    Notes: Five types of sensilla are situated on the apical area of the labial and maxillary palpi and galea of Cicidela sexguttata. Large, conical, and peg-like sensilla are in rows on the central region of each palpus. These sensilla have a hollow cuticular peg, with an apical pore and multi-innervation. This central region of palpal sensilla is surrounded by campaniform sensilla that are disc-shaped and small conical peg sensilla. A similar type of conical sensillum as the found in the palpal central region is situated around the periphery of the palpal apex and apex of the galea. This conical peg sensillum is located in a shallow depression and is structurally similar to the other peg sensilla, but it has a mechanoreceptor neuron attached to the cuticular base of the sensillum. A long, single, trichoid sensillum is situated in the center of the galea and is hollow, thick-walled, porous, and multi-innervated. The apices of the palpi and galea have a large number of dermal gland openings that actively secrete a substance during the feeding process of the tiger beetle. © 1995 Wiley-Liss, Inc.

    Notes: The kinetics of grafting a basic monomer, 2-(dimethylamino)-ethyl methacrylate (DMAEMA) to hydrocarbon substrates have been investigated. These systems were chosen as models for the grafting of a homopolymerizable monomer to polyolefins such as polyethylene. The reactions with squalane and n-eicosane were initiated by an organic peroxide, 2,5-dimethyl 2,5 dit-butylperoxy)-3-hexyne; grafting yields become significant at high reaction temperatures and low monomer concentrations. In squalane, the order of reaction with respect to monomer increased from about 1.1 for 0.22-0.44M DMAEMA to almost 2 at 0.69M DMAEMA; the order with respect to initiator was 0.56. The overall activation energy in the 130-160°C temperature range was, however, surprisingly low, 42±5 kJ mol-1. When analytical data were used to separate the overall rate into those for grafting and homopolymerization, different kinetic paths were observed for the competing reactions. These results are interpreted in terms of two different mechanisms; intramolecular chain transfer plays an important role in grafting, while depropagation becomes a major factor in homopolymerization at temperatures above 150°C. © 1995 John Wiley & Sons, Inc.

    Notes: The first large-scale production of 7-aminocephalosporanic acid (7ACA) from cephalosporin C (CPC) using a wholly enzymatic synthesis method is reported here. We produced 7ACA from CPC in as high a molar yield as 85% using the immobilized enzymes D-amino acid oxidase (D-AOD) and glutaryl-7-ACA acylase (GL-acylase). In the first reactor, CPC is converted to keto-adipyl-7-aminocephalosporanic acid (keto-7ACA) using an immobilized D-AOD isolated from a yeast, Trigonopsis variabilis. The keto-7ACA is then spontaneously converted to glutaryl-7-aminocephalosporanic acid (GL-7ACA) via a chemical reaction with hydrogen peroxide. The hydrogen peroxide is also a product of the D-AOD reaction. Near quantitative conversion of the keto-7ACA to GL-7ACA was observed. The second reactor converts GL-7ACA to 7ACA using an immobilized GL-acylase, which was isolated from a reconbinant Escherichia coli. The final 7ACA crystalline product is a high quality product. The reactions are conducted under very mild aqueous conditions: pH 8.0 and 20° to 25°C. The production of desacetyl side products is minimal. This process is currently being implemented on an industrial scale to produce 7ACA. © 1995 John Wiley & Sons, Inc.

    Notes: Epithelial ovarian cancer is a heterogenous disease. Epidemiologic studies have identified risk factors for this disease including advanced age, nulliparity, history of infertility, early age at menarche, late age at menopause, and perhaps ovulation induction. Cohort selection that includes women who have potential precursor lesions and alterations of select biomarkers may prove useful in the design of chemoprevention trials of epithelial ovarian cancer. Nuclear morphometry, specific genetic alterations, and markers of proliferation and differentiation may be useful biomarker to monitor the efficacy of specific interventions.

    Notes: The potential for encapsulation of arsenic waste by combination with two commodity polymers was evaluated. Initial studies employed nontoxic substitutes, these being gypsum and calcium carbonate, with host polymers being predominately a recycled grade of polyethylene (PE) and a synthetic elastomer. The latter was used at low processing temperatures with rather volatile arsenic compounds. The gypsum caused processing difficulties due to evolved water, but the CaCO3 was able to be readily combined at high volumes. Arsenic trioxide was able to be incorporated at modest levels in PE and was less successfully combined with the rubber. However, the higher processing temperatures needed for PE caused the As2O3 to sublime. Less volatile calcium arsenite, readily prepared from As2O3, was able to be mixed at high volume ratios with the rubber, although processing with PE was less successful. These results may be used in conjunction with other methods for stabilization and would be applicable to other forms of medium- to high-level waste. © 1995 John Wiley & Sons, Inc.

    Notes: Women at risk of uterine cancer include those with one or more of the following characteristics: obesity, nulliparity, late menopause, diabetes mellitus, prolonged unopposed estrogen use, and tamoxifen therapy. Risk is additionally increased by the presence of endometrial hyperplasia. The incorporation of biomarkers into the selection criteria of cohort groups at risk for developing endometrial cancer offers an innovative approach to the clinical design of chemoprevention trials of endometrial adenocarcinoma. Biomarkers that may be useful in cohort selection include nuclear morphometry, specific genetic abnormalities, and markers of proliferation and differentiation.

    Notes: A black charcoal-like material having cation exchange and adsorption properties was prepared by the controlled pyrolysis of starch in the presence of a commercial phytic acid solution. Resins with binding capacities of 0.7-5.7 meq/g of calcium were prepared by varying the phytic acid to starch ratio, the temperature, or the duration of heating of the reaction mixture. SEM photomicrographs of some of these new materials showed that they are composed of particles similar in size and shape to the starting starch granules.     Notes: A pneumatically assisted electrospray liquid chromatography/mass spectrometry interface has been modified for use with open-tubular supercritical fluid chromatography (SFC). The modified interface houses the SFC flow restrictor within the spray device. A sheath-flow liquid of methanol or methanol/water containing ammonium acetate flows coaxially to the restrictor. The addition of the sheath-flow liquid results in the ionization of analytes known to be in a neutral charge state during the chromatographic separation. The sheath-flow liquid also allows for the independent operation of the chromatograph and the mass spectrometer. Both could be operated under optimum conditions. The operation of the interface was optimized using a solution of poly(dimethylsiloxane) in CO2 introduced by continuous infusion. We also report the application of the modified instrument to samples containing amines and propoxylated alcohols.

    Notes: A gas chromatographic/mass spectrometric/mass spectrometric method was developed for the determination of 5-methoxyflavone in plasma. Optimized detection schemes for 5-methoxyflavone readily allow a limit of quantification of 1 ng ml-1 in rat or dog plasma (1.0 ml sample volume). The complete analysis time (injection to injection) is less than 9 min and allows for rapid automated sample throughput. Additional advantages include a high degree of specifity and good accuracy and precision. This type of rapid tandem mass spectrometric assay has been found to be particularly useful in pharmaceutical discovery, since an assay for one compound can be quickly developed, utilized and then modified to determine analogs of similar structure. Application of this methodology to pilot preclinical pharmacokinetic studies of the drug indicated that it is rapidly absorbed and cleared, with an apparent terminal half-life of about 15 min in dogs. Bioavailability in the two dogs tested ranged 33 to 72%, but was about 25% in rats.
