 1α,25-(OH)2D3 decreased cell number, increased alkaline phosphatase activity and osteocalcin production, and had no effect on LTGFβ or PGE2 production by MG63 cells grown on tissue culture polystyrene. These data suggest that bone cell response to systemic hormones is modified by surface roughness and that surface roughness increases the responsiveness of MG63 cells to 1α,25-(OH)2D3.     Notes: Surface roughness affects proliferation, differentiation (alkaline phosphatase and osteocalcin), local factor production [transforming growth factor (TGFβ) and prostaglandin E2 (PGE2)], and response to 1,25-(OH)2D3 (1,25) of MG63 osteoblast-like cells. In this study, we examined whether the effect of surface roughness on MG63 cells is mediated by prostaglandins produced by the cells. Unalloyed titanium (Ti) disks were pretreated with HF/HNO3 (PT) and then machined and acid-etched (MA). Disks were also coarse grit-sandblasted (SB), coarse grit-sandblasted and acid-etched (CA), or plasma-sprayed with Ti particles (PS). The surfaces, from smoothest to roughest, were PT, MA, CA, SB, and PS. MG63 cells were cultured to confluence on the Ti disks in the presence or absence of 10-7M indomethacin (Indo), a specific inhibitor of cyclooxygenase activity, resulting in decreased prostaglandin production. When the cells reached confluence, cell number, cell layer alkaline phosphatase specific activity (ALPase), and osteocalcin (OC) and latent TGFβ (LTGFβ) production were determined. In addition, confluent cultures which had been grown in the absence of Indo were exposed to 10-7M 1,25, 10-7M Indo, or a combination of the two for 24 h. On the rougher surfaces, cell number was decreased and ALPase, OC, and LTGFβ were increased. When indomethacin was present throughout the culture period, the effect of surface roughness on cell number, OC, and LTGFβ was abolished. ALPase was reduced, but surface roughness-dependent effects were still observed. Addition of indomethacin to confluent cultures for 24 h had no effect on any of the parameters examined, with one exception: Cells cultured on MA surfaces exhibited a more differentiated phenotype. 1,25 increased all parameters examined on SB, CA, and PS surfaces. When indomethacin was added with 1,25, the 1,25-dependent effects on cell number and OC and LTGFβ production were abolished; however, ALPase was unaffected. This indicates that bone cell response to systemic hormones may be modified by implant surface roughness.     Notes: The use of perioral bristles (modified vibrissae) by 17 captive Florida manatees and approximately 20 wild manatees was analyzed. Captive manatees were fed six species of aquatic vegetation normally eaten in the wild (four freshwater species and two seagrasses). Inanimate objects were placed in the holding tanks with manatees at Lowry Park Zoological Gardens (Tampa, FL) to determine the degree to which perioral bristles were used in exploration and to define the range of manipulative behavior. In addition, behavioral observations were made on the use of perioral bristles during social interactions with conspecifics. Observations were recorded using a Hi8-format video camera. Florida manatees possess an unusually large degree of fine motor control of the snout and perioral bristles. The large and robust perioral bristle fields of the upper lip were used in a prehensile manner during feeding. Bristle use by manatees feeding on submerged vegetation differed from that seen during feeding on floating vegetation. Other behavioral use of the perioral bristles shows variation depending upon the situation encountered. The degree of plasticity of perioral bristle use supports our hypothesis that the vibrissal-muscular complex of the Florida manatee has evolved to increase the efficiency of grazing and browsing on aquatic vegetation and to fully maximize the potential of the manatee as a generalist feeder. The manipulative and sensitive nature of the manatee snout is likely a manifestation of a complex sensory and motor system which has evolved for marine mammal aquatic herbivores living in shallow turbid habitats.

