    Notes: The desire to replace the amide backbone of renin inhibitors with a new scaffold led us to explore vinylogous amides (enaminones). An initial attempt proved unsuccessful, a result explained after the fact via docking experiments. Based on this lesson, we designed a different vinylogous amide scaffold which incorportated one or more pyrrolinone rings into the backbone. Three of the four compounds gave IC50s in the 0.6 to 18 μM range. These compounds did not inhibit HIV-1 protease. Taken together, the results reported herein provide insights into the role of hydrogen bonding and steric interactions for binding to renin. © 1994 John Wiley & Sons, Inc.

    Notes: A complex of lymphoepithelial organs, the “anal tonsils,” is a consistent structure in the anal canal of the bottlenose dolphin, Tursiops truncatus. This complex occurs as a circumferential cluster of discrete tonsil like aggregations of lymphoid tissues, together with epithelial ducts (“crypts”) and occasional mucus secretory units in the extreme lower portion of the intestinal tract. These structures are concentrated in the segment lined by stratified squamous epithelium and extend for a variable distance cephalad from the anal aperture. The tonsils appear to be most active, judged by the amount of lymphoid tissue present, in young animals. Depletion of lymphocytes and cystic enlargement of the crypts, probably representing functional as well as morphological involution, is a consistent feature of older animals. © 1995 Wiley-Liss, Inc.

    Notes: To examine the possible role of kinesin in pigment granule migration in the retinal pigment epithelium (RPE) of teleosts, we investigated the expression and distribution of kinesin heavy chain (KHC) in RPE. Blots of fish RPE lysates probed with two well-characterized antibodies to KHC (H2 and HD) displayed a prominent band at 120 kD. A third KHC antibody (SUK4) recognized a band at 118 suggesting the presence of two KHC isoforms in teleost RPE. Reverse transcriptase-polymerase chain reaction (RT-PCR) of mRNA from RPE using primers homologous to conserved regions of the KHC motor domain resulted in the homologous to conserved regions of the KHC motor domain resulted in the identification of two putative KHC genes (FKIF1 and FKIF5) based on partial amino acid sequences. Previous studies had demonstrated a requirement for microtubules in pigment granule aggregation in RPE. In addition, the reported microtubule polarity orientation in RPE apical projections is consistent with a role for kinesin in pigment granule aggregation. Immunofluorescent localization of KHC in isolated RPE cells using H2 revealed a mottled distribution over the entire cell body, with no detectable selective association with pigment granules, even in cells fixed while aggregating pigment granules. Microinjected KHC antibodies had no effect on pigment granule aggregation or dispersion, although each of the three antibodies has been shown to block kinesin function in other systems. Thus we found no evidence for KHC function in RPE pigment granule aggregation. However, the two KHC isoforms may participate in other microtubule-dependent processes in RPE.

    Notes: Use of saturation-dependent relative mobilities leads to linear flow; however, experiment and theory show that, in the limit of very large viscosity ratio, the flow is not linear but fractal. Generally, fractional flows and relative mobilities depend on both saturation and time. Use of a standard pore-level model of 2-D flow in the limit of infinite capillary number shows that this flow is fractal for large viscosity ratios (M = 10,000) and the sauration and fractional flows agree with the results of our general arguments. For realistic viscosities (M = 3 → 300), our modeling of the unstable flow shows that, although the flows are initially fractal, they become linear on a time scale, τ increasing as τ = τ0M0.17. Once linear, the saturation front advances as x ≈ v0M0.068 t; the factor M0.068 acts as a 2-D Koval factor.

    Notes: There are three major methods for buffer exchange of proteins at industrial scale: size exclusion chromatography (SEC), tangential flow filtration (TFF), and countercurrent dialysis (CCD). In order to determine the optimal technology for a given process, a study was done to compare these technologies on a technological and economic basis. This comparison required that new mathematical models be developed which enable the common features of each unit operation to be directly compared. The new concept of a diavolume equivalent for SEC, defined as the inverse of the fractional loading, was also introduced to aid in this comparison. Variables that were examined for each unit operation included range of buffer exchange, dilution of protein solution, yield, buffer requirements, total operating time, throughput, plant space, capital, raw materials, and labor costs. It was found that TFF and CCD have a greater range of buffer exchange than SEC. TFF also provides the advantage that concentration of the protein can readily be accomplished in the same step. For processes of equal batch size and yield, TFF and CCD also provide a two- to five- fold improvement in each of the remaining variables. The major economic advantage in using TFF and CCD over SEC is the decreased plant size required for manufacturing and thus the longer term use of existing facilities. Situations where SEC (or CCD) would be favored over TFF are when protein denaturation occurs in TFF but does not occur in SEC. © 1995 John Wiley & Sons, Inc.

    Notes: Continuous kinetics of thermal degradation of poly (styrene-allyl alcohol) (PSA) in solution are explored theoretically and experimentally. Thermal degradation experiments for PSA of number-average molecular weight 1,640 in t-butanol solution were conducted in a steady-state flow reactor at 6.8 MPa and 403-473 K. The molecular-weight distributions (MWDs) of the original polymer and its reaction products were measured as a function of residence time by gel permeation chromatography The MWD of the initial PSA is described by a gamma distribution function. Experimental data indicated that the polymer cracks to specific, low-molecular-weight (MW) products and degrades by random chain scission. Results are interpreted by a mathematical model based on the continuous kinetics for specific and random degradation processes. Rate coefficients are determined separately from the dynamic data of the MW moments for the specific products and the MWDs for the reacting polymer. Activation energies obtained are 5.9 - 7.4 kcal/mol for specific degradation processes and 35 kcal/mol for the random degradation process. The detailed MWDs are related to conventional lumped (numberaverage molecular-weight) data.

    Notes: We discuss a two-step superposition method for calculating linear elastic stress intensity factors. The procedure requires the solution to the full cracked problem and the solution to a problem on the same mesh assuming the singularity due to a crack tip in an infinite region. We show that this is equivalent to the well known subtraction of singularity method if the two solutions are characterized by crack tip stress. The advantages of our procedure are that no modifications need to be made to a standard computer program and that once one singular solution is available on a given cracked mesh, solutions with different boundary conditions on the same mesh may be obtained in one step without including any singular crack effects. The mesh required to represent the singular crack tip field may also be studied independently of the complete problem. The additional computational cost of a two-step procedure is minimal since the solution matrix from step one may be reused with a new right-hand side. Numerical experiments using the boundary element method demonstrate the high accuracy and simplicity of the superposition approach.
