    Notes: Variability in the response to salinity within Chloris gayana (Rhodes grass) germplasm was evaluated under field conditions, and random amplified polymorphic DNA (RAPD) markers were used to assess genetic relatedness among cultivars/accessions. RAPD analysis showed a clustering of cultivars of known relatedness: cv. Pioneer and accessions Local and Trancas (derived from an old Pioneer pasture established in saline soil) belonged to the same cluster, Katambora to another and tetraploid Boma and Callide could be further separated, Boma belonging to a fourth, distant cluster. Field experiments were laid out in two types of plots: control [with electrical conductivity of the saturation extract (EC) = 3·64 dS m−1] and saline (EC=13·10 dS m−1) and two experiments were carried out: one to evaluate the effects of salinity on emergence and establishment, and the other, with a uniform number of plants per plot, to evaluate yield under saline conditions. All cultivars/accessions had salinity-associated decreases in dry-matter (DM) production during the establishment phase. After this stage, 1-year DM yield was similar in all cultivars within each salinity level and production in the saline plots was significantly lower than in controls only in cv. Callide and accession Trancas. Second-year production in the non-saline plots increased by 30% on average over the previous year, whereas an average 40% reduction was observed in the saline plots. Thus, salinity had a negative effect on Rhodes grass establishment and persistence. The cultivars could not be ranked unequivocally by production under saline conditions, but tetraploids Boma and Callide may be said to be less tolerant than the rest on the basis of an increased proportion of dead leaves and decreased number of stolons observed in the saline plot.

    Notes: Barley (Hordeum vulgare L.) exposed to low temperature increases its freezing tolerance. This increase has been associated with several metabolic changes caused by low temperature, including expression of dehydrins (DHN), a family of proteins induced by dehydration and cold acclimation. DHNs play an undetermined role in dehydration responses during freezing. We have studied the accumulation of an 80-kDa DHN-like protein (P-80) in barley under cold acclimation 6/4°C (day/night), postulating that it is localized in tissues where primary ice nucleation occurs. P-80 was absent in nonacclimated plants and was detectable after 48 h of cold acclimation, reaching a stable level after 6 days. P-80 decreased when plants were returned to 20–25°C. Drought, ABA and high temperature did not increase the levels of P-80, suggesting that its expression could be specifically regulated by cold. Immunolocalization by tissue printing and fresh cross sections of leaves showed the protein to be associated with vascular tissues and epidermis. The localization of P-80 is consistent with our hypothesis because vascular tissue and the epidermis are preferential ice nucleation zones during the onset of freezing. The differential accumulation of P-80 may have an adaptive value by participating in tolerance mechanisms during freeze-induced dehydration.

    Notes: The main objective of this study was to analyse differences and similarities in morphometric characteristics among specimens of the same year class of Acipenser naccarii (Bonaparte, 1836), comparing fish raised either in Italy or Spain. All specimens were Fl of captive broodstock in Italy.Fifteen biometric variables were measured, and three derived indices were calculated, on 30 individuals of 3+ year class from Lombardy (Italy) and 90 individuals of 1+, 2+ and 3+ year class from Riofrio (Granada, Spain). A stepwise discriminate analysis (SDA) was carried out, and the results were contrasted using a non-hierarchical cluster analysis. The two groups proved to be similar, so a principal component analysis and a simple regression analysis were performed on all individuals, taking total length (Tl) as the independent variable and all the biometric variables as dependent.The study revealed that although the Soljan index (i.e. ratio of mouth width to snout length) is related to Tl, and is a valid taxonomic characteristic, the CA index (i.e. relative position of the barbels) and FB index (i.e. ratio of snout length to width) are also valid characteristics for individuals longer than 57 cm, being independent of Tl.

