    Beschreibung: Gamma-ray observations of the nearby starburst galaxies NGC 253 and M82 over the energy range (0.05-10) MeV have been obtained with the Oriented Scintillation Spectrometer Experiment (OSSE) spectrometer on the Compton Gamma-Ray Observatory (CGRO). The priority of these galaxies as OSSE targets had been established on the grounds that the average supernova rate may be high in starbursts as indicated by infrared and radio observations, and at distances of approximately 3 Mpc a significant chance of supernova gamm-ray line detection exists. NGC 253 was detected in continuum emission up to 165 keV with a total significance of 4.4 sigma and an estimated luminosity of 3 x 10(exp 40) ergs/s. The spectrum is best fit by a power law of photon index approximately 2.5. We consider the possible contribution of different emission mechanisms, including inverse Compton scattering, bremsstrahlung, discrete sources, and Type Ia/Ib supernova continuum to the measured flux. No significant continuum flux was observed from M82. A search for the gamma-ray line from the decay of the most abundant radioactive element produced in supernovae (Ni-56 yields Co-56 yields Fe-56) yielded no significant detection: the 3 sigma upper limits to the line fluxes at 0.158, 0.812, 0.847, and 1.238 MeV for both galaxies are obtained.

    Notizen: Vitamin D metabolites appear to regulate chondrocytes and osteoblasts via a combination of genomic and nongenomic mechanisms. Specificity of the nongenomic response to either 1,25-(OH)2D3 or 24, 25-(OH)2D3 may be conferred by the chemical composition of the target membrane and its fluid mosaic structure, by the presence of specific membrane receptors, or by the interaction with classic Vitamin D receptors. Nongenomic effects have been shown to include changes in membrane fluidity, fatty acid acylation and reacylation, arachidonic acid metabolism and prostaglandin production, calcium ion flux, and protein kinaase C activity. Chondrocytes metabolize 25-(OH)D3 to 1,25-(OH)2D3 and 24,25-(OH)2D3; production of these metabolites is regulated by both growth factors and hormones and is dependent on the state of cell maturation. 1,25-(OH)2D3 and 24,25-(OH)2D3 may interact directly with extracellular matix vesicles to regulate their function in the matrix, including protease activity, resulting in matrix modefication and calcification. Isolated matrix vesicles, produced by growth zone chondrocytes, can activate latent transforming growth factor-β when incubated with exogenous 1,25-(OH)2D3. These observations suggest that nongenomic regulation of martix vesicle structure and function may be a mechanism by which mesenchymal cells, like osteoblasts and chndrocytes, may modulate events in the extracellular matrix at sites distant from the cell surace.

    Notizen: Recombinant human haemoglobin A (rHbA) was produced by a leucine-requiring strain of Hansenula polymorpha which had been transformed with an integration vector containing the Saccharomyces cerevisiae LEU2 gene and cDNAs for the expression of α and β globin each driven by the H. polymorpha MOX promoter. After 40 generations in a chemostat it was found that the integrated vector had become amplified in the host strain. In some cases this led to an increase in LEU2 gene dosage, but a loss of globin expression cassettes. In other cases the globin gene dosage also increased. These changes coincided with an increase in rHbA production in the culture, which was reversed when the dilution rate was increased. Isolates from a chemostat culture producing elevated levels of rHbA were grown in fed-batch fermentations, resulting in higher productivities than when inoculated with the parent strain. The rHbA produced was purified and characterized. Oxygen binding studies and electrospray mass spectrometry showed that the rHbA had been processed and assembled correctly, and behaved as a fully functional co-operative tetramer.

    Beschreibung: The bright, peculiar elliptical galaxy Centaurus A (NGC 5128, PKS 1322-427) was one of the first extragalactic radio sources to be optically identified (Bolton et al. 1949). At a distance of 4 Mpc, Centaurus A is the closest active radio galaxy and affords the highest linear imaging resolution (1 mas approximately equal to 0.02 pc) and hence the best prospects for studying an active nucleus close to the central radio source. We present the results of multi-epoch, 8.4-GHz, very long baseline interferometry (VLBI), imaging observations of the nucleus made over the past three years. The nucleus possesses a core-jet structure where the inner portion of the jet shows apparent linear motion with a velocity substantially less than the speed of light.

    Beschreibung: Snow feedbacks produced by 14 atmospheric general circulation models have been analyzed through idealized numerical experiments. Included in the analysis is an investigation of the surface energy budgets of the models. Negative or weak positive snow feedbacks occurred in some of the models, while others produced strong positive snow feedbacks. These feedbacks are due not only to melting snow, but also to increases in boundary temperature, changes in air temperature, changes in water vapor, and changes in cloudiness. As a result, the net response of each model is quite complex. We analyze in detail the responses of one model with a strong positive snow feedback and another with a weak negative snow feedback. Some of the models include a temperature dependence of the snow albedo, and this has significantly affected the results.

