    Notes: The structure, genomic organization and transcription of the gene encoding histone H2B in the protozoan parasite Trypanosoma cruzi have been studied. This gene consists of a 746-nucleotlde unit, tandemly repeated at least 18 times in each of two clusters. DNA probes corresponding to histones H2B and H3 hybridized to different chromosomes revealing that the genes coding for these two histones are not physically linked in the genome of T. cruzi. The primary transcription product of the H2B gene is processed by trans-splicing and polyadenylation. Inhibition of DNA synthesis with aphidicolin resulted in the reduction of histone H2B mRNA to undetectable levels in about two hours, suggesting that its abundance is regulated throughout the cell cycle as it occurs in other eukaryotes. in addition, a concomitant inhibition of translation by cycloheximide reverted this effect indicating that de novo protein synthesis is required for RNA instability. Histone mRNA abundance was dependent on the life-cycle stage of T. cruzi: abundant in amastigotes and epimastigotes, the dividing forms in the host cell and the insect vector, respectively, white undetected in trypomastigotes, the parasite's non-dividing life stage.

    Notes: Abstract Mo/Si multilayers are fabricated by electron-beam evaporation in UHV at different temperatures (30° C, 150° C, 200° C) during deposition. After completion their thermal stability is tested by baking them at temperatures (T bak) between 200° C and 800° C in steps of 50° C or 100° C. After each baking step the multilayers are characterized by small angle CuKα-X-ray diffraction. Additionally, the normal incidence soft-X-ray reflectivity for wavelengths between 11 nm and 19 nm is determined after baking at 500° C. Furthermore, the layer structure of the multilayers is investigated by means of Rutherford Backscattering Spectroscopy (RBS) and sputter/Auger Electron Spectroscopy (AES) technique. While the reflectivity turns out to be highest for a deposition temperature of 150° C, the thermal stability of the multilayer increases with deposition temperature. The multilayer deposited at 200° C stands even a 20 min 500° C baking without considerable changes in the reflectivity behaviour.

    Notes: Abstract Five different TiNx coatings have been deposited on steel substrates by reactive magnetron sputtering. A quantitative analysis was carried out in two ways. The coatings were initially analysed by GDOS and after removing the coatings from the substrates and digesting the TiNx powder, nitrogen was quantified by the Kjeldahl technique and titanium by AAS. A comparison of the results shows that AAS and Kjeldahl results correlate very well, whereas GDOS results deviate from these two methods systematically.

    Notes: Summary Background: Prednisone is an active drug in the treatment of multiple myeloma. The optimal dose, frequency, and role of glucocorticoid receptors (GR) in response to prednisone is unknown.Purpose: The purposes of this study were (1) to estimate the response rate of alternate-day high dose prednisone in patients with relapsing and refractory multiple myeloma; (2) to measure the range of GR levels; and (3) to correlate the response of prednisone with GR status.Patients and methods: Between 8/86 and 1/90, 127 patients were entered onto the study with 121 evaluable for response. The number of GR sites/cell was determined on mononuclear cells isolated from pretreatment bone marrow aspirates using a one point GR binding assay. Patients received prednisone 100 mg po qod x 2 weeks, followed by 50 mg po qod x 10 weeks.Results: The overall response rate was 10% (95% CI: 5–15%) with a median survival of 11.8 months. The GR sites/cell ranged from 0–53,212 with a mean of 8,371 sites/cells. Stratification of GR sites into 0–2,500, 2,501–6,000 and 〉 6,000 sites/cells was associated with a response rate of 6%, 27% and 4% respectively (p = 0.009). The median survival of patients in these categories was 8.1, 14.9 and 10.6 months respectively. This was not significant by the logrank test (p = 0.11). Although myeloma patients with intermediate levels of GR sites/cell initially responded more favorably to prednisone, their long-term survival was not significantly improved.Conclusions: Alternate-day high-dose prednisone was well tolerated and may provide palliative benefit for a subset of patients with relapsing and refractory multiple myeloma. The survival of patients on this study was comparable to that reported with other but more toxic doses of glucocorticoids.

    Notes: Abstract Anchihaline habitats occur most frequently in subsiding areas. Typically, they are populated by ancestors of marine origin. These ancestral forms have a much wider distribution in the open sea, thus their hypogean descendents occur in phenetically similar populations on various islands. On the contrary, on rising islands, marine ancestors stranded during the uplift and got isolated in brackish or fresh ground waters, giving rise to phenetic and genetic isolates in very restricted areas. They belong to old genera with a large ditribution (amphi-Atlantic or Tethyan) The stygofaunas of both rising and subsiding areas thus originated in the sea, but contrary to the often uttered suggestions, not in the deep-sea. Phenetic resemblance between deep-sea and anchihaline taxa may indicate common ancestry, but then it must be shallow-water ancestry for both, simply because no deep-sea species survived the two oxygen-crises in the early and mid-Tertiary.