    Notes: Development of multiple insect resistance in tropical maize represents a major effort of the maize breeding programme at CIMMYT. Resistance to the southwestern corn borer (SWCB) is polygenically controlled with primarily additive gene action. Our main objective was to identify quantitative trait loci (QTL) involved in resistance to SWCB. Other objectives were to detect QTL in the same population for plant height, female flowering, and the anthesis-silking interval (ASI). A population of 472 F2 individuals derived from a cross between the susceptible line Ki3 and the resistant inbred CML139, was restriction fragment length polymorphism (RFLP) genotyped using 110 maize probes. F3 families were rated for leaf-feeding damage after artificial infestation at one location in three consecutive years. Height and flowering were measured in protected trials in two locations. QTL analyses were conducted using joint composite interval mapping. Seven QTL on chromosomes 3, 5, 6, 8, and 9 explained 30% of the phenotypic variance (σ2p) for SWCB damage. Most QTL alleles conferring resistance were contributed from CML139. QTL showed dominance, partial dominance and additive gene action. Eleven QTL dispersed across the genome were determined to affect plant height and explained 43% of σ2p. Four of these were in close proximity to loci with qualitative effects on plant height. Thirteen QTL (50% of sigma;2p) were identified for days to female flowering and nine (30% of σ2p) for ASI. Our results, along with those from other mapping studies at CIMMYT, are allowing us to formulate marker-assisted selection schemes to complement the breeding efforts for such complex traits as borer resistance.

    Notes: The photosynthetic response of Larrea tridentata Cav., an evergreen Mojave Desert shrub, to elevated atmospheric CO2 and drought was examined to assist in the understanding of how plants from water-limited ecosystems will respond to rising CO2. We hypothesized that photosynthetic down-regulation would disappear during periods of water limitation, and would, therefore, likely be a seasonally transient event. To test this we measured photosynthetic, water relations and fluorescence responses during periods of increased and decreased water availability in two different treatment implementations: (1) from seedlings exposed to 360, 550, and 700 μmol mol–1 CO2 in a glasshouse; and (2) from intact adults exposed to 360 and 550 μmol mol–1 CO2 at the Nevada Desert FACE (Free Air CO2 Enrichment) Facility. FACE and glasshouse well-watered Larrea significantly down-regulated photosynthesis at elevated CO2, reducing maximum photosynthetic rate (Amax), carboxylation efficiency (CE), and Rubisco catalytic sites, whereas droughted Larrea showed a differing response depending on treatment technique. Amax and CE were lower in droughted Larrea compared with well-watered plants, and CO2 had no effect on these reduced photosynthetic parameters. However, Rubisco catalytic sites decreased in droughted Larrea at elevated CO2. Operating Ci increased at elevated CO2 in droughted plants, resulting in greater photosynthetic rates at elevated CO2 as compared with ambient CO2. In well-watered plants, the changes in operating Ci, CE and Amax resulted in similar photosynthetic rates across CO2 treatments. Our results suggest that drought can diminish photosynthetic down-regulation to elevated CO2 in Larrea, resulting in seasonally transient patterns of enhanced carbon gain. These results suggest that water status may ultimately control the photosynthetic response of desert systems to rising CO2.

    Notes: The effects of CO2 enrichment on photosynthesis and ribulose-1,5-bisphosphate carboxylase/ oxygenase (Rubisco) in current year and 1-year-old needles on the same branch were studied on Pinus radiata D. Don. trees growing for 4 years in large, open-top chambers at ambient (36 Pa) and elevated (65 Pa) CO2 partial pressures. At this age trees were 3·5–4 m tall. Measurements made late in the growing cycle (March) showed that photosynthetic rates at the growth CO2 concentration [(pCO2)a] were lower in 1-year-old needles of trees grown at elevated CO2 concentrations than in those of trees grown at ambient (pCO2)a. At elevated CO2 concentrations Vcmax (maximum carboxylation rate) was reduced by 13% and Jmax (RuBP regeneration capacity mediated by maximum electron transport rate) by 17%. This corresponded with photosynthetic rates at the growth (pCO2)a of 4·68 ± 0·41 μmol m–2 s–1 and 6·15 ± 0·46 μmol m–2 s–1 at 36 and 65 Pa, respectively (an enhancement of 31%). In current year needles photosynthetic rates at the growth (pCO2)a were 6·2 ± 0·72 μmol m–2 s–1 at 36 Pa and 10·15 ± 0·64 μmol m–2 s–1 at 65 Pa (an enhancement of 63%). The smaller enhancement of photosynthesis in 1-year-old needles at 65 Pa was accompanied by a reduction in Rubisco activity (39%) and content (40%) compared with that at 36 Pa. Starch and sugar concentrations in 1-year-old needles were not significantly different in the CO2 treatments. There was no evidence in biochemical parameters for down-regulation at elevated (pCO2)a in fully fexpanded needles of the current year cohort. These data show that enhancement of photosynthesis continues to occur in needles after 4 years’ exposure to elevated CO2 concentrations. Photosynthetic acclimation reduces the degree of this enhancement, but only in needles after 1 year of growth. Thus, responses to elevated CO2 concentration change during the lifetime of needles, and acclimation may not be apparent in current year needles. This transitory effect is most probably attributable to the effects of developmental stage and proximity to actively growing shoots on sink strength for carbohydrates. The implications of such age-dependent responses are that older trees, in which the contribution of older needles to the photosynthetic biomass is greater than in younger trees, may become progressively more acclimated to elevated CO2 concentration.

