    Notes: Abstract In order to identify regulatory elements that direct widespread in vivo expression of a linked gene, we have examined one of the human aldolase A alternative promoters, the ubiquitous pH promoter, which is active in most foetal and adult tissues. We have used the pH promoter region to drive expression of an heterologous CAT reporter gene in transgenic mice. We show that a short 820 bp pH promoter fragment is able to confer a ubiquitous and reproducible activity pattern on the CAT reporter gene in most of the transgenic lines analysed, with a particularly high level of expression in adult skeletal muscle. Activity of this transgene was detected from early embryonic stages. Therefore, this pH promoter region appears to be a powerful tool to direct ubiquitous and early expression of a transgene in vivo. Deletion analysis revealed that: (i) the region between −651 and −369 bp relative to the pH promoter transcription start site includes DNA elements capable of overriding effects of the surrounding chromatin at the integration site, (ii) the region between −285 and −211 bp is involved in pH promoter tissue-specific expression pattern in skeletal muscle and/or nervous tissues, (iii) the region located between −211 and −108 bp is necessary for its ubiquitous and muscle-pre dominant activity and (iv) the most proximal region downstream from −108 bp is still sufficient to confer an activity in brain and lung

    Notes: Abstract A cDNA encoding the multifunctional cytochrome P450, CYP71E1, involved in the biosynthesis of the cyanogenic glucoside dhurrin from Sorghum bicolor (L.) Moench was isolated. A PCR approach based on three consensus sequences of A-type cytochromes P450 – (V/I)KEX(L/F)R, FXPERF, and PFGXGRRXCXG – was applied. Three novel cytochromes P450 (CYP71E1, CYP98, and CYP99) in addition to a PCR fragment encoding sorghum cinnamic acid 4-hydroxylase were obtained. Reconstitution experiments with recombinant CYP71E1 heterologously expressed in Escherichia coli and sorghum NADPH–cytochrome P450–reductase in L-α-dilaurylphosphatidyl choline micelles identified CYP71E1 as the cytochrome P450 that catalyses the conversion of p-hydroxyphenylacetaldoxime to p-hydroxymandelonitrile in dhurrin biosynthesis. In accordance to the proposed pathway for dhurrin biosynthesis CYP71E1 catalyses the dehydration of the oxime to the corresponding nitrile, followed by a C-hydroxylation of the nitrile to produce p-hydroxymandelonitrile. In vivo administration of oxime to E. coli cells results in the accumulation of the nitrile, which indicates that the flavodoxin/flavodoxin reductase system in E. coli is only able to support CYP71E1 in the dehydration reaction, and not in the subsequent C-hydroxylation reaction. CYP79 catalyses the conversion of tyrosine to p-hydroxyphenylacetaldoxime, the first committed step in the biosynthesis of the cyanogenic glucoside dhurrin. Reconstitution of both CYP79 and CYP71E1 in combination with sorghum NADPH-cytochrome P450–reductase resulted in the conversion of tyrosine to p-hydroxymandelonitrile, i.e. the membranous part of the biosynthetic pathway of the cyanogenic glucoside dhurrin. Isolation of the cDNA for CYP71E1 together with the previously isolated cDNA for CYP79 provide important tools necessary for tissue-specific regulation of cyanogenic glucoside levels in plants to optimize food safety and pest resistance.

    Notes: Vigna unguiculata L. (Walp.)] crop residues could be used to reduce fertilizer N inputs with fall broccoli [Brassica oleracea L. (Italica Group)]. Studies in 1992, 1993, and 1994 involved broccoli preceded by cowpeas, with one or without factorial combinations of preplant N and sidedress N from urea on the broccoli. Control broccoli plants were grown on fallowed ground and supplied with 168 kg ha–1 of N from urea. Cowpea crop residues alone did not provide sufficient N for optimum production of marketable broccoli. Marketable broccoli yields sometimes showed no significant differences between controls and cowpea residue treatments receiving only 84 kg ha–1 of fertilizer N. However, there was a tendency for increased broccoli transplant mortality in cowpea residue plots. A fourth study in 1994 indicated a negative main effect of cowpeas on broccoli stand establishment, and a complex interaction of cowpeas and trifluralin treatments on broccoli yield. Petiole N concentrations of broccoli plants with reduced fertilizer N inputs which succeeded cowpeas usually were lower than those of control plants. Soil nitrate levels at 15- to 30-cm soil depths at the time of broccoli planting were consistent indicators of apparent nitrate utilization by microbial decomposition of cowpea crop residues. We were unable to demonstrate a cowpea-broccoli succession cropping system which consistently reduced fertilizer N inputs without some type of adverse effect on the broccoli crop. Therefore, the use of cowpea crop residues to reduce fertilizer N inputs with fall broccoli is not recommended.

    Notes: Abstract Phosphate (P) uptake experiments were conducted to determine the routes of P entry into nitrogen-fixing common bean root nodules and the availability of this P to the bacteroids within the nodules. Nodulated common bean plants were cultured for 30 days in a hydroponic system that contained either a complete nutrient solution (+P) or a nutrient solution where P was not added (−P). The plant roots were then exposed to [32P]-phosphate-labeled +P nutrient solution and P accumulation in bacteroids and nodules was measured. For labeling, plants were suspended in the nutrient solution so that the root system was either fully or partially submerged. Because bean nodules are heavily clustered in the root crown, most of the nodules in the partial submergence treatments were not in contact with the nutrient solution and we could study nodules that had not been directly exposed to 32P. Regardless of P treatment of the host plant, P accumulation in bacteroids was significantly greater and more rapid when the nodule surface was in contact with the labeled nutrient solution, suggesting that nodules can import P from the solution. P entry through the nodule surface was verified in experiments where 32P-labeled nutrient solution was applied to the nodule surface only. In full submergence treatments, 32P accumulation in bacteroids of −P plants was significantly greater than in bacteroids of +P plants. As measured by total leaf label, uptake and translocation of P to the shoot was greater in −P plants and still greater in partial submergence treatments where most of the nodules were not in contact with the labeled nutrient solution. Results of these experiments suggest that, in addition to P acquisition via the vascular system, P flow to symbiotic tissues and bacteroids can originate from the soil solution immediately surrounding the nodule. This was particularly evident in nodules of plants that had been limited for P.