    Notes: Human rhinoviruses, the major cause of mild recurrent infections of the upper respiratory tract, are small icosahedral particles. Over 100 different serotypes have been identified. The majority (91 serotypes) use intercellular adhesion molecule 1 as the cell-attachment site; ten serotypes (the minor group) bind to members of the low-density lipoprotein receptor. Three different crystal forms of the minor-group human rhinovirus serotype 2 (HRV2) were obtained by the hanging-drop vapour-diffusion technique using ammonium sulfate and sodium/potassium phosphate as precipitants. Monoclinic crystals, space group P21, diffracted at least to 2.8 Å resolution, and two complete virus particles were located in the crystal asymmetric unit. A second type of crystals had a compact cubic like morphology and diffracted beyond 2.5 Å resolution. These crystals belong to a primitive orthorhombic space group, with unit-cell parameters a = 309.3, b = 353.5, c = 759.6 Å, and contain one virus particle in the asymmetric unit. A third type of crystals, with a prismatic shape and belonging to space group I222, was also obtained under similar crystallization conditions. These latter crystals, with unit-cell parameters a = 308.7, b = 352.2, c = 380.5 Å, diffracted to high resolution (beyond 1.8 Å) and contained 15 protomers per asymmetric unit; this requires that three perpendicular crystal twofold axes coincide with three of the viral particle's dyad axes.

    Notes: : Determination of the boundary conditions for modeling ground water flow is a critical point especially in regional models. Normally the regional models require model areas that are greater than the given area of interest. This work focuses on the prediction of hydraulic heads in regional models using flux boundary conditions. The model uses flux boundary conditions that were estimated using a radial flow analog and Darcy's law. The regional model that is presented uses no parameter identification (inverse estimation) procedures. In the present work, the Houston area was used. The simulation of the hydrological conditions of the Chicot and Evangeline Aquifers that underlie the Houston area were made using the available information about the geological profile in the Houston region and the current information about the existing production wells. The regional model works as a forward problem. The system parameters such as hydraulic conductivity, specific storage, and hydrological stresses were specified, and the model predicts the hydraulic head. Actual data from piezometers operated by the U.S. Geological Survey (USGS) in many places throughout Houston were used as initial conditions. Some piezometric head data were generated using the regional variable theory called kriging to supply head estimates in areas where data were unavailable. The Modular Three Dimensional Finite Difference Groundwater Flow Model developed by the USGS was used to predict the hydraulic heads. The predicted ground water heads are compared to the actual data. The results show that the model performs well for locations where data were available.

    Notes: Abstract A site-directed mutagenesis method was designed and used to create Cry1Ab mutant proteins in two of the five highly conserved blocks present in the Cry protein family. Region 1 comprises the central α-helix 5 of domain I and has been implicated in the pore formation activity of the toxin. Substitution of arginine by serine at position 173 (R173S) affects neither structural integrity nor toxicity. Region 2 comprises the major part of the domain I/domain II interface, characterized by the presence of numerous hydrogen bonds and electrostatic interactions. Mutations in the salt bridge formed by aspartic acid 242 and arginine 265 (D242N, D242C, R265C, and D242C/R265C) resulted in structurally unstable mutant proteins as is shown by their increased protease sensitivity and lack of biological activity.

    Notes: Abstract. 3H-IAA transport in excised sections of carnation cuttings was studied by using two receiver systems for recovery of transported radioactivity: agar blocks (A) and wells containing a buffer solution (B). When receivers were periodically renewed, transport continued for up to 8 h and ceased before 24 h. If receivers were not renewed, IAA transport decreased drastically due to immobilization in the base of the sections. TIBA was as effective as NPA in inhibiting the basipetal transport irrespective of the application site (the basal or the apical side of sections). The polarity of IAA transport was determined by measuring the polar ratio (basipetal/acropetal) and the inhibition caused by TIBA or NPA. The polar ratio varied with receiver, whereas the inhibition by TIBA or NPA was similar. Distribution of immobilized radioactivity along the sections after a transport period of 24 h showed that the application of TIBA to the apical side or NPA to the basal side of sections, increased the radioactivity in zones further from the application site, which agrees with a basipetal and acropetal movement of TIBA and NPA, respectively. The existence of a slow acropetal movement of the inhibitor was confirmed by using 3H-NPA. From the results obtained, a methodological approach is proposed to measure the variations in polar auxin transport. This method was used to investigate whether the variations in rooting observed during the cold storage of cuttings might be related to changes in polar auxin transport. As the storage period increased, a decrease in intensity and polarity of auxin transport occurred, which was accompanied by a delay in the formation and growth of adventitious roots, confirming the involvement of polar auxin transport in supplying the auxin for rooting.