    Beschreibung: Acetone (CH3COCH3) was found to be the dominant nonmethane organic species present in the atmosphere sampled primarily over eastern Canada (0-6 km, 35 deg-65 deg N) during ABLE3B (July to August 1990). A concentration range of 357 to 2310 ppt (= 10(exp -12) v/v) with a mean value of 1140 +/- 413 ppt was measured. Under extremely clean conditions, generally involving Arctic flows, lowest (background) mixing ratios of 550 +/- 100 ppt were present in much of the troposphere studied. Correlations between atmospheric mixing ratios of acetone and select species such as C2H2, CO, C3H8, C2Cl4 and isoprene provided important clues to its possible sources and to the causes of its atmospheric variability. Biomass burning as a source of acetone has been identified for the first time. By using atmospheric data and three-dimensional photochemical models, a global acetone source of 40-60 Tg (= 10(exp 12) g)/yr is estimated to be present. Secondary formation from the atmospheric oxidation of precursor hydrocarbons (principally propane, isobutane, and isobutene) provides the single largest source (51%). The remainder is attributable to biomass burning (26%), direct biogenic emissions (21%), and primary anthropogenic emissions (3%). Atmospheric removal of acetone is estimated to be due to photolysis (64%), reaction with OH radicals (24%), and deposition (12%). Model calculations also suggest that acetone photolysis contributed significantly to PAN formation (100-200 ppt) in the middle and upper troposphere of the sampled region and may be important globally. While the source-sink equation appears to be roughly balanced, much more atmospheric and source data, especially from the southern hemisphere, are needed to reliably quantify the atmospheric budget of acetone.

    Beschreibung: We present the first Extreme Ultraviolet Explorer (EUVE) satellite observation of the Jupiter system, obtained during the 2 day period 1993 March 30 through April 1, which shows a rich emission-line spectrum from the Io plasma torus spanning wavelengths 370 to 735 A. The emission features correspond primarily to known multiplets of oxygen and sulfur ions, but a blended feature near 372 A is a plausible Na II transition. The summed detected energy flux of (7.2 +/- 0.2) x 10(exp -11) ergs/sq cm(s) corresponds to a radiated power of approximately equal to 4 x 10(exp 11) W in this spectral range. All ansa emissions show a distinct dawn-dusk brightness asymmetry and the measured dusk/dawn ratio of the bright S III lambda-680 feature is 2.3 +/- 0.3, significantly larger than the ratio measured by the Voyager spacecraft ultraviolet (UV) instruments. A preliminary estimate of ion partitioning indicates that the oxygen/sulfur ion ratio is approximately equal to 2, compared to the value approximately equal to 1.3 measured by Voyager, and that (Na(+))/(e) greater than 0.01.

    Beschreibung: We have used the Faint Object Spectrograph of the Hubble Space Telescope to observe interstellar linear polarization from 1300 to 3300 A in two stars with well-studied interstellar polarization at visible wavelenths. The wavelength dependence of linear polarization declines smoothly with decreasing wavelength and is devoid of structure associated with the prominent 2175 A absorption bump in the interstellar extinction curve. The data for one star (HD 161056) are consistent with an extrapolation based on the Serkowski formula of a fit to the ground-based polariztion; the other star (HD 7252) shows excess (super-Serkowski) polarization relative to the extrapolation. Out of a total of 10 stars now studied by means of spectropolarimetry in the satellite ultraviolet, including eight obseved with the Wisconsin Ultraviolet Photopolarimeter, five (those of longest lambda (sub max)) show Serkowski behavior, and four others show super-Serkowski behavior; only one (HD 197770) shows evidence for polarization associated with the 2175 A bump. These results place important constraints on the nature of the bump feature.

    Beschreibung: Aircraft measurements of key reactive nitrogen species (NO, NO2, HNO3, PAN, PPN, NO3(-), NO(y)), C1 to C6 hydrocarbons, acetone, O3, chemical tracers (C2Cl4, CO), and important meteorological parameters were performed over eastern Canada during July to August 1990 at altitudes between 0 and 6 km as part of an Arctic Boundary Layer Expedition (ABLE3B). In the free troposphere, PAN was found to be the single most abundant reactive nitrogen species constituting a major fraction of NO(y) and was significantly more abundant than NO(x) and HNO3. PAN and O3 were well correlated both in their fine and gross structures. Compared to data previously collected in the Arctic/subarctic atmosphere over Alaska (ABLE3A), the lower troposphere (0-4 km) over eastern Canada was found to contain larger reactive nitrogen and anthropogenic tracer concentrations. At higher altitudes (4-6 km) the atmospheric composition was in many ways similar to what was seen over Alaska and supports the view that a large-scale reservoir of PAN (and NO(y)) is present in the upper troposphere over the entire Arctic/subarctic region. The reactive nitrogen budget based on missions conducted from the North Bay site (missions 2-10) showed a small shortfall, whereas the budget for data collected from the Goose Bay operation (missions 11-19) showed essential balance. It is calculated that 15-20 ppt of the observed NO(x) may find its source from the available PAN reservoir. Meteorological considerations as well as relationships between reactive nitrogen and tracer species suggest that the atmosphere over eastern Canada during summer is greatly influenced by forest fires and transported industrial pollution.

    Beschreibung: As a step toward developing poliovirus as a vaccine vector, poliovirus recombinants were constructed by fusing exogenous peptides (up to 400 amino acids) and an artificial cleavage site for viral protease 3Cpro to the amino terminus of the viral polyprotein. Viral replication proceeded normally. An extended polyprotein was produced in infected cells and proteolytically processed into the complete array of viral proteins plus the foreign peptide, which was excluded from mature virions. The recombinants retained exogenous sequences through successive rounds of replication in culture and in vivo.
