    Notizen: Abstract Ingredient substitutions for agar or carrageenan in artificial diets for lepidopterous larvae were evaluated. A sodium polyacrylamide polyacrylate gelling agent, Water-Lock G-400, was compared with agar for solidifying diets of the cabbage looper, Trichoplusia ni (Hübner), omnivorous looper, Sabulodes aegrotata (Gueneé), and the western avocado leafroller, Amorbia cuneana Walsingham. Water-Lock G-400 is considerably less expensive than agar or carrageenan, it does not require heating to gel, and it is made from a dependable supply of agricultural starch. Sabulodes aegrotata pupae from larvae reared on agar were significantly heavier than those reared on Water-Lock G-400. However, the remainder of the variables observed (larval survival, developmental times, fecundity, sex ratio and pupal weights for A. cuneana and T. ni) were all independent of diet. Decreasing the total amount of ingredients in a Water-Lock-G-400 diet produced fewer and smaller T. ni pupae than did an agar diet when the larvae were reared using a mass production protocol. However, these lower values were still above acceptable levels for mass production and reduced the cost of diet production by 65%. Further ingredient substitutions impaired larval development (i.e., replacing pinto beans with soy protein or combinations of soy protein and α-cellulose), while others did not (i.e., excluding pinto beans and reducing water content and replacing pinto beans with α-cellulose or corn cob grits). Moisture control by adding corn cob grits to diets increased the yield of pupae, while removing water from diets increased survivorship and the yield of pupae.

    Notizen: Abstract In this paper we present an application of computational reflection in the programming oftime-dependent systems. A time-dependent system performs its tasks according to timing specifications specified within the system or imposed from outside the system. Reflective techniques can be applied to programming time-dependent systems because (1) some application programs require the introduction of a new language construct for specifying timing requirements and (2) different applications may require domain-specific scheduling algorithms. To allow a programmer to add or modify language constructs or scheduling algorithms, however, a clear reflective architecture and program interfaces must be provided. This paper proposes a concurrent object-based reflective architecture (R 2 architecture) for time-dependent computing. This architecture is based on anindividual reflection scheme and introduces new meta-level objects (real-time meta objects) that are responsible for time-dependent capabilities. An alarm-clock object and a scheduler object are introduced, and message protocols between them and real-time meta objects are defined. We implemented this architecture on ABCL/R2 and created the Sampled Sound Player program as an application. With this application we provided three different scheduler objects and measured the impact of different scheduling algorithms on sound playback. The measured results show that a scheduler with more complex computations at the meta level exhibited less scheduling overhead, thus was capable of better sound playback. The other example, Time-dependent Graceful Degradation Scheme, demonstrates the programming of functionality degradation triggered by failure to satisfy timing specifications.

    Notizen: Abstract A recently proposed theory for diffusion-limited aggregation (DLA), which models this system as a random branched growth process, is reviewed. Like DLA, this process is stochastic, and ensemble averaging is needed in order to define multifractal dimensions. In an earlier work by Halsey and Leibig, annealed average dimensions were computed for this model. In this paper, we compute the quenched average dimensions, which are expected to apply to typical members of the ensemble. We develop a perturbative expansion for the average of the logarithm of the multifractal partition function; the leading and subleading divergent terms in this expansion are then resummed to all orders. The result is that in the limit where the number of particlesn→∞, the quenched and annealed dimensions areidentical; however, the attainment of this limit requires enormous values ofn. At smaller, more realistic values ofn, the apparent quenched dimensions differ from the annealed dimensions. We interpret these results to mean that while multifractality as an ensemble property of random branched growth (and hence of DLA) is quite robust, it subtly fails for typical members of the ensemble.

    Notizen: Abstract Vitamin B12 (VB12) is a putative modulator of the human circadian clock, improving entrainability to the 24 h light-dark cycle. The present study was intended to elucidate the mechanism of VB12 action in an animal model. In male rats free-running under constant dim illumination, a single light pulse of 50–1000 lux for 20 min given at circadian time (CT) 20 induced a 0.28 to 1.08 h phase advance and at CT 14 induced a 0.54 to 2.10 h phase delay. A 3 h intracerebroventricular (icv) infusion of 30 nmol VB12 starting 2 h prior to a 20 min 200 lux light pulse significantly amplified phase shifts in comparison with saline-treated or untreated controls. The mean phase advance (1.13 h) was 1.8-fold greater than that of saline-infused controls, whereas the mean phase delay (2.28 h) was 2.9-fold greater. These values were comparable to the maximal phase shifts caused by 1000 lux light pulses in untreated rats. Since the same VB12 treatment alone had failed to induce a phase shift in a previous experiment, these results indicate that VB12 strongly enhanced light pulse-induced phase shifts and thus augmented the entrainability of the circadian clock to light.

