    Notes: Many felids are threatened by loss of habitat, lack of genetic diversity, and over-exploitation. The reintroduction of bobcats (Felis rufus) to Cumberland Island, Georgia provided an opportunity to reintroduce a mid-sized felid without the concern for species survival that is paramount with endangered species. We captured bobcats from the coastal plain region of Georgia, briefly held them in captivity, and released them on Cumberland Island. We describe and evaluate the protocols and techniques used to accomplish the reintroduction. Future reintroductions of felids should consider the problem of post-release dispersal, although our island was relatively isolated and inhibited dispersal. Also, any reintroduction effort should invest effort and resources into post-release monitoring of the population. Empirical knowledge about the effects of spatial distribution, genetics, population dynamics, especially mechanisms of population regulation, behavior, and environmental conditions on the viability of populations is critical to the conservation of endangered species. Future research of the bobcats on Cumberland Island will be able to address aspects of the population and genetic dynamics of a small, insular felid population.

A segment of ribosomal DNA, about 1,350 base pairs long, was amplified from the microsporidian species Encephalitozoon hellem, isolated from AIDS patients, and Encephalitozoon cuniculi. the amplified DNA segment extends from position 530 in the small ribosomal RNA subunit to position 580 in the large ribosomal RNA subunit. A comparison of sequence data from this region for Encephalitozoon hellem and Encephalitozoon cuniculi shows relatively high sequence similarity, supporting the placement of these two organisms in the same genus. At the same time, sequence differences between the two organisms confirm that they are not the same species. Three separate isolates of E. hellem were sequenced in the highly variable intervening spacer region. the sequence was identical for all three isolates. Within the amplified DNA segment, regions were sequenced which yield highly variable, moderately variable and highly conserved sequence information, appropriate for comparison with other species in the phylum Microspora at all taxonomic levels. We suggest that sequence data from these regions be included in future species descriptions for the purposes of species identification and phylogenetic analysis. Restriction digests of the amplified region are presented and give a rapid method for distinguishing between the two Encephalitozoon species.

    Notes: : Experimental work in animals and, to a more limited extent, in humans, has demonstrated that the cholinergic system is involved in mechanisms which control learning and memory. Since there is cholinergic loss in a variety of dementing illnesses, any treatment designed to alleviate the mental symptoms of these diseases must address the issue of cholinergic dysfunction even if other treatments are also required to overcome other neurotransmitter imbalances. Work in rodents has demonstrated that cholinergic-rich fetal neural tissue transplants can, under certain circumstances, alleviate the behavioral effects of cholinergic lesions or of cholinergic decline associated with aging. More complex cognitive testing can be achieved using primates and, in this case, the common marmoset is a suitable species to use because its rapid and reliable reproductive rate aids the provision of appropriate transplant tissue. Marmosets with transection of the fornix are deprived of a cholinergic input into the dentate gyrus, posterior hippocampus and entorhinal cortex and are specifically impaired on learning tasks which require remembering a rule of responding (non-evaluative memory). Transplantation of cholinergic-rich fetal septal tissue into the hippocampus of such animals completely restores their ability to learn this type of task, whereas transplantation of cholinergic-poor fetal hippocampal tissue into the same area produces no such improvements. These results demonstrate that where a learning impairment is produced by a relatively simple procedure which has a major effect on one neurotransmitter, that function can be restored by transplantation of tissue containing that neurotransmitter even where the impairment consists of a very “high level” cognitive dysfunction.

    Notes: This paper presents a subset of the family of diagonally implicit Runge-Kutta (DIRK) time-stepping methods for finite-difference models of parabolic (diffusion-like) equations. It includes the first-order-accurate Euler implicit (backward in time, DIRK1) and the second-order Crank-Nicolson (DIRK2) methods as special cases. It combines a series of DIRK1 partial time steps so as to eliminate additional power terms of the time step δt, in the local error, by the number of partial steps used. This offers a large increase in computational efficiency, going from a DIRK1 to a DIRK2 method, and improves on the Crank-Nicolson method with a better choice of Runge-Kutta parameters. For a linear diffusion example, an optimal-parameter DIRK2 method offers the same accuracy as the Euler implicit method at two orders of magnitude larger time step, with an order of magnitude better accuracy than the Crank-Nicholson method. In a highly nonlinear horizontal unsaturated water flow example, using eight simulated medium to coarse soils, a DIRK2 method produces either an average maximum accuracy improvement of 4.7 times over the Euler implicit method, without Newton or Picard iteration, or from 3.8 to 48 times faster computer run times for the same accuracy.

    Notes: Tissue-specific effects of low growth temperature on maize chloroplast thylakoid protein accumulation were analysed using immunocytology. Sections of leaves from plants grown at 25 and 14°C were probed with antibodies to specific chloroplast thylakoid proteins from the four major protein multisubunit complexes of the thylakoid membrane followed by fluorescein-conjugated goat anti-rabbit antibodies. At a normal growth temperature of 25°C, the 32 kDa D1 protein of the photosystem II reaction centre and the 33 kDa protein of the extrinsic oxygen-evolving complex of photosystem II are both accumulated to a greater degree in the mesophyll than in the bundle sheath chloroplasts. In contrast, subunit II of photosystem I, cytochrome f and the α- and β-subunits of ATP synthetase are predominant in the bundle sheath thylakoids at 25°C. A striking difference between the 25°C-grown and the 14°C-grown leaf tissue was the presence in the latter of (20–30%) cells whose chloroplasts apparently completely lack several of the thylakoid proteins. In plants grown at 14°C, the accumulation of the 33 kDa protein of the extrinsic oxygen-evolving complex of photosystem II was apparently unchanged, but other thylakoid proteins showed a significant reduction. The uneven distribution of proteins between the bundle sheath and mesophyll chloroplasts observed at 25°C was also maintained at 14°C. Reduction in the fluorescence at 14°C was manifested either as an overall reduction in the diffuse fluorescence across the chloroplast profiles or less frequently as a reduction to small discrete bodies of intense fluorescence. The significance of these results to low-temperature-induced reduction in the photosynthetic productivity of maize is discussed.

    Notes: Leaves of Triticum aestivum cv. Avalon were grown in an atmosphere that contained 150 nmole mol-1 ozone for 7h each day. After leaves had reached maximum size, the leaf blade was divided into three sections to provide tissue of different age, the youngest at the base of the blade and the oldest at the leaf tip. The ozone treatment was found to decrease significantly the light-saturated rate and quantum yield of CO2 assimilation and the maximum quantum yield of photosystem II photochemistry in the oldest leaf section. No effects were found on the basal and middle sections of the leaf. These ozone-induced decreases in the photosynthetic parameters were associated with decreases in the efficiency of utilization of light for CO2 assimilation at the photon flux density under which the leaves were grown. The depression in photosynthetic performance of tissue near the leaf tip was accompanied by large decreases in the contents of total, soluble and thylakoid proteins and chlorophyll. There was also found to be a preferential loss of ribulose-1,5-carboxylase-oxygenase. These ozone-induced changes in chlorophyll and protein contents and the photosynthetic activities of the leaf tissue were similar to changes normally associated with leaf senescence. Two-dimensional polyacrylamide gel analyses of leaf proteins demonstrated the loss of some minor, and unidentified, proteins, whilst another group of minor proteins appeared. It is concluded that daily exposure of the leaf to 150 nmol mol-1 ozone for 7h had no effect on the development of the photosynthetic apparatus and its activities during leaf expansion, but it did promote the onset of premature senescence in fully expanded tissue that resulted in a loss of pigments, proteins and photosynthetic capacity and efficiency.
