Beschreibung: Plasmodium vivax and P. falciparum are the major causes of human malaria, except in sub-Saharan Africa where people lack the Duffy blood group antigen, the erythrocyte receptor for P. vivax. Duffy negative human erythrocytes are resistant to invasion by P. vivax and the related monkey malaria, P. knowlesi. Several lines of evidence in the present study indicate that the Duffy blood group antigen is the erythrocyte receptor for the chemokines interleukin-8 (IL-8) and melanoma growth stimulatory activity (MGSA). First, IL-8 binds minimally to Duffy negative erythrocytes. Second, a monoclonal antibody to the Duffy blood group antigen blocked binding of IL-8 and other chemokines to Duffy positive erythrocytes. Third, both MGSA and IL-8 blocked the binding of the parasite ligand and the invasion of human erythrocytes by P. knowlesi, suggesting the possibility of receptor blockade for anti-malarial therapy.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Horuk, R -- Chitnis, C E -- Darbonne, W C -- Colby, T J -- Rybicki, A -- Hadley, T J -- Miller, L H -- HL 41382/HL/NHLBI NIH HHS/ -- New York, N.Y. -- Science. 1993 Aug 27;261(5125):1182-4.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Genentech, Inc., South San Francisco, CA 94080.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/7689250" target="_blank"〉PubMed〈/a〉

Notizen: Resistance to the prevailing races of sunflower rust, Puccinia hehanthi Schw., is lacking in the commercial hybrids (Helianthus annuus L.). The objective of this study was to identify new sources of resistance to the four North American rust races in wild Helianthus species, and to determine their mode of inheritance. Seventy-eight accessions of H. annuus L., H. argophyllus Torrey and Gray, and H. petiolans Nutt. were evaluated in the greenhouse. Resistance to races 1, 2, 3, and 4 was observed in 25, 28, 15, and 26% of the plants, respectively, and 10% of the plants were resistant to all four races. Seven accessions that had a high percentage of resistant plants to all the four races were selected and one resistant plant from each accession was crossed with susceptible inbred line HA89. Three to four F1 plants resistant to all four races from each cross were backcrossed with HA89. F1 plants from PI-413118 × HA89 and PI 413175 × HA89 were resistant to all four races. The PI 413023 × HA89 F1 plants were 100 % resistant to races 3 and 4 and segregated in a 3: 1 resistant (R) to susceptible (S) ratio to races 1 and 2. The other four F1 combinations segregated 3R: IS ratios to all four races. Bc1F1 progenies revealed that plants from PI 413048, PI 413037, PI 413038, and PI 413171 used in the crosses possessed two dominant genes in heterozygous condition for resistance to each of the four races, whereas plants from PI 413023 possessed two dominant genes in heterozygous condition for resistance to each of races 1 and 2, and one dominant resistance gene in homozygous condition for each of races 3 and 4. Plants from PI 413118 and PI 413175 carried a single dominant gene in homozygous condition for resistance against each of the four races.

Notizen: Xylem sap was collected from wheat and barley growing in a drying soil, and the effect of the sap on transpiration was detected by a bioassay with detached wheat leaves. The inhibitory activity of fresh sap was small, and could be largely accounted for by the abscisic acid content (about 2×10-5mol m-3). When fresh sap was stored at -20°C for several days, the activity increased. Maximum activity developed after a week. This increase in activity was due to a compound that increased in size with storage at -20°C. When fresh sap was fractionated with filters of different molecular size exclusion characteristics, and the separated fractions stored at -20°C for a week, activity developed only in the fraction containing compounds smaller than 0·3 kDa. However, when sap already stored at -20°C was fractionated, activity was only in fractions containing compounds larger than 0·3 kDa. The increase in activity and in size did not occur with storage in liquid nitrogen (-196°C) or at -80°C. These results suggest that storage at -20°C causes the aggregation or polymerization of a small compound with low activity to form a large compound with high activity. This change is not catalysed by an enzyme because it can occur in a fraction from which molecules larger than 0·3 kDa are removed. It is probably promoted by high solute concentrations when ice crystals form. Sap collected from plants in soils of high water potential had little or no activity after storage at -20°C.

Notizen: Leaflet abscission in Sambucus nigra is the result of cell wall breakdown at the site of separation. Associated with wall degradation is an increase in the activity of the enzyme β1,4 glucanase (E.C.3.1.2.4) in the cells that comprise the abscission zone. The enzyme has been extracted from abscission zone tissue and purified using a substrate affinity column. A qualitative enzyme assay procedure has been developed and this has facilitated the purification process. The β1,4 glucanase enzyme has a pH optimum of 7 and a molecular mass of 54kDa. Antibodies have been raised to the purified protein. The role of the enzyme in the abscission process is discussed.

Notizen: The melting point Tm of iron at conditions of the Earth's inner core boundary (ICB) has been calculated from the dislocation theory of melting in metals. These calculations take into account the effects of pressure at ICB conditions and of possible freezing point depression resulting from dilution of pure iron in the outer core. Monte Carlo calculations have been used to estimate the uncertainties introduced by uncertainty in the geophysical parameters that are used in the calculations. With this approach Tm of pure e-Fe at a pressure of 330 GPa and without freezing point depression is 6160 ± 250 K, and 6110 K for a 1000 K freezing-point depression, Tm of pure y-Fe is not significantly different; these values agree well with shock-wave determinations of Tm. The estimated temperature of the ICB is lower by the freezing point depression, perhaps by 500-1000 K.