    Notes: The influence of vesicular–arbuscular mycorrhizal (M) colonization on biomass production and photosynthesis of Trifolium repens L. was investigated in two experiments in which the foliar nitrogen and phosphorus contents of non-mycorrhizal (NM) plants were manipulated to be no lower than that of M plants. Throughout both experiments there was a stimulation in the rate of CO2 assimilation of the youngest, fully expanded leaf of M compared with NM plants. In addition, M plants exhibited a higher specific leaf area compared with NM plants, a response that maximized the area available for CO2 assimilation per unit of carbon (C) invested. Despite the increased rate of photosynthesis in M plants there was no evidence that the additional C gained was converted to biomass production of M plants. It is suggested that this additional C gained by colonized plants was allocated to the mycorrhizal fungus and that it is the fungus, by acting as a sink for assimilates, that facilitated the stimulation in the rate of photosynthesis of the plant partner.

    Notes: The paper describes an experimental programme on an existing full-scale plant to demonstrate the conditions which are necessary to achieve nitrification. The programme was guided by parallel computer modelling of the plant using the WRc STOAT simulation package. Nitrification was successfully achieved, and the studies and computer modelling indicated how the plant should be extended to provide year-round nitrification. The effect of storm flows on effluent quality was assessed for each process option. The effluent quality consent of 5 mg/l ammoniacal nitrogen as a 95 percentile was achieved, and there was a significant improvement in river water quality with respect to ammoniacal nitrogen.

    Notes: Metabolism is the conversion of available energy sources to those energy forms required for sustaining and propagating living organisms; this is simply biological energy conversion. Proteins are the machines of metabolism; they are the engines of motility and the other machines that interconvert energy forms not involving motion. Accordingly, metabolic engineering becomes the use of natural protein-based machines for the good of society. In addition, metabolic engineering can utilize the principles, whereby proteins function, to design new protein-based machines to fulfill roles for society that proteins have never been called upon throughout evolution to fulfill.This article presents arguments for a universal mechanism whereby proteins perform their diverse energy conversions; it begins with background information, and then asserts a set of five axioms for protein folding, assembly, and function and for protein engineering. The key process is the hydrophobic folding and assembly transition exhibited by properly balanced amphiphilic protein sequences. The fundamental molecular process is the competition for hydration between hydrophobic and polar, e.g., charged, residues. This competition determines Tt, the onset temperature for the hydrophobic folding and assembly transition, Nhh, the numbers of waters of hydrophobic hydration, and the pKa of ionizable functions.Reported acid-base titrations and pH dependence of microwave dielectric relaxation data simultaneously demonstrate the interdependence of Tt, Nhh and the pKa using a series of microbially prepared protein-based poly(30mers) with one glutamic acid residue per 30mer and with an increasing number of more hydrophobic phenylalanine residues replacing valine residues. Also, reduction of nicotinamides and flavins is shown to lower Tt, i.e., to increase hydrophobicity.Furthermore, the argument is presented, and related to an extended Henderson-Hasselbalch equation, wherein reduction of nicotinamides represents an increase in hydrophobicity and resulting hydrophobic-induced pKa shifts become the basis for understanding a primary energy conversion (proton transport) process of mitochondria. © 1998 John Wiley & Sons, Inc.
