An active recombinant preparation of the carboxy-terminal ribonuclease H (RNase H) domain of HIV-I reverse transcriptase has produced crystals of several different forms, including a trigonal prism form (P31; a = b = 52.03, c = 113.9 Å with two molecules per asymmetric unit) and a hexagonal tablet form (P6222 or P6422; a = b = 93.5, c = 74.1 Å with one molecule per asymmetric unit). The former appears to be isomorphous with crystals of a similar, but inactive, version of the enzyme that was used for a prior crystal structure determination [Davies, Hostomska, Hostomsky, Jordan & Matthews (1991). Science, 252, 88–95]. We have also obtained a structure solution for this crystal form and have refined it with 2.8 Å resolution data (R = 0.216). We report here details of our crystallization studies and some initial structural results that verify that the preparation of active HIV-1 RNase H yields a protein that is not just enzymatically, but also structurally, distinguishable from the inactive form. Evidence suggests that region 538–542, which may be involved in the catalytic site and which is disordered in both molecules in the prior structure determination, is ordered in the crystal structure of the active enzyme, although the ordering may include more than one conformation for this loop. It should also be noted that, in the crystal structure of the trigonal form, RNase H monomers associate to form noncrystallographic twofold-symmetric dimers by fusing five-stranded mixed β sheets into a single ten-stranded dimerwide sheet, an assembly that was not remarked upon by previous investigators.

    Notizen: Abstract This paper examines the directionality of tuberous electroreceptor responses and relates them to a polarity bias seen for passive electrolocation by electric fish (Hypopomus). We recorded from Burst Duration Coders (BDCs) while stimulating with 1 kHz single period sine waves with electric fields oriented horizontally in different directions. Electroreceptors have figure-8 directional sensitivity profiles with two, usually unequal lobes of sensitivity separated by 180°. For most units the larger lobe points inward, while for a few, the lobes are symmetrical or the larger lobe points outward. The differences correlate with differences in frequency tuning of the receptors. We can alter, and even reverse, the directional asymmetry of a single unit by changing the frequency of the stimulus. Two general response profiles result, with two corresponding classes of tuning curves. The degree of asymmetry varies with position on the body surface. The asymmetries and the effects of stimulus frequency and of tuning can be modeled with a linear/non-linear/linear cascade filter. The behavioral preference for approaching the head end ( + ) of an electrode is difficult to understand in light of the asymmetry of responses we report for amplitude-coding BDCs but can be understood by reference to the time-coding Pulse Marker (PM) receptors.

    Notizen: Abstract This paper is an electrophysiological study of the directionality of the tuberous electroreceptors of weakly electric fish. We recorded from two classes of tuberous electroreceptors known for pulse gymnotiforms: Burst Duration Coders (BDCs), and Pulse Markers (PMs). Both code for stimulus amplitude, although the dynamic range for BDCs is greater, and both exhibit strong directional preferences. Polar plots of spike number (for BDCs) or spike threshold (for PMs) versus electric field azimuth, are figure-8 shaped with two asymmetrical, elliptical lobes separated by 180°. The best azimuth of these two types of receptors from a given body region correlate with each other and with measures of best azimuth for transepidermal current flow. The shape and asymmetry of the directionality profiles appear to be caused by filter dynamics of the receptors. Pulse Markers are located on the anterior part of the body surface while Burst Duration Coders are located all over. The best directions of receptors in the anterior third of the body vary systematically with location from 0° to 180°. This region is probably critical for determining the direction of local electric fields. Together these receptors provide the CNS with sufficient information to construct a map of horizontal plane electric field directions.

    Notizen: Abstract Thirteen species of fleshy fungi were identified from two agricultural fields during the 1989 and 1990 growing seasons at the Rural Development Center (RDC) and Horticulture Farm, Coastal Plain Experiment Station, Tifton, Georgia. The three most common fungi wereCoprinus plicatilis, Cyathus olla, andPanaeolina foenisecii. At the former site, residue management practices were established for evaluating crop production and at the Horticulture Farm different crops were grown with conservation tillage employing a Low Input Sustainable Vegetable Production system. Tillage treatments at the RDC were no-till, row-till, ridge-plant, and moldboard-plowing. Residue management of triticale included burning or cutting the stubble at a height of 20 or 60 cm at harvest. At the Horticulture Farm, seven winter cover crops were compared to a fallow control. No significant differences were shown between observations of fungal species and individual treatments at the Horticulture Farm during 1989 and 1990. However, there were treatment differences for observed fungi at the RDC farm on specific dates for both years. Furthermore, no individual treatment(s) during the investigation increased observation rates of specific fungal species. Non-burned debris treatments showed consistently higher populations and diversity of fungi than burn debris plots.

    Notizen: Abstract The saline lakes of the Vestfold Hills, Antarctica, are unique ecosystems in which microorganisms survive and grow in cold, saline and often anaerobic conditions. A polyphasic approach to taxonomy of the microbiota has extended the known range of microbial diversity through description of nine new species including bacteria and archaea. Low temperature and reduced water activity are the major constraints on microbial growth in the lakes. Bělhrádek-type models provide a good description of temperature dependence and indicate the potential of the microbiota to colonise their natural habitats. Physiological traits of the microbiota selected by the physical and chemical characteristics of the saline lakes suggest biotechnological potential in areas such as polyunsaturated fatty acid production and hydrocarbon degradation.

    Notizen: Abstract A fully automatic, ultrasonic instrument to measure texture and formability parameters on metal sheet is described. Arrays of EMAT transducers are used to transmit and receiveS o Lamb waves propagating at 0°, 45°, and 90° with respect to the rolling direction. By analyzing the frequency dependence of the phase of the received signals, the long wavelength limit of the velocities is obtained. Included is a discussion of this algorithm, and subsequent processing steps to predict the ODC'sW 400,W 420, andW 440. On steel, the prediction of drawability parametersr and Δr based on a correlation developed previously by Mould and Johnson is also discussed. Results of blind field trials at facilities of three suppliers/users of steel sheet for automotive applications and one supplier of aluminum sheet for beverage can production are reported. The former confirmed the Mould-Johnson correlation for lowr material but indicated that refinements are needed for modern steels with highr. The aluminum data suggest a correlation between W440 and the degree of four-fold earing.