Bone morphogenetic proteins (BMPs) are novel growth and differentiation factors that act on mesenchymal stem cells to initiate new bone formation in vivo and promote the growth and differentiation of cells in the osteoblastic lineage. In the present study, we examined the effects of recombinant human osteogenic protein-1 (also known as BMP-7) on the expression of related members of the BMP family using SaOS-2 and U2-OS, two human osteosarcoma cell strains. Evaluation of BMP-2, -4, and -6 mRNA expression indicates that OP-1 stimulated the mRNA levels of BMP-6 in both SaOS-2 cells (threefold) and U2-OS cells (fivefold) after 24 hours of treatment, while decreasing the mRNA levels of BMP-4 in SaOS-2 cells (80%) and BMP-2 and BMP-4 in U2-OS cells by 50% and 72%, respectively. BMP-2 mRNA expression, as examined by Northern blot analysis, was below detectable limits in SaOS-2 cultures. These results demonstrate that OP-1 modulates the mRNA expression of related members of the BMP family, suggesting a possible mode of action of OP-1 on the growth and differentiation of cells in the osteoblastic lineage in vitro.

    Notizen: Abstract  It has been reported that in the chick embryonic retina, N-cadherin first appears at the very early stages and is subsequently substituted by R-cadherin at the middle to late stages of development. To examine the role of R-cadherin in the morphogenesis of chick retinal pigment epithelium (RPE), the distribution of this adhesion molecule was studied by immunofluorescence cytochemistry and immunoelectron microscopy from embryonic day (E) 6 to hatching. R-cadherin immunoreactivity was detected at E6, and was strongest at E12–13. During these stages, R-cadherin was expressed uniformly on the lateral plasma membranes of RPE cells in contact with each other. Thereafter, R-cadherin immunoreactivity was markedly decreased, with intense immunoreactivity restricted to zonulae adherentes in latero-apical regions at E16. R-cadherin immunoreactivity was no longer detectable in the newly hatched chick RPE, even though morphologically well developed zonulae adherentes were present in latero-apical regions. No immunoreactivity was detected on the apical side facing the neural retina or on the basal side facing the basal lamina at any stage of development. These findings indicate that R-cadherin plays an important role as a major cadherin subtype in the morphogenesis of chick embryo RPE, and is involved initially in non-specific cell-cell adhesions, and subsequently in the formation and maintenance of developing zonulae adherentes.

    Notizen: Abstract  CD1 and TL were once thought to be genetic homologues because of their thymus-specific expression. We investigated their equivalents in the rat to clarify whether their structure and pattern of expression are conserved in rodents. Two rat class Ib genes, containing 3′ sequences very similar to mouse TL, were identified and designated RT1.P. Neither of them, however, can encode ordinary class I molecules due to the accumulation of harmful mutations in the 5′ regions that are unique to RT1.P, while the 3′TL-like regions still retain protein-coding capacity. Comparison of the structural organization of three types of TL family genes, which include mouse T3/T18-encoding TL antigens, mouse T1/T16, and rat RT1.P1/P2 pseudogenes, revealed the presence of a clear demarcation between the type-specific and TL-specific sequences at intron 3. This finding suggests that recombination plays an important role in creating the TL family genes in rodents. Characteristic features of TL, such as a low level of polymorphism and linkage to the major histocompatibility complex, were also observed in the rat. On the other hand, rat CD1 molecules were expressed at a high level on the surface of thymocytes. Absence of authentic TL antigens and thymic expression of CD1d molecules in the rat suggest the plasticity and conservation of class Ib genes in rodent evolution. Functions of TL may be substituted with CD1 or other class Ib molecules expressed by rat thymus.

    Notizen: Abstract Objectives: The present study was conducted to compare pharmacokinetic behaviors of nicardipine enantiomers given in different doses with different formulations of racemic nicardipine in healthy volunteers. Methods: One or two 20-mg racemic nicardipine tablets, and a 40-mg sustained-release capsule of nicardipine were administered to eight healthy volunteers in a cross-over fashion and pharmacokinetic parameters were evaluated. Enantiomer concentrations were determined by GC-MS combined with chiral stationary phase HPLC. Results and conclusions: Serum concentration of (+)-nicardipine was approximately 2–3 times higher than that of (−)-nicardipine in 20- and 40-mg doses of conventional formulations and a non-linear increase in bioavailability with dose was demonstrated. The value for AUC of (+)-nicardipine was approximately 2.3–2.8 times greater than that of the (−)-nicardipine (P 〈 0.05) when 20 and 40 mg racemic nicardipine were administered in a conventional preparation. Relative bioavailability of the sustained-release preparation vs the conventional preparation was 28% and 44% for (+)- and (−)-nicardipine, respectively, for the 40-mg dose.
